Cross Standardized coefficients from the logistic regression analyses indicated that pup survival explained the most variability in the probability of extinction, regardless of whether or not the model incorporated density dependence. Adult survival and the standard deviation of pup survival were the next most important parameters in density-dependent simulations, whereas the severity and probability of catastrophe were more important during density-independent simulations. The inclusion of density dependence decreased the probability of extinction, but neither the abruptness nor the inclusion of density dependence were important model parameters. Results of both relative sensitivity analyses that altered each parameter by 10% of its range and life-stage-simulation analyses of deterministic matrix models supported the logistic regression results, indicating that pup survival and its variation were more important than other parameters. But both conventional sensitivity analysis of the stochastic model which changed each parameter by 10% of its mean value and elasticity analyses indicated that adult survival was more important than pup survival. We evaluated the advantages and disadvantages of using logistic regression to analyze the sensitivity of stochastic population viability models and conclude that it is a powerful method because it can address interactions among input parameters and can incorporate the range of parameter variability, although the standardized regression coefficients are not comparable between studies. Model structure, method of analysis, and parameter uncertainty affect the conclusions of sensitivity analyses. Therefore, rigorous model exploration and analysis should be conducted to understand model behavior and management implications. Resumen: Utilizamos la regresión logística como un método de análisis de sensibilidad par a un modelo de análisis de viabilidad poblacional de perros silvestres Africanos ( Lycaon pictus) y comparamos estos resultados con análisis de sensibilidad convencionales de modelos estocásticos y determinísticos. Coeficientes estandarizados de los análisis de regresión logística indicaron que la supervivencia de cachorros explicaba la mayor variabilidad en la probabilidad de extinción, independientemente de que el modelo incorporara la denso-dependencia. La supervivencia de adultos y la desviación estándar de la supervivencia de cachorros fueron los parámetros que siguieron en importancia en simulaciones de denso-dependencia, mientras que la severidad y la probabilidad de catástrofes fueron más importantes durante simulaciones denso-independientes. HIGH Summary., Gene transfer is a novel area of therapeutics in which the active agent is a nucleic acid rather than a protein or small molecule. As early as 1997, investigators reported long-term expression of therapeutic levels of factor IX using gene transfer techniques in hemophilia B mice, and similar data were thereafter reported in mice with hemophilia A. Efforts to translate these results to hemophilic dog models at first yielded only marginally therapeutic levels (1%,2% normal circulating levels), but within the past few years have achieved levels in the range of 10%,20% through multiple different gene transfer strategies. Early phase clinical testing has revealed that many aspects of gene transfer in humans were accurately predicted by studies in hemophilic dogs, but that other aspects were not, and were only appreciated as a result of clinical testing. Studies in the next few years will determine whether the problems identified in preclinical and early phase clinical testing can be solved to develop a therapeutic gene transfer approach to hemophilia. Pharmacol. Therap.33, 109,117. Historically, aldosterone receptor antagonists (ARA) have been classified as ,potassium sparing diuretics'. However, the positive effect of spironolactone, the most extensively studied ARA, on morbidity and mortality observed in humans suffering cardiac insufficiency could not be explained by the renal effect of the drug alone, and a pivotal clinical study has led to extensive research. Many experimental studies have demonstrated that ARA have previously unexpected beneficial effects on the cardiovascular system including reduction in remodelling of the vascular smooth muscle cells and myocytes and improvement of endothelial cell dysfunction in heart failure. These effects improve vascular compliance and slow down the progression of left ventricular dysfunction and end-organ damage. Furthermore, aldosterone receptor blockade also restores the baroreceptor reflex, improving heart rate variability in heart failure in humans. Some of these effects have been demonstrated in dog models of cardiac disease and so justified further investigation of the potential benefit of ARA in dogs with congestive heart failure (CHF). Positive effects of spironolactone on morbidity and mortality appear to have been seen in studies conducted in dogs suffering from naturally occurring CHF. In addition, eplerenone has been shown to have benefits in canine models of heart failure. The precise mechanisms by which ARA produce these beneficial effects in dogs remain to be determined but this group of drugs clearly provide therapeutic actions out-with their diuretic effects. Coulet The pharmacodynamic properties of a new veterinary fluoroquinolone antimicrobial agent, ibafloxacin, were evaluated. Minimal inhibitory concentrations (MIC), time-kill kinetics, postantibiotic effect (PAE) and postantibiotic subminimal inhibitory concentration effects (PA-SME) were determined against pathogenic canine Gram-negative and Gram-positive bacterial isolates from dermal, respiratory and urinary tract infections. The synergistic interactions between ibafloxacin and its main metabolite, 8-hydroxy-ibafloxacin were investigated. Finally, the efficacy of ibafloxacin was tested in in vivo canine infection models. Ibafloxacin had good activity against Pasteurella spp., Escherichia coli, Klebsiella spp., Proteus spp. and Staphylococcus spp. (MIC90=0.5 µg/mL), moderate activity against Bordetella bronchiseptica, Enterobacter spp. and Enterococcus spp. (MIC50=4 µg/mL) and low activity against Pseudomonas spp. and Streptococcus spp. The time-killing analysis confirmed that ibafloxacin was bactericidal with a broad spectrum of activity. The PAE and PA-SME were between 0.7,2.13 and 1,11.5 h, respectively. Finally, studies in dog models of wound infection and cystitis confirmed the efficacy of once daily oral ibafloxacin at a dosage of 15 mg/kg. Additional studies are needed to better define the importance of AUC/MIC (AUIC) and Cmax/MIC ratios on the outcome of fluoroquinolone therapy in dogs. [source] The effect of capsaicin on blood glucose, plasma insulin levels and insulin binding in dog modelsPHYTOTHERAPY RESEARCH, Issue 5 2001I. Tolan Abstract Capsicum frutescens has been used to treat diabetes mellitus by traditional healers in Jamaica. This study was designed to identify any hypoglycaemic principle(s) and to determine the mechanism of action. Purification experiments employing thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) led to the extraction of the active principle, capsaicin. Capsaicin caused a decrease in blood glucose levels of 4.91,±,0.52 (n,=,6),mmol/dL versus 6.40,±,0.13,mmol/dL (n,=,6) for the control (p,<,0.05) at the 2.5,h time interval when the oral glucose tolerance test (OGTT) was performed on dogs treated with capsaicin and compared with the control. Plasma insulin levels measured at the 2.5,h time interval showed that there was an increase in plasma insulin levels of 5.78,±,0.76,µIU/mL (n,=,6) for the capsaicin treated dogs versus 3.70,±,0.43,µIU/mL (n,=,10) for the control (p,<,0.05). Insulin receptor studies, using a modification of the method of Gambhir et al. done on monocytes obtained from blood at the 2.5,h time interval showed that there was a decrease in the percentage receptor binding for the capsaicin treated dogs when compared with the control. Insulin affinity results showed that there was a decrease of 2.4,×,10,4 in monocytes for the capsaicin treated dogs versus 8.77,×,10,4 for the control (p,<,0.05). Also, insulin receptor calculations showed a decrease in number, 2.63,×,108,±,5.73,×,107, compared with 8.77,×,108,±,1.47,×,108 for the control. In conclusion it can be stated that capsaicin is responsible for the hypoglycaemic episodes seen in the dogs and that it also causes an increase in insulin secretion which leads to a reduction of insulin binding on the insulin receptors.
