An X-ray Spectromicroscopy Study of Albumin Adsorption to Crosslinked Polyethylene Oxide Films,

Abstract Synchrotron-based X-ray photoemission electron microscopy (X-PEEM) is used to characterize the near surface composition of polyethylene oxide (PEO) combined with 1.5, 5, and 10,wt.-% pentaerythritol triacrylate (PETA) crosslinker. It is found that as the concentration of PETA increases, it becomes the dominant component in the top 10,nm of the film surface. The same surfaces are also exposed to human serum albumin (HSA) and the distributions of the protein relative to PEO and PETA measured with X-PEEM. A positive correlation is found between levels of PETA and HSA at the surface. Above PETA concentrations of 5,wt.-%, HSA adsorption is significant, which suggests high levels of PETA (often used to immobilize PEO by crosslinking) can significantly reduce the non-fouling properties of PEO. Nitrification in terrestrial hot springs of Iceland and Kamchatka

Abstract Archaea have been detected recently as a major and often dominant component of the microbial communities performing ammonia oxidation in terrestrial and marine environments. In a molecular survey of archaeal ammonia monooxygenase (AMO) genes in terrestrial hot springs of Iceland and Kamchatka, the amoA gene encoding the ,-subunit of AMO was detected in a total of 14 hot springs out of the 22 investigated. Most of these amoA -positive hot springs had temperatures between 82 and 97 °C and pH range between 2.5 and 7. In phylogenetic analyses, these amoA genes formed three independent lineages within the known sequence clusters of marine or soil origin. Furthermore, in situ gross nitrification rates in Icelandic hot springs were estimated by the pool dilution technique directly on site. At temperatures above 80 °C, between 56 and 159 ,mol NO3, L,1 mud per day was produced. Furthermore, addition of ammonium to the hot spring samples before incubation yielded a more than twofold higher potential nitrification rate, indicating that the process was limited by ammonia supply. Our data provide evidence for an active role of archaea in nitrification of hot springs in a wide range of pH values and at a high temperature. [source]


Comparative study of the essential oils of seven Melaleuca (Myrtaceae) species grown in Brazil

Abstract The compositions of the essential oils obtained from seven species of Melaleuca grown in the municipal district of Viçosa, State of Minas Gerais, Brazil, were analysed by both GC and GC,MS. The various Melaleuca species showed differences within their relative essential oil compositions. Each of the oils analysed revealed a dominant component: terpinen-4-ol (53.7 ± 0.2%) for M. alternifolia; 1,8-cineole for M. armillaris (80.2 ± 0.0%), M. ericifolia (79.5 ± 0.4%), M. cajuputi, subspecies cajuputi (43.7 ± 0.5%) and M. cajuputi subspecies platyphylla (41.0 ± 8.5%); viridiflorol (71.0 ± 0.9%) for M. quinquenervia; and methyl eugenol (96.6 ± 0.7%) for M. leucadendra. Chemical constituents of the essential oils of Goniothalamus malayanus Hook. f. and Thoms.

Abstract The chemical composition of the leaf, bark and root oils of Goniothalamus malayanus Hook. f. and Thoms. was examined by capillary GC and GC,MS. The leaf oil was made up mainly of sesquiterpenoids, of which ,-selinene (33.6%) was the dominant component. The other major compounds present were viridiflorol (13.1%), epi -globulol (7.7%), (E)-nerolidol (4.4%) and globulol (3.8%). The bark and root oils, which were qualitatively similar but with some variation in levels of the individual constituents, were also rich in sesquiterpenoids (97% and 95%, respectively), with eudesmols as the most abundant compounds. The bark oil contained a higher amount of ,-eudesmol (32.2%), ,-eudesmol (21.8%) and ,-eudesmol (6.6%) than the root oil. Copyright © 2002 John Wiley & Sons, Ltd. [source]


Exoenzyme activities as indicators of dissolved organic matter composition in the hyporheic zone of a floodplain river

Summary 1. We measured the hyporheic microbial exoenzyme activities in a floodplain river to determine whether dissolved organic matter (DOM) bioavailability varied with overlying riparian vegetation patch structure or position along flowpaths. 2. Particulate organic matter (POM), dissolved organic carbon (DOC), dissolved oxygen (DO), electrical conductivity and temperature were sampled from wells in a riparian terrace on the Queets River, Washington, U.S.A. on 25 March, 15 May, 20 July and 09 October 1999. Dissolved nitrate, ammonium and soluble reactive phosphorus were also collected on 20 July and 09 October 1999. Wells were characterised by their associated overlying vegetation: bare cobble/young alder, mid-aged alder (8,20 years) and old alder/old-growth conifer (25 to >100 years). POM was analysed for the ash-free dry mass and the activities of eight exoenzymes (,-glucosidase, ,-glucosidase, , -N-acetylglucosaminidase, xylosidase, phosphatase, leucine aminopeptidase, esterase and endopeptidase) using fluorogenic substrates. 3. Exoenzyme activities in the Queets River hyporheic zone indicated the presence of an active microbial community metabolising a diverse array of organic molecules. Individual exoenzyme activity (mean ± standard error) ranged from 0.507 ± 0.1547 to 22.8 ± 5.69 ,mol MUF (g AFDM),1 h,1, was highly variable among wells and varied seasonally, with the lowest rates occurring in March. Exoenzyme activities were weakly correlated with DO, DOC and inorganic nutrient concentrations. 4. Ratios of leucine aminopeptidase : ,-glucosidase were low in March, May and October and high in July, potentially indicating a switch from polysaccharides to proteins as the dominant component of microbial metabolism. 5. Principal components analysis indicated that there were patch effects and that these effects were strongest in the summer. 6. DOM degradation patterns did not change systematically along hyporheic flowpaths but varied with overlying forest patch type in the Queets River hyporheic zone, suggesting that additional carbon inputs exist. We hypothesise that the most likely input is the downward movement of DOM from overlying riparian soils. Understanding this movement of DOM from soils to subsurface water is essential for understanding both the hyporheic metabolism and the carbon budget of streams and rivers. [source]


Robustness of inference on measured covariates to misspecification of genetic random effects in family studies

Abstract Family studies to identify disease-related genes frequently collect only families with multiple cases. It is often desirable to determine if risk factors that are known to influence disease risk in the general population also play a role in the study families. If so, these factors should be incorporated into the genetic analysis to control for confounding. Pfeiffer et al. [2001 Biometrika 88: 933,948] proposed a variance components or random effects model to account for common familial effects and for different genetic correlations among family members. After adjusting for ascertainment, they found maximum likelihood estimates of the measured exposure effects. Although it is appealing that this model accounts for genetic correlations as well as for the ascertainment of families, in order to perform an analysis one needs to specify the distribution of random genetic effects. The current work investigates the robustness of the proposed model with respect to various misspecifications of genetic random effects in simulations. When the true underlying genetic mechanism is polygenic with a small dominant component, or Mendelian with low allele frequency and penetrance, the effects of misspecification on the estimation of fixed effects in the model are negligible. Competitive dynamics in two- and three-component intercrops

Summary 1Intercropping is receiving increasing attention because it offers potential advantages for resource utilization, decreased inputs and increased sustainability in crop production, but our understanding of the interactions among intercropped species is still very limited. 2We grew pea Pisum sativum, barley Hordeum vulgare and rape Brassica napus as sole crops and intercrops under field conditions using a replacement design. We collected total dry matter data from sequential harvests and fitted the data to a logistic growth model. At each harvest we estimated the relative Competitive Strength (CS) of the three crops by fitting the data to a simple interspecific competition model. 3The pea monocrop produced the largest amount of biomass from the middle to the end of the growth period, but pea was not dominant in intercrops. 4Fitting data to a logistic growth model emphasizes the importance of initial size differences for interactions among intercrops. Barley was the dominant component of the intercrops largely because of its initial size advantage. The competitive effect of barley on its companion crops, measured as CS, increased throughout most of the growing season. 5The performance of each crop species was very different when it grew with a second species rather than in monoculture, but addition of a third crop species had only minor effects on behaviour of the individual crops. 6Synthesis and applications. Including sequential harvests in experiments on intercropping can provide important information about how competitive hierarchies are established and change over time. Our results suggest that increased understanding of the role of asymmetric competition among species and the resulting advantages of early germination and seedling emergence would be valuable in designing intercrops. More focus on understanding the mechanisms that govern interactions between intercropped species is needed for designing optimized intercropping systems. [source]


Richness and diversity of helminth communities in tropical freshwater fishes: empirical evidence

Abstract Aim Published information on the richness and diversity of helminth parasite communities in tropical freshwater fishes is reviewed in response to expectations of species-rich parasite communities in tropical regions. Location Areas covered include the tropics and some subtropical areas. In addition, the north temperate area of the nearctic zone is included for comparison. Methods Data from 159 communities in 118 species of tropical freshwater fish, summarized from 46 published studies, were used for this review. Parasite community descriptors used in the analyses included component community richness and calculated mean species richness. Data from 130 communities in 47 species of nearctic north temperate freshwater fish were summarized from 31 studies and used for comparison. Results The component helminth communities of many tropical freshwater fish are species-poor, and considerable proportions of fish from certain parts of the tropics, e.g. West African drainages, are uninfected or lightly infected. Mean helminth species richness was low and equaled or exceeded 2.0 in only 22 of 114 communities. No single group of helminths was identified as a dominant component of the fauna and species composition was variable among and within broader geographical areas. The richest enteric helminth assemblages were found in mochokid and clariid catfish with a mixed carnivorous diet, whereas algal feeders, herbivores and detritivores generally had species-poor gut helminth communities. Comparisons indicated that certain areas in the north temperate region had higher helminth species richness in fishes than areas in the tropics. Main conclusions Expectations of high species richness in helminth communities of tropical freshwater fishes are not fulfilled by the data. Direct comparisons of infracommunities and component communities in host species across widely separated phylogenetic and geographical lines are inappropriate. Examination of latitudinal differences in richness of monophyletic parasite groups or of compound communities may uncover patterns different from those found in this study. Richness of helminth communities may be ultimately determined not by the number of host species present but by the degree of divergence of host lineages and by their diversification modes. A phylogenetic framework for hosts and parasites will reveal if increased host species richness within host clades, when host speciation is accompanied by habitat or diet specialization, or both, leads to lower helminth diversity in host species by fragmentation of a core helminth fauna characteristic or specific of the larger host clade. This pattern may be analysed in the context of cospeciation and acquisition from other unrelated hosts (host-sharing or host-switching). [source]


Influence of boron concentration on the XPS spectra of the (100) surface of homoepitaxial boron-doped diamond films

Abstract As-grown (100) homoepitaxial diamond films with boron concentrations [B] from 4.6 × 1016 to 1.5 × 1021 cm,3 have been analysed using X-ray photoelectron spectroscopy (XPS). Their C 1s core levels contain a dominant component around 284.17 ± 0.2 eV ascribed to sp3 C and a main secondary component around 284.88 ± 0.2 eV ascribed to CHx (x , 2) on surface defects. Their relative concentration decreases and increases, respectively, as [B] increases. A significant component around 286.4 ± 0.2 eV ascribed to ether group (C,O,C) remains nearly constant up to [B] , 3 × 1020 cm,3, then increases for greater boron concentrations. Other components around 283.0 ± 0.2, 287.69 ± 0.2 and 288.76 ± 0.2 eV ascribed, respectively, to sp2 C, carbonyl (C=O) and carboxyl (HO,C=O) on surface defects remain with low concentrations. The occurrence of these XPS components, their assignments and their relative concentrations are satisfactorily compared to those previously found for IIb crystals with (100) surface and for polycrystalline films with [B] around 1019 and 7 × 1020 cm,3. Environmental modulation of reproductive activity of the invasive mussel Limnoperna fortunei: implications for antifouling strategies

Abstract Limnoperna fortunei (Dunker, 1857) (Bivalvia) invaded Argentina through the Río de la Plata estuary around 1990 and is presently established throughout five South American countries as a dominant component of the benthic fauna and a major nuisance for industry and power plants. Between 1997 and 2006 we monitored the reproductive activity of L. fortunei through weekly measurements of planktonic larvae in six South American water bodies: Río de la Plata estuary, Paraná and Carapachay rivers, Salto Grande, Itaipú and Embalse de Río Tercero reservoirs. Mean larval densities varied between 4000 and 7000 individuals m,3; except in the reservoirs of Itaipú (450 ind. m,3) and Salto Grande (869 ind. m,3), where the mussel was first recorded shortly before our surveys, and upstream dispersal is limited (Salto Grande). In all cases, reproductive output decreases during the winter. At four of the six sites surveyed larval densities were comparatively high for 8.8,10.2 months per year. A lower food supply is possibly responsible for the shorter reproductive period of 5.9 months at Embalse. At Salto Grande, there is a well-defined mid-summer drop in larval numbers, coinciding with blooms of cyanobacteria. We propose that, in addition to temperature, two major factors may regulate the reproductive activity of L. fortunei: (i) the availability of food; and (ii) blooms of toxic cyanobacteria, significantly shortening the otherwise very long reproductive period. This information is important for the design of antifouling programmes involving the use of molluscicides, and has potential for reduced biocide use. These results provide supporting evidence for some fundamental ecological theories of invasions discussed here. [source]


The Chemistry of Escapin: Identification and Quantification of the Components in the Complex Mixture Generated by an L -Amino Acid Oxidase in the Defensive Secretion of the Sea Snail Aplysia californica

Abstract A complex mixture of products in an enzymatic reaction: Aplysia californica releases amino acid oxidase and its substrate lysine in defensive secretions to produce a mixture of multiple compounds (see figure). Escapin is an L -amino acid oxidase in the ink of a marine snail, the sea hare Aplysia californica, which oxidizes L -lysine (1) to produce a mixture of chemicals which is antipredatory and antimicrobial. The goal of our study was to determine the identity and relative abundance of the constituents of this mixture, using molecules generated enzymatically with escapin and also using products of organic syntheses. We examined this mixture under the natural range of pH values for ink,from ,5 at full strength to ,8 when fully diluted in sea water. The enzymatic reaction likely forms an equilibrium mixture containing the linear form ,-keto-,-aminocaproic acid (2), the cyclic imine ,1 -piperidine-2-carboxylic acid (3), the cyclic enamine ,2 -piperidine-2-carboxylic acid (4), possibly the linear enol 6-amino-2-hydroxy-hex-2-enoic acid (7), the ,-dihydroxy acid 6-amino-2,2-dihydroxy-hexanoic acid (8), and the cyclic aminol 2-hydroxy-piperidine-2-carboxylic acid (9). Using NMR and mass spectroscopy, we show that 3 is the major component of this enzymatic product at any pH, but at more basic conditions, the equilibrium shifts to produce relatively more 4, and at acidic conditions, the equilibrium shifts to produce relatively more 2, 7, and/or 9. Studies of escapin's enzyme kinetics demonstrate that because of the high concentrations of escapin and L -lysine in the ink secretion, millimolar concentrations of 3, H2O2, and ammonia are produced, and also lower concentrations of 2, 4, 7, and 9 as a result. We also show that reactions of this mixture with H2O2 produce ,-aminovaleric acid (5) and ,-valerolactam (6), with 6 being the dominant component under the naturally acidic conditions of ink. Thus, the product of escapin's action on L -lysine contains an equilibrium mixture that is more complex than previously known for any L -amino acid oxidase. [source]


THE EFFECT OF EPISTASIS ON THE EXCESS OF THE ADDITIVE AND NONADDITIVE VARIANCES AFTER POPULATION BOTTLENECKS

Abstract The effect of population bottlenecks on the components of the genetic variance generated by two neutral independent epistatic loci has been studied theoretically (VA, additive; VD, dominant; VAA, additive × additive; VAD, additive × dominant; VDD; dominant × dominant components of variance). Nonoverdominance and overdominance models were considered, covering all possible types of marginal gene action at the single locus level. The variance components in an infinitely large panmictic population (ancestral components) were compared with their expected values at equilibrium, after t consecutive bottlenecks of equal size N (derived components). Formulae were obtained in terms of allele frequencies and effects at each locus and the corresponding epistatic value. An excess of VA after bottlenecks can be assigned to two sources: (1) the spatiotemporal changes in the marginal average effects of gene substitution ai, which are equal to zero only for additive gene action within and between loci; and (2) the covariance between a2i and the heterozygosity at the loci involved, which is generated by dominance, with or without epistasis. Numerical examples were analyzed, indicating that an increase in VA after bottlenecks will only occur if its ancestral value is minimal or very small. For the nonoverdominance model with weak reinforcing epistasis, that increase has been detected only for extreme frequencies of the negative allele at one or both loci. With strong epistasis, however, this result can be extended to a broad range of intermediate frequencies. With no epistasis, the same qualitative results were found, indicating that dominance can be considered as the primary cause of an increase in VA following bottlenecks. In parallel, the derived total nonadditive variance exceeded its ancestral value (VNA= VD+ VAA+VAD+ VDD) for a range of combinations of allele frequencies covering those for an excess of VA and for very large frequencies of the negative allele at both loci. For the overdominance model, an increase in VA and VNA was respectively observed for equilibrium (intermediate) frequencies at one or both loci or for extreme frequencies at both loci. For all models, the magnitude of the change of VA and VNA was inversely related to N and t. At low levels of inbreeding, the between-line variance was not affected by the type of gene action. For the models considered, the results indicate that it is unlikely that the rate of evolution may be accelerated after population bottlenecks, in spite of occasional increments of the derived VA over its ancestral value. Supercritical carbon dioxide extraction of volatiles from Satureja fruticosa Béguinot

Abstract Dried flowers and leaves of Satureja fruticosa Béguinot were submitted to supercritical CO2 extraction (SFE), followed by a two-stage fractional separation. The extraction was carried out at 40 °C in a flow apparatus, provided with a 1 l extraction vessel and two separators of 0.27 l each. Conditions of extraction, such as pressure, particle size of plant material and CO2 flow rate, were studied in order to assess their influence on the yield and composition of the volatiles. The best conditions of extraction were 90 bar pressure at 1.32 kg/h CO2 flow rate and a plant material particle size of 0.5 mm. The SFE volatiles obtained and the essential oil isolated by hydrodistillation (HD) were analysed and compared by GC and GC,MS. The oxygen-containing monoterpenes, pulegone (33,36% for SFE and 40,41% for HD), isomenthone (14,18% for SFE and 21% for HD), piperitenone (11,12% for SFE and 9,11% for HD) and piperitenone oxide (11,13% for SFE and 7,9% for HD) were the dominant components in all samples. Fatty acid analyses reveal high degrees of omnivory and dietary plasticity in pond-dwelling tadpoles

Summary 1.,Understanding the trophic relationships of consumers is central to ecology, but constructing meaningful food webs is often difficult because of a lack of detailed information on consumption versus assimilation and high degrees of omnivory. 2.,We used fatty acid analyses to examine the trophic relationships of three common larval anurans (Pseudacris crucifer, Lithobates catesbeianus and Lithobates clamitans) that are often classified as grazers or detritivores. Tadpoles and potential food sources were sampled in four ponds in southern Illinois and analysed for fatty acid composition. Single linkage cluster analysis was then used to compare fatty acid profiles among tadpole gut contents, tadpole muscle tissues and available food resources. 3.,Diets varied among species and within species among ponds, but organic sediments consistently contributed most to the fatty acid composition of the gut contents of all species. Fatty acid profiles also indicated that larval insects and phytoplankton were consumed by both L. catesbeianus and L. clamitans in one pond, while L. clamitans and P. crucifer consumed mainly periphyton along with sediments in another pond, and these diet differences appeared linked to physical differences among ponds, with periphyton and/or phytoplankton contributing more to tadpole diets in less shaded ponds. 4.,The fatty acid composition of muscle tissues of L. clamitans, the dominant tadpole in these systems, indicated that plant detritus and bacteria, which were the dominant components of organic sediments in the ponds, were common components of the assimilatory diet. 5.,Results demonstrate the utility of fatty acid analyses for assessing both consumption and assimilation. The tadpole assemblages we examined derive much of their energy from heterotrophic and allochthonous sources and exhibit high dietary plasticity. This information will allow for more accurate and comprehensive assessments of trophic interactions in freshwater habitats, as well as aid in amphibian conservation, management and captive propagation efforts. The response of volume and returns to the information shocks in China's commodity futures markets

This study investigates the response of returns and volume to different information shocks in China's commodity futures markets using bivariate moving average representation (BMAR) and bivariate vector autoregression (BVAR) methodologies. Consistent with the conclusions from stock market studies that have used these methodologies, it is found that the informational/permanent components are the dominant components for returns movements, and the noninformational/transitory components are the dominant components for trading volume. It is also found that the market response of copper futures improved during the sample period, and the market responses of actively traded futures (copper and soybeans) are better than those of the less actively traded futures (aluminum and wheat). © 2005 Wiley Periodicals, Inc.
