Downstream variation in bed sediment size along the East Carpathian rivers: evidence of the role of sediment sources

Abstract Taking as an example six main rivers that drain the western flank of the Eastern Carpathians, a conceptual model has been developed, according to which fluvial bed sediment bimodality can be explained by the overlapping of two grain size distribution curves of different origins. Thus, for Carpathian tributaries of the Siret, coarse gravel joins an unimodal distribution presenting a right skewness with enhanced downstream fining. The source of the coarse material distributions is autohtonous (by abrasion and hydraulic sorting mechanisms). A second distribution with a sandy mode is, in general, skewed to the left. The source of the second distribution is allohtonous (the quantity of sand that reaches the river-bed through the erosion of the hillslope basin terrains). The intersection of the two distributions occurs in the area of the 0·5,8 mm fractions, where, in fact, the right skewness (for gravel) and left skewness (for sand) histogram tails meet. This also explains the lack of particles in the 0·5,8 mm interval. For rivers where fine sediment sources are low, the 0·5,8 mm fractions have a higher proportion than the fractions under 1 mm. For the Siret River itself, bed sediment bimodality is greatly enhanced due to the fact that the second mode is more than 25% of the full sample. As opposed to its tributaries, the source of the first mode, of gravel, is allohtonous to the Siret river, generated by the massive input of coarse sediment through the Carpathian tributaries, while the second mode, of the sands, is local. In this case we can also observe that the two distributions of particles of different origins overlap in the 0·5,8 mm fraction domain, creating the illusion of ,particle lack' in the fluvial bed sediments. Preference and performance of the sawfly Diprion pini on host and non-host plants of the genus Pinus

Abstract The sawfly, Diprion pini L., is a pest of Pinus in Europe and is mainly found on P. sylvestris L. and P. nigra laricio Poiret. The relative importance of female oviposition capacity and behaviour, egg development, and larval survival on a new host plant was measured on 11 pine species. Five were natural host plants and six non-host plants, five of which are not indigenous to Europe. Oviposition choice tests showed that females discriminated between the pine species. Egg and larval development also differed between pine species. However, the female choice was not linked with hatching rate and larval development. Results of biological tests clearly indicated that there were different response patterns of D. pini life stages in relation to pine species, and these patterns were the same with insects of four different origins. We discuss the importance of each potential barrier to colonisation of a new host. [source]


Ultra-trace analysis of multiple endocrine-disrupting chemicals in municipal and bleached kraft mill effluents using gas chromatography,high-resolution mass spectrometry

Abstract A comprehensive gas chromatographic,high-resolution mass spectrometric (GC-HRMS),based method was developed that permitted the simultaneous determination of 30 estrogenic endocrine-disrupting chemicals (EDCs) and related compounds, including surfactants, biogenic and synthetic steroids, fecal sterols, phytoestrogens, and plasticizers, in wastewater. Features of the method include low sample volume (,40 ml), optimized Florisil® cleanup to minimize matrix interferences and optimized analyte derivatization to improve sensitivity via GC-HRMS. Detection limits were in the low- to mid-ng/L range, and recoveries were greater than 60% for most target analytes. This new method allows for high throughput analysis of many organic wastewater contaminants in a complex matrix with relative standard deviation of less than 15% for most measurable compounds. The applicability of the method was demonstrated by examining wastewater samples from different origins. Compounds such as di(2-ethylhex-yl)phthalate, cholesterol, cholestanol, and other cholesterol derivatives were measured in much higher concentrations in untreated sewage and were reduced substantially in concentration by the treatment process. However, steroidal compounds, particularly estrone (E1), 17,-estradiol (E2), and estriol (E3), as well as plant sterols (except stigmastanol), were greater in the treated municipal wastewater versus the untreated effluent. Plant and fungi sterols, stigmastanol and ergosterol, were found largely associated with bleached kraft mill effluent (BKME) as compared to the municipal effluents. [source]


Forebrain projections to the hypothalamus are topographically organized in anurans: conservative traits as compared with amniotes

Abstract The organization of the forebrain in amphibians (anamniotes) is currently being re-evaluated in terms of evolution and several evidences have corroborated numerous traits shared by amphibians and amniotes, such as the organization of the basal ganglia and the amygdaloid complex. In the present study we have analysed the organization of forebrain afferent systems to the hypothalamus of the frog Rana perezi. In vivo and in vitro tract-tracing techniques with dextran amines and immunohistochemistry for localizing nitric oxide synthase (NOS) in a series of single or combined experiments were used as NOS labelling reveals hypothalamic afferents arising from the lateral amygdala and the combination allowed analysis of the relationship between fibers of different origins in the same section. The results showed a large segregation of afferents in the hypothalamic region depending on their site of origin in the forebrain. Four highly topographically organized prosencephalic tracts reaching the anuran hypothalamus were observed: (i) the medial forebrain bundle, from the medial pallium and septal complex; (ii) the caudal branch of the stria terminalis formed by fibers arising in the lateral and medial amygdala; (iii) part of the lateral forebrain bundle with fibers from the central amygdala and (iv) the dorsal thalamo-hypothalamic tract. Fibers coursing in each tract reach the hypothalamus and terminate in distinct fields. The resemblance in pattern of forebrain-hypothalamic organization between amphibians and amniotes suggests that this feature represents an important trait conserved in the evolution of all tetrapods and therefore essential for the hypothalamic function. High rates of marital dissolution and easy access to divorce are not unprecedented, historically or cross-culturally. But contemporary divorce in North America and Western Europe has different origins and features than divorce in previous cultures. The origins of modern divorce patterns date back more than 200 years, to the invention of the historically unprecedented idea that marriage should be based on love and mutual affection. Ironically, then, the fragility of modern marriage stems from the same values that have elevated the marital relationship above all other personal and familial commitments: the concentration of emotion, passion, personal identity, and self-validation in the couple relationship and the attenuation of emotional attachments and obligations beyond the conjugal unit. The immediate causes of divorce may range from factors as diverse as the personal psychological characteristics of one or both spouses to the stresses of economic hardship and community disintegration. But in a larger perspective, the role of divorce in modern societies and its relatively high occurrence both flow from the same complex of factors that have made good marriages so much more central to people's happiness than through most of the past, and deterioration of a marital relationship so much more traumatic. Volatile compounds after spontaneous fermentation of musts from Vitis vinifera cv. Albariño grapes cultivated in different origins from Rías Baixas AOC, Spain

Abstract White wines experimentally produced from the white grape variety Albariño from three different areas from Rias Baixas AOC, north-western Spain, have been analysed in this study. The contents of terpenes, C13 -norisoprenoids, alcohols, acetates and ethyl esters were determined by GC,MS. The Albariño wines from O Rosal are characterized by a high content of higher alcohols, while wines from Val do Salnés show the highest concentrations of free terpenes, acetates and ethyl esters; and wines of Condado do Tea show the highest concentrations of C13 -norisoprenids, principally due to the ,-ionone. Two principal components accounting for 90.7% of the total variance were calculated by means of a principal components analysis (PCA), and the graphical representation of these two components allows grouping the wines according their respective origins. Volatile compounds with fruity and floral odours showed the highest odour activity values, contributing in a great measure to the aroma of Albariño wines. Copyright © 2006 John Wiley & Sons, Ltd. Magnetic investigation of heavy metals contamination in urban topsoils around the East Lake, Wuhan, China

SUMMARY Magnetic measurements and heavy metal analyses were performed on 133 samples from the urban soils around the East Lake in Wuhan, China. Samples were collected from four areas with different environmental settings: a heavy industrial area well known for thermal power generation and steel works; villages located in the downwind area of the industrial area; a main road with heavy traffic and roads around the East Lake. Results show that concentrations of magnetic particle and heavy metals in urban topsoils are significantly elevated due to the input of coarser-grained magnetite from industrial (e.g. power generation and steel production) and other anthropogenic activities (e.g. vehicle emissions). Concentration-related magnetic parameters, for example, magnetic susceptibility, saturation isothermal remanent magnetization and anhysteretic remanent magnetization, significantly correlate with the concentration of heavy metals. Moreover, in terms of grain sizes, the magnetic particles of different origins can be efficiently discriminated at the studied region. Therefore, magnetic measurements provide a basis for discrimination and identification of different contamination sources, and can be used as an economic alternative to chemical analysis when mapping heavy metal contamination in urban soil around the East Lake region, Wuhan, China. Magnetic grain-size distribution of the enhanced component in the loess,palaeosol sequences in the western Loess Plateau of China

SUMMARY Rock magnetic measurements of Chinese loess,palaeosol samples from the Beiyuan section in Linxia City suggest the presence of two magnetic components: a background component and an enhanced component. The magnetic properties of the enhanced component suggest strong grain-size control, which is in contrast with variable grain-size distribution in loess,palaeosol from the central Loess Plateau. Chemically formed magnetic grains do not fulfil the requirements in the case of the western Loess Plateau because they may show shifts in the grain-size distribution. The difference in climate between the western and central Loess Plateau may lead to different origins and different preservation conditions of the pedogenic magnetite. [source]


Determination of Selenium Concentration in Sixty Five Reference Materials for Geochemical Analysis by GFAAS after Separation with Thiol Cotton

Samples were decomposed with a mixture of nitric and hydrofluoric acids. Selenium was reduced to SeIV with hydrochloric acid, and then fixed and separated from the matrix on thiol cotton. After digestion of the thiol cotton in hot nitric acid, the Se concentration was measured using palladium and magnesium nitrates as a matrix modifier. The limit of determination was 0.02 ,g g,1, the precision of the results (relative standard deviation of 3 to 8 replicates) varied from 2.6 to 17.7% with an average of 7.9% in the range 0.02-42.7 ,g g,1 and was similar to the value obtained for synthetic samples. Our results are in good agreement with available literature values. Le sélénium a été dosé sur 65 standards géochimiques de référence de différentes provenances par spectrométrie d'absorption atomique avec four graphite. Les échantillons ont été mis en solution par une attaque acide HNO3 -HF. Le sélénium a été réduit en SeIV avec de l'acide chlorhydrique, fixé et séparé de la matrice sur du coton thiol. Après décomposition du coton thiol par chauffage avec de l'acide nitrique, les concentrations en sélénium ont été mesurées en utilisant du nitrate de palladium et de magnésium comme modificateur de matrice. La limite de détermination est de 0.02 ,g g,1, la précision des résultats (écart type relatif sur 3 à 8 mesures) pour les standards géochimiques varie de 2.6 à 17.7% avec une moyenne de 7.9% dans la game 0.02-42.7 ,g g,1et est comparable à celle obtenue pour des échantillons synthétiques. Nos résultats sont en bonne concordance avec les résultats déjà publiés dans la littérature. [source]


Generation of endoderm-derived human induced pluripotent stem cells from primary hepatocytes,

Recent advances in induced pluripotent stem (iPS) cell research have significantly changed our perspective on regenerative medicine. Patient-specific iPS cells have been derived not only for disease modeling but also as sources for cell replacement therapy. However, there have been insufficient data to prove that iPS cells are functionally equivalent to human embryonic stem (hES) cells or are safer than hES cells. There are several important issues that need to be addressed, and foremost are the safety and efficacy of human iPS cells of different origins. Human iPS cells have been derived mostly from cells originating from mesoderm and in a few cases from ectoderm. So far, there has been no report of endoderm,derived human iPS cells, and this has prevented comprehensive comparative investigations of the quality of human iPS cells of different origins. Here we show for the first time reprogramming of human endoderm-derived cells (i.e., primary hepatocytes) to pluripotency. Hepatocyte-derived iPS cells appear indistinguishable from hES cells with respect to colony morphology, growth properties, expression of pluripotency-associated transcription factors and surface markers, and differentiation potential in embryoid body formation and teratoma assays. In addition, these cells are able to directly differentiate into definitive endoderm, hepatic progenitors, and mature hepatocytes. Conclusion: The technology to develop endoderm,derived human iPS cell lines, together with other established cell lines, will provide a foundation for elucidating the mechanisms of cellular reprogramming and for studying the safety and efficacy of differentially originated human iPS cells for cell therapy. For the study of liver disease pathogenesis, this technology also provides a potentially more amenable system for generating liver disease-specific iPS cells. Effects of restricted thoracic movement on the regional distribution of ventilation

Background: Restricted thoracic movement is often encountered in patients, necessitating mechanical ventilation during surgery or intensive care treatment. High intraabdominal pressure, obesity or thorax rigidity and deformity reduce the chest distensibility and deteriorate the lung function. They render the selection of proper ventilator settings difficult and complicate the weaning process. Electrical impedance tomography (EIT) is currently being proposed as a bedside imaging method for monitoring regional lung ventilation. The objective of our study was to establish whether the effects of decreased chest compliance on regional lung ventilation can be determined by EIT. Methods: Ten healthy male volunteers were studied in our pilot study under three conditions: (1) unrestricted breathing and (2) restricted breathing by abdominal and (3) lower rib cage strapping. The subjects were followed during spontaneous tidal breathing in five postures (sitting, supine, prone, left and right side). EIT and spirometry data were acquired in each condition. Results: The distribution of ventilation in subjects with unrestricted breathing corresponded with the physiologically expected values. In the left and right lateral postures, abdominal and thoracic cage restrictions reduced the ventilation in the dependent lung areas; the non-dependent areas were unaffected. In the prone position, the ventilation of the dependent and non-dependent areas was reduced. The effects of strapping were least pronounced in the supine posture. Conclusions: We conclude that EIT is able to measure changes in the regional distribution of ventilation induced by restricted chest movement and has the potential for optimising artificial ventilation in patients with limited chest compliance of different origins. A study on the temporal dynamics of tourism demand in the Asia Pacific Region

Abstract Using the monthly statistics of tourists from four different origins (America, Europe, Asia and Oceania) among seven places in the Asia Pacific region for a period from January 1991 to December 2005, we investigate these 28 time series using the Holt-Winters method, artificial neural network and numerical graphical plots. Interesting comparisons on level fluctuations, trends and seasonal patterns of the time series among the Asia Pacific region are explored. These findings enable us to understand more about the temporal aspect of tourism demand in the Asia Pacific region. Small intestinal digestibility of microbial and endogenous amino acids in dairy cows

The small intestinal digestibility of microbial amino acids and the apparent re-absorption of endogenous amino acids were estimated in three ruminal, duodenal and ileal fistulated lactating dairy cows. The cows were assigned to a 3 × 3 Latin square and fed diets low in amino acids, composed of straw, starch, molasses and urea. The treatments were three straw to concentrate ratios. The digestibilities and re-absorptions were estimated from duodenal and ileal flows of amino acids from feed, microbial and endogenous sources. The duodenal distribution of amino acids between different origins was determined by the difference method and by the amino acid profile method. The ileal distribution was estimated by a simple method where the feed fraction was estimated by the mobile bag method, the endogenous fraction as the water soluble fraction and the microbial fraction as the rest. In ileal samples, 8.0 ± 1.1, 52.2 ± 2.9 and 39.8 ± 2.6% of total amino acid nitrogen (AAN) were of feed, microbial and endogenous origin, respectively, compared with 5.1 ± 0.9, 61.6 ± 6.6 and 32.7 ± 6.9% in duodenal samples. The small intestinal digestibility of microbial AAN was estimated to be 75.1 ± 1.8% and the apparent re-absorption of endogenous AAN to be 62.3 ± 11.1%. [source]


Evidence for dose-dependent effects on plant growth by Stenotrophomonas strains from different origins

Abstract Aims: To assess the influence of Stenotrophomonas on plants, the interaction of 16 Stenotrophomonas strains from clinical and environmental sources with strawberry plant seedlings was analysed. Methods and Results:In vitro, all Stenotrophomonas strains influenced plant growth when applied to seedlings. Whereas most of the Stenotrophomonas strains promoted root growth and hair development, a statistically significantly negative influence on the length of stem was found. Although strains from a clinical origin also showed statistically significant effects on plants, this was generally lower when compared with environmental strains. For three selected strains, a strong dose-dependent effect was observed for all parameters. In vitro, a correlation was found between plant growth promotion and production of a plant growth hormone, indole-3-acetic acid (IAA). Xanthomonas campestris, a phylogenetically very closely related species to Stenotrophomonas, was used as a phytopathogenic control. It too confirmed the reduction of plant growth in this in vitro system. Conclusions: Independent of their origin, Stenotrophomonas strains can produce IAA in vitro and subsequently, influence plant growth. The effect of Stenotrophomonas presence on plants was dose-dependent. Significance and Impact of the Study: The dose-dependent effect of Stenotrophomonas, a bacterium of both biotechnological and medical interest, is of great interest for biocontrol applications of plant-associated strains. This paper is the first report that clearly demonstrates the phytopathogenic capacity of Stenotrophomonas. [source]


Prevalence and virulence properties of Vibrio cholerae non-O1, Aeromonas spp. and Plesiomonas shigelloides isolated from Cambé Stream (State of Paraná, Brazil)

The incidence of Vibrio cholerae, Aeromonas spp. and Plesiomonas shigelloides was determined in water samples from Cambé Stream. The samples were collected from seven different sites. The serogroups, virulence markers and drug resistance profiles were also evaluated. Twelve Aer. hydrophila, 12 Aer. caviae, eight Aer. sobria, seven Ple shigelloides and two V. cholerae non-O1 were isolated. They belonged to different serogroups and all produced haemolysis in different assays. Five of the Aeromonas strains and one of V. cholerae non-O1 were positive for enterotoxin activity. Haemagglutination and its inhibition, using erythrocytes of different origins, was variable for Aeromonas spp. and V. cholerae, while none of the Ple. shigelloides haemagglutinated in association with any type of erythrocyte. All isolates exhibited multiple drug resistance. These results indicate that the occurrence of V. cholerae non-O1, Aeromonas spp. and Ple. shigelloides, in water used for vegetable irrigation, human recreation and animal consumption, among others, represents a potential risk for humans. Evolution, origin and age of lineages in the Californian and Mediterranean floras

Abstract This paper addresses some of the conceptual issues involved in the analysis of the age and origin of mediterranean-climate plant taxa, paying particular attention to three topics: (1) the importance of an explicit time frame in the definition of biogeographical origins, (2) the distinction between the age of traits and the age of taxa, and (3) the idea of mediterranean-type ecosystems as environmental islands. (1) In California, recent analyses demonstrate that the diversity of species derived from different biogeographical origins is significantly correlated with temperature and precipitation gradients. These patterns support the hypothesis that niche conservatism is an important factor structuring modern diversity gradients. However, depending on how far back in time one looks, a species may be assigned to different origins; future discussions of biogeographical origins need to address the appropriate time frame for analysis. (2) Past research has demonstrated distinctive trait syndromes among woody plants of the Mediterranean, Chile, California and Mexico, and proposed that the syndromes are associated with lineages of different age in these floras. Reanalysis of individual traits demonstrates greater variability among regions than previously reported. The classification of plants into ,old' and ,new' genera is re-evaluated, and it is suggested that greater attention be paid to the age of traits, rather than to the age of taxa, especially at an arbitrary rank such as genus. (3) The idea of mediterranean-climate regions as ,climatic islands' is examined. Space,time diagrams of climate enable one to view the emergence of distinctive climatic regions in a continental context. The terms ,synclimatic' and ,anticlimatic' are proposed, referring to migration routes that parallel climate contours in space and time versus those that cross contours (including the case of geographic stasis in the face of climate change), respectively. Mediterranean-climate regions have served as important case studies in plant ecology and evolution, and merit continued close examination in the light of continued advances in phylogenetics and palaeoecology. Use of sweet sorghum juice for lactic acid fermentation: preliminary steps in a process optimization

Abstract BACKGROUND: Lactic acid has many applications in the chemical industries and it can be produced economically by microorganisms using biomass raw materials of different origins. Sweet sorghum juice is a high sugar content raw material with potential for lactic acid production because after hydrolysis of its sucrose content the remaining glucose and fructose can supply the carbon demand of most lactic acid bacteria. However, satisfying the nitrogen and B-vitamin needs of the bacteria by supplementation with yeast extract and/or other alternative nitrogen-containing supplements can make the process too expensive. RESULTS: Using a statistical optimization process much of the yeast extract can be replaced by a cheaper alternative nitrogen source, namely wheat gluten. This resulted in a fermentation with 99% lactic acid yield and 3.04 g L,1 h,1 volumetric productivity. CONCLUSION: Using response surface methodology (RSM) media optimization was performed for lactic acid fermentation with an industrially acceptable result, reducing the costs of raw materials by half, replacing yeast extract by an alternative nitrogen source and applying yeast extract only as a source of micro-elements (vitamins, salts, etc.) Copyright © 2010 Society of Chemical Industry [source]


No correlation of five gene polymorphisms with periodontal conditions in a Greek population

Abstract Background: Various studies have examined possible correlations between a number of cytokine gene polymorphisms and periodontal disease in populations of different origins. The present study sought the correlation between four single-nucleotide polymorphisms (IL1A+3954, IL1B+4845, TNFA,308, COL1A1 Sp1), a variable number of tandem repeats polymorphism (IL1RN intron 2) and periodontal conditions in subjects of Greek origin. Methods: One hundred and ninety-two healthy subjects, stratified as non-periodontitis and periodontitis (chronic and aggressive) cases, participated in the present study. Genotyping was performed by polymerase chain reaction-based techniques using the primers and conditions described in the literature. The frequencies of genotypes between study groups were compared using Genepop v3.3 genetic software and Instat statistical package. Results: No differences were observed among the groups concerning the distributions of genotypes under investigation. Conclusions: Carriage rates of the polymorphisms under investigation in systemically healthy subjects of Greek origin are well within the range reported for Caucasians but these polymorphisms cannot discriminate between non-periodontitis and periodontitis (chronic or aggressive) cases. Detecting local adaptation in a natural plant,pathogen metapopulation: a laboratory vs. field transplant approach

Abstract Antagonistic coevolution between hosts and parasites in spatially structured populations can result in local adaptation of parasites. Traditionally parasite local adaptation has been investigated in field transplant experiments or in the laboratory under a constant environment. Despite the conceptual importance of local adaptation in studies of (co)evolution, to date no study has provided a comparative analysis of these two methods. Here, using information on pathogen population dynamics, I tested local adaptation of the specialist phytopathogen, Podosphaera plantaginis, to its host, Plantago lanceolata at three different spatial scales: sympatric host population, sympatric host metapopulation and allopatric host metapopulations. The experiment was carried out as a field transplant experiment with greenhouse-reared host plants from these three different origins introduced into four pathogen populations. In contrast to results of an earlier study performed with these same host and parasite populations under laboratory conditions, I did not find any evidence for parasite local adaptation. For interactions governed by strain-specific resistance, field studies may not be sensitive enough to detect mean parasite population virulence. Given that parasite transmission potential may be mediated by the abiotic environment and genotype-by-environment interactions, I suggest that relevant environmental variation should be incorporated into laboratory studies of parasite local adaptation. DIFFERENTIATION OF CURED COOKED HAMS BY PHYSICO-CHEMICAL PROPERTIES AND CHEMOMETRICS

ABSTRACT Comparison of physico-chemical and compositional traits was carried out on cooked hams. Deboned fresh pig thighs of three different origins were divided into three batches: 200 pig thighs from the Italian market, H1; 200 from The Netherlands, H2; and 200 from Denmark, H3. Boneless pig thighs were processed under commercial guidelines for production of cooked hams, using brine at 25% level of injection. After processing, 12 cooked hams from each batch were sampled randomly and analyzed for proximate and fatty acid composition. Color measurement was performed on the muscles: biceps femoris, semimembranosus, and semitendinosus. H1 hams showed a higher weight loss and a lower technological yield compared to H2 and H3 hams. Analysis of variance on compositional data showed that H1 hams had a lower moisture/protein ratio, a higher fat content, a lower percentage of, -linolenic, eicosapentaenoic and docosahexaenoic acid, and a higher percentage of myristic and palmitic acids when compared to H2 and H3 hams (P < 0.05). Analysis of color of the three muscles demonstrated that hams from the H1 group had the highest a* values. The application of linear discriminant analysis demonstrated that the use of only four variables allowed to correctly discriminate among groups of cooked hams. PRACTICAL APPLICATIONS The following are the practical applications of this research. The comparison of physico-chemical and compositional traits were carried out on cooked hams. Pig thighs of different origin were processed under commercial guidelines. The physicochemical parameters of cooked hams were defined and showed some differences characterizing the products. Raman identification of natural red to yellow pigments: ochre and iron-containing ores

Abstract Red and yellow natural (roasted) pigments have been used for millennia. This paper reports on the Raman characterization of about 50 pigments of the Ôkhra ,Mathériauthèque' collection from different origins: violet, red, orange, apricot, yellow ochre, Umbrian and Siena earths from France (Puisaye, Ardennes, Vaucluse), Italy (Siena, Sardinia, Venice, etc.), Germany, Sweden, Cyprus and India (Madras). Comparison is made with ,Bengara' Japanese pigments. Goethite is the chromophore of yellow pigments, and haematite (sometimes with magnetite) that of red ochre and earths. Umbrian pigment (,Terre d'Ombre') contains additional manganese oxides. Color shades are related to the content of secondary phases: iron oxides (haematite, goethite, magnetite), quartz, gypsum and anhydrite, calcite, dolomite, alums (jarosites), and to the Fe/M substitution in the haematite structure. Simultaneous determination of iridoids, phenolic acids, flavonoids, and saponins in Flos Lonicerae and Flos Lonicerae Japonicae by HPLC-DAD-ELSD coupled with principal component analysis

Abstract A method, HPLC coupled with diode-array and evaporative light scattering detectors (HPLC-DAD-ELSD), was newly developed to evaluate the quality of Flos Lonicerae (FL) and Flos Lonicerae Japonicae (FLJ), through a simultaneous determination of multiple types of bioactive components. By employing DAD, the detection wavelengths were set at 240 nm for the determination of iridoids, 330 nm for phenolic acids, and 360 nm for flavonoids, respectively. While ELSD, connected in series after DAD, was applied to the determination of saponins. This assay was fully validated with respect to precision, repeatability, and accuracy. Moreover, principal component analysis (PCA) was used for the similarity evaluation of different samples, and it was proven straightforward and reliable to differentiate FL and FLJ samples from different origins. For PCA, two principal components have been extracted. Principal component 1 (PC1) influences the separation between different sample sets, capturing 54.598% variance, while principal component 2 (PC2) affects differentiation within sample sets, capturing 12.579% variance. In conclusion, simultaneous quantification of bioactive components by HPLC-DAD-ELSD coupled with PCA would be a well-acceptable strategy to differentiate the sources and to comprehensively control the quality of the medicinal plants FL and FLJ. Variations in cadmium accumulation among rice cultivars and types and the selection of cultivars for reducing cadmium in the diet

Abstract A pot trial was conducted with 52 rice cultivars of different types collected from different origins. The results showed that there were great differences in Cd concentrations in straw, brown rice and grain chaff among the rice cultivars grown in a soil containing a Cd concentration of 100 mg kg,1; the Cd concentrations in brown rice ranged from 0.22 to 2.86 mg kg,1. The great genotypic differences in Cd concentrations indicated that it is possible to lower the Cd content of rice through cultivar selection and breeding. Significant differences were found in the Cd concentrations of the rice types of Indica consanguinity and those of Japonica consanguinity, but not between their origins. There were significant correlations between straw and brown rice in Cd concentration and in the total amount of Cd accumulated. The distribution ratios of the Cd accumulated in brown rice to the total Cd accumulation in the above-ground rice plant varied greatly from 12.9 to 137.8 g kg,1, and there was significant correlation between the distribution ratios and Cd concentrations in brown rice. These indicated that Cd concentration in rice grain is governed by the transport of Cd from root to shoot and also from shoot to grain. Cd concentrations in brown rice also correlated significantly with some important agronomic traits, as well as with nitrogen concentrations, one of the most important criteria for rice quality. Gemini near-infrared integral field spectroscopy of the narrow-line region of ESO 428,G14: kinematics, excitation and the role of the radio jet

ABSTRACT We present two-dimensional (2D) gas kinematics and excitation of the inner 300 pc of the Seyfert galaxy ESO 428,G14 at a sampling of 14 pc2, from near-infrared spectroscopic observations at R, 6000 obtained with the Integral Field Unit (IFU) of the Gemini Near-Infrared Spectrograph. From measurements of fluxes and profiles of the emission lines [Fe ii],1.257 ,m, Pa,, H2, 2.121 ,m and Br,, we construct 2D maps of line intensities and ratios, radial velocities and velocity dispersions. Emission line ,tomography' is provided by velocity slices obtained across the line profiles, a unique capability of IFUs, which allows the mapping of not only the peak velocities but including also the wings. We compare these maps with a previously published high spatial resolution radio map and find a tight relation between the radio structure and the emission-line flux distributions and kinematics, revealing that the radio jet plays a fundamental role not only in shaping the narrow-line region but also in the imprint of its kinematics. Blueshifts of up to 400 km s,1 and velocity dispersions of up to 150 km s,1 are observed in association with the radio jet at a position angle (PA) = 129°, which is also the PA of the photometric major axis of the galaxy. We conclude that the radio jet is launched at a small angle relative to the galactic plane, with the north-western side slightly oriented towards us. This angle is small enough for the radio jet to shock and compress the gas in the plane of the galaxy, and for the nuclear continuum to ionize and heat it. The distinct kinematics and flux distributions observed for the different emission lines suggest different origins for their emission. The [Fe ii] shows the largest blueshifts and velocity dispersions and its flux distribution is concentrated along the jet, while the H2 shows the lowest velocity dispersions and has additional flux contribution from regions beyond the jet. Both X-rays emitted by the active galactic nucleus and shocks produced by the radio jet can excite the H2 and [Fe ii] emission lines. We use the 2D velocity dispersion maps to estimate upper limits to the contribution of the radio jet to the excitation of [Fe ii] and H2 which may reach 90 per cent for [Fe ii] and 80 per cent for H2 in the jet region. The [Fe ii]/Pa, emission-line ratios and the association of the [Fe ii] flux distribution and kinematics with the radio structure support a stronger contribution of the radio jet to the [Fe ii] excitation than that of H2. In the regions beyond the jet, the observations favour X-ray excitation. [source]


Molecular evidence for multiple polyploidization and lineage recombination in the Chrysanthemum indicum polyploid complex (Asteraceae)

Summary ,,The Chrysanthemum indicum polyploid complex comprises morphologically differentiated diploids, tetraploids and hybrids between C. indicum and C. lavandulifolium. The relationships between species and cytotypes within this complex remain poorly understood. ,,Random amplified polymorphic DNAs (RAPDs), intersimple sequence repeats (ISSRs) and chloroplast SSR markers were used to elucidate the genetic diversity and relationships of the C. indicum polyploid complex. ,,Molecular analysis of three diploid and nine tetraploid populations provided strong evidence for recurrent origins and lineage recombination in the C. indicum polyploid complex. The high similarity in molecular marker profiles and cpDNA haplotypes between the diploids and tetraploids distributed in the Shen-Nong-Jia Mountain area of China suggested an autopolyploid origin of the tetraploids, while the tetraploids from other populations may have originated via allopolyploidization. Lineage recombination was revealed by the extensive sharing of chloroplast haplotypes and genetic markers among the tetraploid populations with different origins. ,,Multiple differentiation and hybridization/polyploidization cycles have led to an evolutionary reticulation in the C. indicum polyploid complex, and resulted in the difficulties in systematic classification. Is tile drainage water representative of root zone leaching of pesticides?

Abstract Given the methods presently available, determination of flux-averaged concentrations of pesticides in structured soils is always a compromise. Most of the available methods entail major uncertainties and limitations. Tile drainage monitoring has several advantages, but the extent to which it is representative of overall leaching has been questioned because it comprises a mixture of water of different origins. This literature review evaluates whether drainage water pesticide concentrations are representative of root zone leaching of pesticides. As there are no reports quantifying the extent to which the flux-averaged concentration of pesticides in drainage water differs from that found between the drains, evidence-based conclusions cannot be drawn. Nevertheless, the existing literature does suggest that the concentration in drainage water does not always correspond to the concentration at drain depth between the drains; depending on the conditions pertaining, the concentrations may be higher or lower. As to whether the flux-averaged concentration of pesticides in drainage water is representative of the interdrain concentration at drain depth it is concluded that (1) the representativeness of drainage water concentrations can be questioned on very well-drained soils and on poorly drained soils with little capacity for lateral transport beneath the plough layer, (2) the conditions provided by relatively porous soils and moderate climatic conditions are conducive to the drainage water concentration being representative and (3) drainage water will be more representative in the case of weakly sorbed pesticides than for strongly sorbed pesticides. Used critically, it is thus believed that drainage water concentrations can serve to characterize the flux-averaged concentration of pesticides at drain depth. However, the use of drainage water for determining average concentrations necessitates thorough investigation and interpretation of precipitation, percolation, drain outflow and concentration dynamics. Fluorescence in situ hybridization polymorphism using two repetitive DNA clones in different cultivars of wheat

Abstract Twenty-two wheat cultivars and a wheat line were analysed with two-colour fluorescence in situ hybridization (FISH) using the pSc119.2 and pAs1 repetitive DNA clones to detect if polymorphism could be observed in the hybridization patterns of different wheat cultivars. The FISH hybridization pattern of ,Chinese Spring' was compared with wheat cultivars of different origins. Differences were observed in the hybridization patterns of chromosomes 4A, 5A, 1B, 2B, 3B, 5B, 6B, 7B, 1D, 2D, 3D and 4D. Although a low level of polymorphism exists in the FISH pattern of different wheat cultivars, it is possible to identify 17 pairs of chromosomes according to their hybridization patterns with these two probes. This study will help to predict the expected variation in the FISH pattern when analysing wheat genetic stocks of different origin. It is presumed that variation in hybridization patterns are caused by chromosome structural rearrangements and by differences in the amount and location of repetitive sequences in the cultivars analysed. Synthesis, UV photo-polymerization and degradation study of PEG containing polyester polyol acrylates

Abstract Succinic acid-co-polyethylene glycol polyester polyol acrylates, capable of in situ polymerization with UV radiation, were synthesized and characterized. Effect of structural changes such as internal branching, external crosslinking on their polymerization and degradation properties were investigated. Use of external crosslinkers reduced the polymerizability of the polymer. Lipases of different origins expedited the degradation to different extents and lipase isolated from porcine pancreas appeared to be more efficient than lipases from other sources. Three different origins for apparent triploid/diploid mosaics

Abstract Four apparent triploid/diploid mosaic cases were studied. Three of the cases were detected at prenatal diagnosis and the other was of an intellectually handicapped, dysmorphic boy. Karyotypes were performed in multiple tissues if possible, and the inheritance of microsatellites was studied with DNA from fetal tissues and parental blood. Non-mosaic triploids have a different origin from these mosaics with simple digyny or diandry documented in many cases. Three different mechanisms of origin for these apparent mosaics were detected: (1) chimaerism with karyotypes from two separate zygotes developing into a single individual, (2) delayed digyny, by incorporation of a pronucleus from a second polar body into one embryonic blastomere, and (3) delayed dispermy, similarly, by incorporation of a second sperm pronucleus into one embryonic blastomere. In three of the four cases, there was segregation within the embryos of triploid and diploid cell lines into different tissues from which DNA could be isolated. In case 2 originating by digyny, the same sperm allele at each locus could be detected in both triploid and diploid tissues, which is supportive evidence for the involvement of a single sperm and for true mosaicism rather than chimaerism. Similarly, in case 4 originating by dispermy, the same single ovum allele at each locus could be detected in diploid and triploid tissues, confirming mosaicism. In the chimaeric case (case 3), the diploid line had the karyotype 47,XY,+16 while the triploid line was 69,XXY. This suggests a chimaera, since, in a true mosaic, the triploid line should also contain the additional chromosome 16. Supporting the interpretation of a chimaeric origin for this case, the DNA data showed that the triploidy was consistent with MII non-disjunction (i.e. involving a diploid ovum). In the mosaic cases (1, 2, 4), there was no evidence of the involvement of a diploid sperm or a diploid ova, and in triploid/diploid mosaicism, an origin from a diploid gamete is excluded, since all such conceptuses would be simple triploids. In one of these triploid/diploid mosaics detected at prenatal diagnosis by CVS, the triploid line seemed to be sequestered into the extra-fetal tissues (confined placental mosaicism). Membrane microdomains and proteomics: Lessons from tetraspanin microdomains and comparison with lipid rafts

Abstract Biological membranes are compartmentalized into microdomains that exhibit particular lipid and protein compositions. Membrane microdomains, such as tetraspanin-enriched microdomains and lipid rafts, have been suggested to play a role in a variety of physiological and pathological processes. Therefore, the characterization of the protein compositions of these microdomains, which is the focus of this review, appears to be a crucial step to better understanding their function. Proteomics has recently allowed the characterization of tetraspanin-enriched microdomains in colon cancer cells. This demonstrated the presence of different categories of membrane proteins and suggested a variation in the composition of tetraspanin-enriched microdomains during tumor progression. On the other hand, proteomics has permitted the identification of hundreds of proteins in lipid rafts of different origins. However, the diversity of methodologies in sample preparation and of strategies in protein identification led to a broad variability in the data obtained. These methodological issues are discussed. Moreover, proteomics has revealed that different sets of proteins were present in tetraspanin-enriched microdomains as compared with lipid rafts, strengthening the idea that these microdomains are distinct structures.
