Solid Solubility and Precipitation in a Single-Crystal Alumina,Zirconia System

Solid solubility was examined in Zr-doped sapphire and Al-doped yttria-stabilized zirconia (YSZ) single crystals from 1200° to 1600°C. Specimens were fabricated via ion implantation of single crystals, followed by annealing in air. Secondary ion mass spectroscopy (SIMS) was used to quantify solute redistribution during annealing. Comparison of SIMS results with analytical electron microscopy (AEM) revealed an alumina solubility of 0.2,0.3 wt% in zirconia, and a zirconia solubility of 0.004,0.027 wt% in alumina. Direct imaging of zirconia precipitates revealed that tetragonal zirconia precipitates from supersaturated sapphire with the following orientation relationship: (100)tetragonal, (0001)sapphire and [01¯1]tetragonal, [12¯10]sapphire. A Graphene Oxide,Streptavidin Complex for Biorecognition , Towards Affinity Purification

Abstract In our postgenomic era, understanding of protein-protein interactions by characterizing the structure of the corresponding protein complex is becoming increasingly important. An important problem is that many protein complexes are only stable for a few minutes. Dissociation will occur when using the typical, time-consuming purification methods such as tandem affinity purification and multiple chromatographic separations. Therefore, there is an urgent need for a quick and efficient protein-complex purification method for 3D structure characterization. The graphene oxide (GO)·streptavidin complex is prepared via a GO·biotin·streptavidin strategy and used for affinity purification. The complex shows a strong biotin recognition capability and an excellent loading capacity. Capturing biotinylated DNA, fluorophores and Au nanoparticles on the GO·streptavidin complexes demonstrates the usefulness of the GO·streptavidin complex as a docking matrix for affinity purification. GO shows a high transparency towards electron beams, making it specifically well suited for direct imaging by electron microscopy. The captured protein complex can be separated via a filtration process or even via on-grid purification and used directly for single-particle analysis via cryo-electron microscopy. Therefore, the purification, sample preparation, and characterization are rolled into one single step. [source]


Direct atomic force microscopy observations of monovalent ion induced binding of DNA to mica

Summary Multivalent ions in solution are known to mediate attraction between two like-charged molecules. Such attraction has proved useful in atomic force microscopy (AFM) where DNA may be immobilized to a mica surface facilitating direct imaging in liquid. Theories of DNA immobilization suggest that either ,salt bridging' or fluctuation in the positions of counter ions about both the mica surface and DNA backbone secure DNA to the mica substrate. Whilst both theoretical and experimental evidence suggest that immobilization is possible in the presence of divalent ions, very few studies identify that such immobilization is possible with monovalent ions. Here we present direct AFM evidence of DNA immobilized to mica in the presence of only monovalent ions. Our data depict E. coli plasmid pBR322 adsorbed onto the negatively charged mica both after short (10 min) and long (24 h) incubation periods. These data suggest the need to re-explore current theories of like-charge attraction to include the possibility of monovalent interactions. We suggest that this DNA immobilization strategy may offer the potential to image natural processes with limited immobilization forces and hence enable maximum conformational freedom of the immobilized biomolecule. Matrix-assisted laser desorption/ionization imaging mass spectrometry of oxaliplatin derivatives in heated intraoperative chemotherapy (HIPEC)-like treated rat kidney

Oxaliplatin [1,2-diaminocyclohexane (dach)-Pt complex] is a platinum anticancer drug which is mainly used in the treatment of advanced colorectal cancer, particularly in Heated Intraoperative Chemotherapy (HIPEC) for the treatment of colorectal peritoneal carcinomatosis. In order to better understand the penetration of oxaliplatin in treated tissues we performed a direct imaging of tissue sections from HIPEC-like treated rat kidney using matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. This procedure allowed the detection and localization of oxaliplatin and its metabolites, the monocysteine and monomethionine complexes, in kidney sections. Specifically, oxaliplatin and its metabolites were localized exclusively in the kidney cortex, suggesting that it did not penetrate deeply into the organ. Based on these results, an imaging analysis of human tumors collected after HIPEC is currently in progress to assess the distribution of oxaliplatin and/or metabolites with the aim of defining clinical conditions to improve drug penetration. Realisation of a fully-deterministic microlensing observing strategy for inferring planet populations,

Abstract Within less than 15 years, the count of known planets orbiting stars other than the Sun has risen from none to more than 400 with detections arising from four successfully applied techniques: Doppler-wobbles, planetary transits, gravitational microlensing, and direct imaging. While the hunt for twin Earths is on, a statistically well-defined sample of the population of planets in all their variety is required for probing models of planet formation and orbital evolution so that the origin of planets that harbour life, like and including ours, can be understood. Given the different characteristics of the detection techniques, a complete picture can only arise from a combination of their respective results. Microlensing observations are well-suited to reveal statistical properties of the population of planets orbiting stars in either the Galactic disk or bulge from microlensing observations, but a mandatory requirement is the adoption of strictly-deterministic criteria for selecting targets and identifying signals. Here, we describe a fully-deterministic strategy realised by means of the ARTEMiS (Automated Robotic Terrestrial Exoplanet Microlensing Search) system at the Danish 1.54-m telescope at ESO La Silla between June and August 2008 as part of the MiNDSTEp (Microlensing Network for the Detection of Small Terrestrial Exoplanets) campaign, making use of immediate feedback on suspected anomalies recognized by the SIGNALMEN anomaly detector. We demonstrate for the first time the feasibility of such an approach, and thereby the readiness for studying planet populations down to Earth mass and even below, with ground-based observations. While the quality of the real-time photometry is a crucial factor on the efficiency of the campaign, an impairment of the target selection by data of bad quality can be successfully avoided. With a smaller slew time, smaller dead time, and higher through-put, modern robotic telescopes could significantly outperform the 1.54-m Danish, whereas lucky-imaging cameras could set new standards for high-precision follow-up monitoring of microlensing events (© 2010 WILEY-VCH Verlag GmbH & Co. IR spectroscopy of COmosphere dynamics with the CO ?rst overtone band

Abstract We discuss observations of the weak ?rst overtone (,, = 2) CO absorption band near 2300 nm with the U.S. National Solar Observatory Array Camera (NAC), a modern mid-infrared detector. This molecular band provides a thermal diagnostic that forms lower in the atmosphere than the stronger fundamental band near 4600 nm. The observed center-to-limb increase in CO line width qualitatively agrees with the proposed higher temperature shocks or faster plasma motions higher in the COmosphere. The spatial extent of chromospheric shock waves is currently at or below the diffraction limit of the available CO lines at existing telescopes. Five minute period oscillations in line strength and measured Doppler shifts are consistent with the p-mode excitation of the photospheric gas. We also show recent efforts at direct imaging at 4600 nm. We stress that future large-aperture solar telescopes must be teamed with improved, dynamic mid-infrared instruments, like the NAC, to capitalize on the features that motivate such facilities (© 2010 WILEY-VCH Verlag GmbH & Co. Increased nitric oxide production in nasal epithelial cells from allergic patients , RT-PCR analysis and direct imaging by a fluorescence indicator: DAF-2 DA*

Background Nitric oxide (NO) is believed to participate in the regulation of airway clearance and non-specific cellular immunity. Recent studies have suggested that airway epithelial cells of allergic and non-allergic individuals may differ in their ability to produce this molecule. Objective The aim of this study was to detect the difference in NO production in human nasal epithelial cells between normal subjects and patients with perennial allergic rhinitis (AR), and to assess the relationship between the expression of nitric oxide synthase (NOS) isoforms and the severity of the disease. Methods Nasal epithelial cells were obtained from the inferior turbinate. The expression of mRNAs encoding constitutive endothelial NOS (eNOS) and inducible NOS (iNOS) was studied by reverse transcription-polymerase chain reaction (RT-PCR). Direct NO production in living cells was visualized and quantified by a fluorescent indicator, DAF-2 DA. Results RT-PCR analysis demonstrated that AR patients with a RAST score of 5 or 6 showed significant increases in the levels of iNOS mRNA and slight reductions in those of eNOS mRNA. Patients with a RAST score of 2,4 also revealed the same tendency however, the difference was not significant. DAF-2 DA imaging demonstrated that epithelial cells, especially the ciliated cells, produced a larger amount of NO than non-epithelial inflammatory cells. Preincubation with L-NAME resulted in an approximate 40% decrease in both groups. Conclusion These results directly indicate that nasal epithelial cells of AR patients overall produce higher levels of NO through the concomitant expression of different NOS isoforms. Continuous NO production by the epithelial cells in normal subjects further support the hypothesis that NO derived from epithelium may play dual roles in the regulation of nasal airway clearance and in the host defense. In addition, the use of DAF-2 DA provides a reliable method to visualize and quantify the direct NO production of living cells. [source]
