Domestic dogs as an edge effect in the Brasília National Park, Brazil: interactions with native mammals

Abstract Edge effects are a well-known result of habitat fragmentation. However, little has been published on fragmentation, isolation and the intrusive influence from the surrounding matrix at the landscape level. The objectives of the present study are to evaluate the presence of dogs in the Brasília National Park (BNP) in relation to habitat type and the influence from the surrounding matrix. In addition, this study examines the response of the native mammal fauna to the presence of dogs. Track stations were built along dirt roads in the BNP and subsequently examined for the presence or absence of tracks. We used a stepwise logistic regression to model the occurrence of five mammal species relative to habitat variables, with an ,=0.05 to determine whether to enter and retain a variable in the model. A simulation of each species occurrence probability was conducted using a combination of selected habitat variables in a resource selection probability function. Results indicate a negative relationship between distance from the BNP edge and the probability of dog occurrences. From an ecological perspective, the presence of dogs inside the BNP indicates an edge effect. The occurrence of the maned wolf was positively associated with distance from a garbage dump site and negatively associated with the presence of dog tracks. The maned wolf and giant anteater seem to avoid areas near the garbage dump as well as areas with dog tracks. There is no support for the possible existence of a feral dog population inside the BNP, but the effects of free-ranging dogs on the wildlife population in such an isolated protected area must not be neglected. Domestic dog Canis familiaris populations and disease control programs should be established in the urban, sub-urban and rural areas surrounding the BNP, along with the complete removal of the garbage dump from the BNP surroundings. Analysis of Forensic SNPs in the Canine mtDNA HV1 Mutational Hotspot Region,

Abstract:, A 60 bp sequence variation hotspot in the canine mitochondrial DNA hypervariable region 1 was evaluated for its use in forensic investigations. Nineteen haplotypes containing 18 single nucleotide polymorphisms were observed among laboratory-generated and GenBank-derived domestic dog sequences representing five regional localities in the U.S. Samples from the different localities were highly variable with the levels of intra-population variability being similar among the populations studied. AMOVA further confirmed that there was no significant genetic structuring of the populations. Assays using these haplotypes were robust, canid specific and portend a rapid method for correctly excluding individual dogs as noncontributors of forensic evidence. Species-specificity of the primers was confirmed by means of in-tube polymerase chain reaction of human and cat DNA and in-silico assessment of the genomes of several animal species. Breed-specific fragments were not detected among the common haplotypes but there is evidence that this assay may be capable of differentiating domestic dog, wolf, and coyote sequences. [source]


Hair Whorls in the Dog (Canis familiaris).

Abstract Hair whorl characteristics were assessed in the domestic dog (Canis familiaris) in the regions of cephalic, cervical (dorsal, ventral, and lateral), thoracic and brachial axillary regions, the chest, shoulders, elbows, ventral abdominal region, and on the caudal thighs (ischiatic). They were classified as simple or tufted, and their position was recorded as the distance between their centers and bony landmarks within each region. The distribution of whorls was explored in a cohort of domestic dogs (N = 120) comprising a variety of breeds and cross-breeds, sourced from shelters (N = 60) and the general public (N = 60). Whorls observed in the majority of dogs in this cohort typically occurred on the chest, brachial axillary region, elbows, and ischiatic region. Atypical whorls were present in fewer than 20% of the population, and included those on the head (cephalic), cervical regions (dorsal, ventral, and lateral), shoulders, thoracic axillary region, and on the ventral abdominal region. The majority of whorls on dogs were classified as simple. In contrast, those located on the elbows and the majority of chest whorls were tufted. The presence and position of whorls were often associated with several variables including coat length and thickness, and the sex and source of the dog. The palpation and hair-cluster method of whorl assessment described in this article is best suited to dogs with short-to-medium coat lengths. The current methodology developed to assess hair whorl characteristics provides a framework for future investigations into any associations between hair whorl characteristics and other canine traits such as temperament. The Distribution of Ganglion Cells in the Equine Retina and its Relationship to Skull Morphology

Summary It has recently been reported that a strong correlation exists between the distribution of retinal ganglion cells and nose length in the domestic dog. To determine if this phenomenon occurs in another domestic species with diverse skull morphology, the current study examined the distribution of retinal ganglion cells in 30 horses from a variety of breeds. There was a significant variation in the density of ganglion cells found across the retinae. Breed was a significant predictor for ganglion cell density within the visual streak. A strong positive correlation exists between the density of ganglion cells in the visual streak and nasal length. Significant variation was also seen in the area centralis but did not correlate with any of the recorded skull measurements. The findings of this study provide us with further understanding of the equine visual system and the level of variation that exists between individuals of the same species. [source]


Coding repeats and evolutionary "agility"

The rapid generation of new shapes observed in the living world is the result of genetic variation, especially in "morphological" developmental genes. Many of these genes contain coding tandem repeats. Fondon and Garner have shown that expansions and contractions of these repeats are associated with the great diversity of morphologies observed in the domestic dog, Canis familiaris.1 In particular, they found that the repeat variations in two genes were significantly associated with changes in limb and skull morphology. A new autosomal STR nineplex for canine identification and parentage testing

Abstract A single multiplex PCR assay capable of simultaneously amplifying nine canine-specific autosomal STR markers (FH3210, FH3241, FH2004, FH2658, FH4012, REN214L11, FH2010, FH2361 and the newly described C38) was developed for individual identification and parentage testing in domestic dogs. In order to increase genotyping efficiency, amplicon sizes were optimized for a 90,350,bp range, with fluorescently labelled primers for use in Applied Biosystems, Inc., platforms. The performance of this new multiplex system was tested in 113 individuals from a case-study population and 12 random dogs from mixed-breed origin. Co-dominant inheritance of STR alleles was investigated in 101 father, mother and son trios. Expected heterozygosity values vary between 0.5648 for REN214L11 and 0.9050 for C38. The high level of genetic diversity observed for most markers provides this multiplex with a very high discriminating power (matching probability=1.63/1010 and matching probability among siblings=4.9/103). Allele sequences and a proposal for standardized nomenclature are also herein presented, aiming at implementing the use of this system in forensic DNA typing and population genetic studies. This approach resulted in an optimized and well-characterized canine DNA genotyping system that is highly performing and straightforward to integrate and employ routinely. Although this STR multiplex was developed for use and tested in a case-study population, the Portuguese breed Cão de Gado Transmontano, it proved to be useful for general identification purposes or parentage testing. Competition between domestic dogs and Ethiopian wolf (Canis simensis) in the Bale Mountains National Park, Ethiopia

Abstract The potential effects of the domestic dogs (Canis familiaris) on the Endangered Ethiopian wolf (Canis simensis) through exploitative and interference competition were studied in the Web Valley of Bale Mountains national park between November 2001 and February 2003. All dogs were owned in the study area and no feral dogs were reported or observed during the research period. The diet of domestic dogs was dominated by barley husks and human faeces which contributed 45% and 20.7% of the total 382 meals observed during focal watch observations. Analysis of dog faeces provided similar results with barley husks, human faeces and animal carcasses occurring in 86.8%, 21.4% and 19.4% of the 1200 faecal samples analysed. Both focal watch and faecal analyses revealed that rodents contributed only a very small proportion of the diet of dogs accounting for only 4.2% of the focal watch and 2.8% of the faecal analysis of roaming dogs. As Ethiopian wolves fed almost exclusively on rodent year round, no significant exploitative competition between dogs and wolves were assessed. Only small proportion of the domestic dogs roamed in the Ethiopian wolf range and interference competition did not appear to be a serious threat for the Ethiopian wolf. Résumé De novembre 2001 à février 2003, on a étudié les effets potentiels des chiens domestiques (Canis familiaris) sur le loup d'Ethiopie (Canis simensis) qui est en danger, en raison d'une compétition par exploitation alimentaire ou par interférences, dans la Vallée de Web, dans le Parc National des montagnes de Bale. Tous les chiens de l'étude avaient un propriétaire, et nous n'avons ni rapporté ni observé de chien féral pendant cette période. Le régime alimentaire des chiens comprenait principalement de la balle d'orge et des excréments humains qui composaient respectivement 45% et 20,7% du total des 382 repas observés pendant des observations focalisées. L'analyse des excréments de chiens donne des résultats semblables : la balle d'orge, les excréments humains et les carcasses d'animaux sont présents dans 86,8%, 21,4% et 19,4% des 1 200 échantillons fécaux analysés. Et les observations focalisées et les analyses des crottes ont révélé que les rongeurs ne composent qu'une toute petite proportion du régime des chiens, avec 4,2% des observations et 2,8% des analyses fécales des chiens errant en liberté. Comme les loups d'Ethiopie se nourrissent toute l'année presque exclusivement de rongeurs, nous avons estimé qu'il n'y avait aucune compétition par exploitation significative entre les chiens et les loups. Detecting introgressive hybridization between free-ranging domestic dogs and wild wolves (Canis lupus) by admixture linkage disequilibrium analysis

Abstract Occasional crossbreeding between free-ranging domestic dogs and wild wolves (Canis lupus) has been detected in some European countries by mitochondrial DNA sequencing and genotyping unlinked microsatellite loci. Maternal and unlinked genomic markers, however, might underestimate the extent of introgressive hybridization, and their impacts on the preservation of wild wolf gene pools. In this study, we genotyped 220 presumed Italian wolves, 85 dogs and 7 known hybrids at 16 microsatellites belonging to four different linkage groups (plus four unlinked microsatellites). Population clustering and individual assignments were performed using a Bayesian procedure implemented in structure 2.1, which models the gametic disequilibrium arising between linked loci during admixtures, aiming to trace hybridization events further back in time and infer the population of origin of chromosomal blocks. Results indicate that (i) linkage disequilibrium was higher in wolves than in dogs; (ii) 11 out of 220 wolves (5.0%) were likely admixed, a proportion that is significantly higher than one admixed genotype in 107 wolves found previously in a study using unlinked markers; (iii) posterior maximum-likelihood estimates of the recombination parameter r revealed that introgression in Italian wolves is not recent, but could have continued for the last 70 (± 20) generations, corresponding to approximately 140,210 years. Bayesian clustering showed that, despite some admixture, wolf and dog gene pools remain sharply distinct (the average proportions of membership to wolf and dog clusters were Qw = 0.95 and Qd = 0.98, respectively), suggesting that hybridization was not frequent, and that introgression in nature is counteracted by behavioural or selective constraints. SNPs in ecological and conservation studies: a test in the Scandinavian wolf population

Abstract Single nucleotide polymorphisms (SNPs) have the potential to become the genetic marker of choice in studies of the ecology and conservation of natural populations because of their capacity to access variability across the genome. In this study, we provide one of the first demonstrations of SNP discovery in a wild population in order to address typical issues of importance in ecology and conservation in the recolonized Scandinavian and neighbouring Finnish wolf Canis lupus populations. Using end sequence from BAC (bacterial artificial chromosome) clones specific for dogs, we designed assays for 24 SNP loci, 20 sites of which had previously been shown to be polymorphic in domestic dogs and four sites were newly identified as polymorphic in wolves. Of the 24 assayed loci, 22 SNPs were found to be variable within the Scandinavian population and, importantly, these were able to distinguish individual wolves from one another (unbiased probability of identity of 4.33 × 10,8), providing equivalent results to that derived from 12 variable microsatellites genotyped in the same population. An assignment test shows differentiation between the Scandinavian and neighbouring Finnish wolf populations, although not all known immigrants are accurately identified. An exploration of the misclassification rates in the identification of relationships shows that neither 22 SNP nor 20 microsatellite loci are able to discriminate across single order relationships. Despite the remaining obstacle of SNP discovery in nonmodel organisms, the use of SNPs in ecological and conservation studies is encouraged by the advent of large scale screening methods. Furthermore, the ability to amplify extremely small fragments makes SNPs of particular use for population monitoring, where faecal and other noninvasive samples are routinely used. [source]

Diet of free-ranging domestic dogs (Canis familiaris) in rural Zimbabwe: implications for wild scavengers on the periphery of wildlife reserves

Numbers of free-ranging dogs (Canis familiaris) have reached unprecedented levels in Zimbabwean communal lands (agropastoralist rural areas). This study examined the potential competitive interactions between dogs and wild scavengers on the boundary of Gokwe Communal Land (GCL) and the Sengwa Wildlife Research Area (SWRA) in 1995,96. Dietary studies showed that dogs were primarily scavengers of human waste and animal carcasses. Twelve experimental carcasses indicated that dogs were the most successful species in the vertebrate scavenger guild, consuming 60% of available biomass and finding 66.7% of carcasses. Dogs monopolized the supply of domestic animal carrion within GCL, but also consumed wild carrion up to 1 km within the SWRA, and were seen 3 km inside the reserve. Their principal competitors for carcasses were vultures, and to a lesser degree lions (Panthera leo), leopards (P. pardus) and spotted hyaenas (Crocuta crocuta). Dogs outcompete vultures on wildlife reserve boundaries owing to their high densities, nocturnal and diurnal activity, physical dominance and greater tolerance of human disturbance. With a population growth rate of 6.5% per annum the influence of dogs will intensify on the peripheries of reserves, exacerbating their existing threat to wild scavengers. This scenario is probably occurring in many other African countries. [source]


Clinical diagnosis and treatment of suspected neuropathic pain in three dogs

Three dogs were referred to The Queen's Veterinary School Hospital at University of Cambridge for chronic behavioural or locomotor disorders associated with pain. All three had been unsuccessfully treated with conventional analgesics, including non-steroidal anti-inflammatory drugs, glucocorticoids and opiate agonists, prior to referral, with minimal or no response. They were investigated by neurological examination plus conventional ancillary diagnostic tests and therapeutic drug trials. Ruling out other causes of pain and applying previously well-described criteria, each case was diagnosed as consistent with neuropathic pain, a poorly recognised condition in domestic dogs. Treatment with the tricyclic antidepressant drug, amitriptyline, or the antiepileptic drug, gabapentin, resulted in either a dramatic improvement or full resolution of clinical signs in all cases. Genetic analysis of canine parvovirus from dogs in Australia

Objective To determine the genetic variants of canine parvovirus-2 (CPV) present in domestic dogs in Australia and to investigate 26 cases of apparent vaccine failure. Design Thirty-three samples of faeces or intestinal tissues and 16 cell culture virus isolates collected over a period from 1980 to 2005 from five Australian states were analysed. Procedure DNA was extracted from the samples and a 1975 bp fragment of the VP1/2 gene of CPV was amplified by polymerase chain reaction (PCR) and sequenced. Sequences were compared to published strains of CPV-2, CPV-2a, CPV-2b and CPV-2c. Results Forty-one of 43 PCR-positive samples contained CPV-2a viruses. One sample collected in 2002 from a pup in northern NSW contained a CPV-2b virus. One sample that had been included in the study as a CPV-antigen negative control sample contained a CPV-2 virus. Conclusion CPV-2a remains the predominant genetic variant of CPV in dogs in Australia and has not been replaced by CPV-2b or CPV-2c as in many other countries. The vaccine failures investigated in the study were likely caused not by genetic variation of field viruses but by maternal antibody interference in the response of pups to vaccination. [source]


A retrospective report of 90 dogs with suspected cane toad (Bufo marinus) toxicity

Objective To report the clinical characteristics of toad toxicity in domestic dogs in Brisbane. Design A retrospective analysis of clinical cases. Procedure All cases of toad poisoning which presented to a northern suburbs emergency clinic in Brisbane over a 30-month period beginning in April 1999 were reviewed. Results A total of 90 canine cases of suspected toad poisoning were reviewed. Small breed dogs accounted for 76% of cases. Jack Russell, Silky, and Fox Terriers were the most represented breeds. Cases were reported year round, with fewest cases over the winter months. The most common clinical signs were increased salivation (78% of cases), and red oral mucous membranes (63% cases). Seizures occurred in 31% of cases. Generally the outcome was excellent with 96% survival. [source]

Objective To determine whether dogs and cats are potential reservoirs of Ross River (RR) and Barmah Forest (BF) viruses Method Young seronegative female dogs and cats were experimentally exposed to the viruses using Ochlerotatus vigilax(Skuse) mosquitoes. Results Only one of the 10 dogs and one of the 10 cats exposed to RR developed neutralising antibody. None of the animals developed detectable viraemia or clinical signs. One dog and three cats exposed to BF developed neutralising antibody. In addition, a serological survey of sera obtained from domestic dogs and cats residing in the Brisbane region indicated that 23.7% and 1.3% of dogs, and 14% and 2% of cats, had neutralising antibodies to RR and BF respectively. Conclusions Although dogs and cats are exposed naturally to these viruses, and can become infected, they are unlikely to be important urban reservoirs of either virus. [source]

Serological survey for Ehrlichia canis in urban dogs from the major population centres of northern Australia

Objective To detect evidence of Ehrlichia canis infection of dogs from the major population centres of northern Australia, if present. Design Serological investigation for E canis. Procedure The sera of 316 domestic dogs, collected from the northern Australian population centres of Townsville, Cairns, Darwin, Kununurra and Broome from May 1997 to August 1999, were investigated for evidence of infection with E canis. Samples were tested for antibodies to E canis using an indirect fluorescent antibody (IFA) test. The buffy coats from blood of dogs whose serum reacted in the IFA test were subsequently tested with a nested PCR to detect E canis DNA. When available, blood from these dogs was injected into suckling mice, which were then examined for clinical disease and tested for the presence of E canis antibodies. Results Of the 316 samples tested seven reacted in the IFA test for E canis. None of the dogs from which these samples were obtained exhibited clinical signs of acute or chronic ehrlichiosis. The six positive samples available for testing were negative when tested with the nested PCR. Suckling mice inoculated with blood from three of the dogs whose serum was positive by IFA test showed no signs of clinical disease nor did their give positive reactions in the IFA test. Conclusions No evidence of E canis infection was confirmed in any of the dogs examined. Northern Australia would appear to remain free of this obligate parasite. [source]
