Biophysical studies of the development of amyloid fibrils from a peptide fragment of cold shock protein B

The peptide CspB-1, which represents residues 1,22 of the cold shock protein CspB from Bacillus subtilis, has been shown to form amyloid fibrils when solutions containing this peptide in aqueous (50%) acetonitrile are diluted in water [M. Großet al. (1999) Protein Science8, 1350,1357] We established conditions in which reproducible kinetic steps associated with the formation of these fibrils can be observed. Studies combining these conditions with a range of biophysical methods reveal that a variety of distinct events occurs during the process that results in amyloid fibrils. A CD spectrum indicative of ,,structure is observed within 1 min of the solvent shift, and its intensity increases on a longer timescale in at least two kinetic phases. The characteristic wavelength shift of the amyloid-binding dye Congo Red is established within 30 min of the initiation of the aggregation process and corresponds to one of the phases observed by CD and to changes in the Fourier transform-infrared spectrum indicative of ,,structure. Short fibrillar structures begin to be visible under the electron microscope after these events, and longer, well-defined amyloid fibrils are established on a timescale of hours. NMR spectroscopy shows that there are no significant changes in the concentration of monomeric species in solution during the events leading to fibril formation, but that soluble aggregates too large to be visible in NMR spectra are present throughout the process. A model for amyloid formation by this peptide is presented which is consistent with these kinetic data and with published work on a variety of disease-related systems. These findings support the concept that the ability to form amyloid fibrils is a generic property of polypeptide chains, and that the mechanism of their formation is similar for different peptides and proteins. Short-term dynamics of bacterial communities in a tidally affected coastal ecosystem

Abstract Tidal effects on the composition of free-living (FL) and particle-associated (PA) bacterial communities were studied in a tidal flat ecosystem in the southern North Sea. Denaturing gradient gel electrophoresis targeting the 16S rRNA gene and the 16S rRNA of Bacteria, Bacteroidetes, Alphaproteobacteria and the Roseobacter clade was applied. Despite strong tidal variations in the quantity and, depending on the season, also the quality of suspended matter as well as variations in bacterial activity, the bacterial community composition remained rather stable. FISH showed some variations of the community composition, but these were not related to typical tidal situations. Variations were higher during tidal cycles in May and July compared with November. Bacteroidetes, Alpha - and Gammaproteobacteria constituted the majority of the bacterial communities but relative proportions of the different groups varied considerably. On particles, Betaproteobacteria were also detected to substantial proportions. The Roseobacter clade constituted up to 90% of FL but only 30% of PA Alphaproteobacteria. Banding patterns of the Bacteroidetes -specific amplicons, and in particular those targeting the 16S rRNA, revealed tidally induced effects, as several bands appeared or disappeared at distinct events such as slack water or resuspension. Sequencing of prominent bands revealed predominantly phylotypes reported previously from this ecosystem. [source]


International Media's Role on U.S.,Small State Relations: The Case of Nepal

U.S. foreign policy relationships toward states with which it assumes limited geostrategic significance are often simplistic in design and misguided in their calculations because of the disproportionate weight given to the limited information from which policy is synthesized. International media outlets exacerbate this problem by underreporting, improperly framing stories, combining distinct events, piggybacking upon their domestic counterparts, encouraging simplifications, and misrepresenting reality on the ground. Recent U.S.,Nepal policy is a prototypical example, as a complex civil war with multiple actors was reduced in the eyes of U.S. policy makers to a simplistic terrorist uprising and treated as such until additional media attention propagated a substantial re-examination of policy. Although this case is more explanatory than predictive, this basic framework may enlighten a more nuanced overall understanding of U.S.,small state relations. [source]


Embryo developmental events and the egg case of the Aleutian skate Bathyraja aleutica (Gilbert) and the Alaska skate Bathyraja parmifera (Bean)

Embryo development events were correlated with egg-case changes for the Aleutian skate Bathyraja aleutica and the Alaska skate Bathyraja parmifera. Yolk absorption underwent two phases: that of steady absorption during early development and that of rapid yolk absorption during the final development stages. Total length (LT) for 50% of the pre-hatching embryos egg-case jelly disappearance was 92·04 mm (range 81,102 mm) and 99·36 mm (range 81,100 mm) for B. aleutica and B. parmifera, respectively, allowing the inner chamber to open to seawater flow. The tail filament underwent three phases of growth: rapid elongation during early development (<100 mm embryo LT), stasis of tail filament length during the remainder of embryo development and rapid absorption soon after hatching. Complete tail filament development coincided with the disappearance of egg-case jelly. Clasper buds first developed at embryos >70 mm LT for both species and the sex ratio was 1:1 well before hatching. Egg cases that were devoid of an ova or developing embryo were c. 5·0 and 6·5% of the egg cases examined for B. aleutica and B. parmifera, respectively. Measurements showed that egg cases containing only egg jelly were smaller in both width and length than those possessing an ova. Embryo stages were punctuated with distinct events that correlated with egg case changes controlling the internal environment of the developing embryo. [source]


Habitat use, migration pattern and population dynamics of chevron snakehead Channa striata in a rainfed rice farming landscape

Habitat use, migration, mortality and growth of the chevron snakehead Channa striata in a rainfed rice farming landscape of north-east Thailand were studied through a tagging experiment. A total of 751 fish were captured, tagged and released during three distinct events in the late dry season, and in the beginning and at the end of the wet season. Rice fields provided the major wet season habitat for C. striata. Small trap ponds built to provide dry season habitat on farms provided 20% of catches and, if not harvested, could increase recruitment to the spawning stock by >30% despite accounting for <1% of dry season habitat by area. Migrations were localized (mostly <500 m). Up-migration from perennial to seasonal water bodies at the beginning of the wet season involved longer distances and took place over a longer time than down-migration at the end of the wet season. Natural mortality rates were extremely high, particularly during the period of down-migration. Fishing mortality rates were high in absolute terms, but contributed only 6,36% to the total mortality. Growth was seasonal with a maximum towards the end of the wet season. Snakeheads have successfully colonized the rainfed rice farming landscape, where populations can withstand intensive exploitation and respond well to aquatic habitat management on farmland. Phylogeographic analysis of a recent radiation of Enallagma damselflies (Odonata: Coenagrionidae)

Abstract A phylogenetic hypothesis revealed two recent radiations among species of Enallagma damselflies, and extensive ecological work suggests that both adaptive and nonadaptive processes are involved in these radiations. We analysed the geographical pattern of genetic variability at 868 bp of mitochondrial DNA (mtDNA) among 283 individuals of 5 species displaying little ecological differentiation to identify the ancestral lineage, support their independent evolutionary trajectories and identify historical events and the underlying mechanism for one of these radiations. Nested clade analysis results clearly support a past event of range fragmentation in E. hageni. These Atlantic and Continental hageni races experienced distinct dispersal histories and still maintain nearly nonoverlapping ranges All four other species derive from the Continental hageni. Whereas three species endemic to the Atlantic coastal plain show little genetic variation, E. ebrium shared several haplotypes with the Continental hageni. Contrasting levels of genetic differentiation between E. hageni and E. ebrium in geographical areas associated with distinct events of E. hageni's recent history support the recent origin of this species. Altogether, our results are compatible with a process of radiation via divergence in mate recognition systems within the Continental hageni race following secondary contacts between putative refugial races.
