Short-Term Traffic Volume Forecasting Using Kalman Filter with Discrete Wavelet Decomposition

Short-term traffic volume data are often corrupted by local noises, which may significantly affect the prediction accuracy of short-term traffic volumes. Discrete wavelet decomposition analysis is used to divide the original data into several approximate and detailed data such that the Kalman filter model can then be applied to the denoised data and the prediction accuracy can be improved. Two types of wavelet Kalman filter models based on Daubechies 4 and Haar mother wavelets are investigated. Traffic volume data collected from four different locations are used for comparison in this study. The test results show that both proposed wavelet Kalman filter models outperform the direct Kalman filter model in terms of mean absolute percentage error and root mean square error. [source]


Detecting Cycle Failures at Signalized Intersections Using Video Image Processing

Cycle failure detection is essential for identifying signal control problems at intersections. However, typical traffic sensors do not have the capability of capturing cycle failures. In this article, we introduce an algorithm for traffic signal cycle failure detection using video image processing. A cycle failure for a particular movement occurs when at least one vehicle must wait through more than one red light to complete the intended movement. The proposed cycle failure algorithm was implemented using Microsoft Visual C#. The system was tested with field data at different locations and time periods. The test results show that the algorithm works favorably: the system captured all the cycle failures and generated only three false alarms, which is approximately 0.9% of the total cycles tested. Some Learning Methods in Functional Networks

This article is devoted to learning functional networks. After a short introduction and motivation of functional networks using a CAD problem, four steps used in learning functional networks are described: (1) selection of the initial topology of the network, which is derived from the physical properties of the problem being modeled, (2) simplification of this topology, using functional equations, (3) estimation of the parameters or weights, using least squares and minimax methods, and (4) selection of the subset of basic functions leading to the best fit to the available data, using the minimum-description-length principle. Several examples are presented to illustrate the learning procedure, including the use of a separable functional network to recover the missing data of the significant wave height records in two different locations, based on a complete record from a third location where the record is complete. Motor units in cranial and caudal regions of the upper trapezius muscle have different discharge rates during brief static contractions

Abstract Aim:, To compare the discharge patterns of motor unit populations from different locations within the upper trapezius muscle during brief submaximal constant-force contractions. Methods:, Intramuscular and surface electromyographic (EMG) signals were collected from three sites of the right upper trapezius muscle distributed along the cranial-caudal direction in 11 volunteers during 10 s shoulder abduction at 25% of the maximum voluntary force. Results:, A total of 38 motor units were identified at the cranial location, 36 from the middle location and 17 from the caudal location. Initial discharge rate was greatest at the caudal location (P < 0.05; mean ± SD, cranial: 16.7 ± 3.6 pps, middle: 16.9 ± 4.0 pps, caudal: 19.2 ± 3.3 pps). Discharge rate decreased during the contraction for the most caudal location only (P < 0.05). Initial estimates of surface EMG root mean square values were highest at the most caudal location (P < 0.05; cranial: 32.3 ± 20.9 ,V, middle: 41.3 ± 21.0 ,V, caudal: 51.6 ± 23.6 ,V). Conclusion:, This study demonstrates non-uniformity of motor unit discharge within the upper trapezius muscle during a brief submaximal constant-force contraction. Location-dependent modulation of discharge rate may reflect spatial dependency in the control of motor units necessary for the development and maintenance of force output. [source]


The mental health of female sex workers

Objective:, There is limited information available about the mental health of female sex workers. Therefore, we aimed to make a comprehensive assessment of the mental status of female sex workers over different outdoors and indoors work settings and nationalities. Method:, As the prerequisites of a probability sampling were not given, a quota-sampling strategy was the best possible alternative. Sex workers were contacted at different locations in the city of Zurich. They were interviewed with a computerized version of the World Health Organization Composite International Diagnostic Interview. Additional information was assessed in a structured face-to-face interview. Results:, The 193 interviewed female sex workers displayed high rates of mental disorders. These mental disorders were related to violence and the subjectively perceived burden of sex work. Conclusion:, Sex work is a major public health problem. It has many faces, but ill mental health of sex workers is primarily related to different forms of violence. [source]


Survival of mammalian B104 cells following neurite transection at different locations depends on somal Ca2+ concentration

Abstract We report that cell survival after neurite transection in a mammalian neuronal model (cultured B104 cells) critically depends on somal [Ca2+]i, a novel result that reconciles separate long-standing observations that somal survival decreases with more-proximal axonal transections and that increased somal Ca2+ is cytotoxic. Using fluorescence microscopy, we demonstrate that extracellular Ca2+ at the site of plasmalemmal transection is necessary to form a plasmalemmal barrier, and that other divalent ions (Ba2+, Mg2+) do not play a major role. We also show that extracellular Ca2+, rather than injury per se, initiates the formation of a plasmalemmal barrier and that a transient increase in somal [Ca2+]i significantly decreases the percentage of cells that survive neurite transection. Furthermore, we show that the increased somal [Ca2+]i and decreased cell survival following proximal transections are not due to less frequent or slower plasmalemmal sealing or Ca2+ entry through plasmalemmal Na+ and Ca2+ channels. Rather, the increased somal [Ca2+]i and lethality of proximal neurite injuries may be due to the decreased path length/increased diameter for Ca2+ entering the transection site to reach the soma. A ryanodine block of Ca2+ release from internal stores before transection has no effect on cell survival; however, a ryanodine- or thapsigargin-induced buildup of somal [Ca2+]i before transection markedly reduces cell survival, suggesting a minor involvement of Ca2+ -induced release from internal stores. Finally, we show that cell survival following proximal injuries can be enhanced by increasing intracellular Ca2+ buffering capacity with BAPTA to prevent the increase in somal [Ca2+]i. © 2004 Wiley Periodicals, Inc. A power function model for the basic geometry of solution dolines: considerations from the classical karst of south-central Slovenia

Abstract Solution doline geometry is generally too complex to be expressed in terms of a few parameters. A methodology previously designed by the author, based on acquisition of specific field data and subsequent processing by various Fourier techniques, enabled doline geometry to be considered in its entirety. This paper sets out to investigate which simple rotational body (rotated power function) best fits the geometry of 38 dolines studied within the Classical Karst of Slovenia. It transpires that doline radius to depth ratio changes as a function of volume, whereas the mode of change remains stable regardless of the actual dimensions of individual dolines, and is basically the same at different locations. Doline volumes are very close of those of regular cones of the same dimensions, growing relatively deeper as the volume increases. Two poly-segment models implied by the power function model and based on earlier work are discussed. It transpires that the studied dolines may be relict forms superimposed upon pre-existing hidden shafts formed by epikarstic water. Copyright © 2006 John Wiley & Sons, Ltd. [source]


An experimental evaluation of ice cover effects on the dynamic behaviour of a concrete gravity dam

Abstract An extensive forced-vibration testing programme has been carried out on an 84-m concrete gravity dam located in northeastern Québec, Canada. The dam was subjected to a harmonic load on the crest in summer and severe winter conditions with temperatures ranging from ,10°C to ,15°C and a 1.0,1.5m ice cover. Acceleration and hydrodynamic frequency responses were obtained in different locations on the dam and in the reservoir. The main objective of the repeated tests was to investigate the effects of the ice cover on the dynamic behaviour of the dam,reservoir,foundation system, by comparing summer and winter results. Modifications in damping and resonance frequencies were observed, as well as an additional resonance that was attributed to an interaction of the dam with the ice cover. These findings provided a reliable and unique database for the investigations of dam,reservoir,foundation interaction and, in particular, the ice-cover effects for dams located in northern regions. Responses of global plant diversity capacity to changes in carbon dioxide concentration and climate

Abstract We model plant species diversity globally by country to show that future plant diversity capacity has a strong dependence on changing climate and carbon dioxide concentration. CO2 increase, through its impact on net primary production and warming is predicted to increase regional diversity capacity, while warming with constant CO2 leads to decreases in diversity capacity. Increased CO2 concentrations are unlikely to counter projected extinctions of endemic species, shown in earlier studies to be more strongly dependent on changing land use patterns than climate per se. Model predictions were tested against (1) contemporary observations of tree species diversity in different biomes, (2) an independent global map of contemporary species diversity and (3) time sequences of plant naturalisation for different locations. Good agreements between model, observations and naturalisation patterns support the suggestion that future diversity capacity increases are likely to be filled from a ,cosmopolitan weed pool' for which migration appears to be an insignificant barrier. Residency and movement of stream-dwelling Japanese charr, Salvelinus leucomaenis, in a central Japanese mountain stream

Abstract,,,The residency and movement of stream-dwelling adult (2+ and older) Japanese charr, Salvelinus leucomaenis, were studied by mark and recapture experiments in the Jadani Stream, a headwater tributary of the Tedori River, central Japan, from 1986 to 1989. Of the marked fish, 31.3,58.3% were recaptured in the same pools where they had been caught during the study periods of summer (June,August), autumn (August,November) and winter,spring (November to the next June) and no seasonal movement was observed. The mean distances that the fish moved during the study periods ranged from 139.0 to 502.3 m and many movements longer than 1000 m were observed. Between the resident and the moved fish, there was little difference in body length, growth rate or sex. For the fish that had been resident in the same pools and riffles at previous recaptures, most fish were recaptures in the same locations at the next recapture. Conversely, for the fish that moved previously, most fish were recaptured in different locations from previous sites at the next recapture. These results suggest that Japanese charr exhibit relatively high residency throughout the year, but many fish moved longer distance. The results also suggest the presence of static and mobile components in the charr population. [source]


Evaluation of river water genotoxicity using the piscine micronucleus test

Abstract The Berdan River, which empties into the Mediterranean Sea on the east coast of Turkey, receives discharges of industrial and municipal waste. In the present study, the in vivo piscine micronucleus (MN) test was used to evaluate the genotoxicity of water samples collected from different locations along the Berdan River. Nile tilapia (Oreochromis niloticus) were exposed in the laboratory for 2, 4, and 6 days, and micronuclei were evaluated in peripheral blood erythrocytes, gill cells, and caudal fin epithelial cells. A single dose of 5 mg/L cyclophosphamide was used as a positive control. In addition to micronuclei, nuclear abnormalities (NAs), such as binucleated cells and blebbed, notched, and lobed nuclei, were assessed in the erythrocytes, and chemical analyses were carried out to determine the amount of heavy metals in the water samples. MN and NA frequencies were significantly elevated (up to 2- to 3-fold) in fish exposed to river water samples taken downstream of potential discharges, and the elevated responses in gill and fin cells were related to the concentration of heavy metals in the water. MN frequencies (expressed as micronucleated cells/1,000 cells), in both treated and untreated fish, were greatest in gill cells (range: 0.80,3.70), and generally lower in erythrocytes (range: 0.50,2.80), and fin cells (range: 0.45,1.70). The results of this study indicate that the Berdan River is contaminated with genotoxic pollutants and that the genotoxicity is related to the discharge of wastes into the river water. Environ. Mol. Biogeography of wetland rice methanotrophs

Summary We focused on the functional guild of methane oxidizing bacteria (MOB) as model organisms to get deeper insights into microbial biogeography. The pmoA gene was used as a functional and phylogenetic marker for MOB in two approaches: (i) a pmoA database (> 4000 sequences) was evaluated to obtain insights into MOB diversity in Italian rice paddies, and paddy fields worldwide. The results show a wide geographical distribution of pmoA genotypes that seem to be specifically adapted to paddy fields (e.g. Rice Paddy Cluster 1 and Rice Paddy Cluster 2). (ii) On the smaller geographical scale, we designed a factorial experiment including three different locations, two rice varieties and two habitats (soil and roots) within each of three rice fields. Multivariate analysis of terminal restriction fragment analysis profiles revealed different community patterns at the three field sites, located 10,20 km apart. Root samples were characterized by high abundance of type I MOB whereas the rice variety had no effect. With the agronomical practice being nearly identical, historical contingencies might be responsible for the field site differences. Considering a large reservoir of viable yet inactive MOB cells acting as a microbial seed bank, environmental conditions might have selected and activated a different subset at a time thereby shaping the community. Estimation of microbial cover distributions at Mammoth Hot Springs using a multiple clone library resampling method

Summary We propose the use of cover as a quick, low-resolution proxy for the abundance of microbial species, which reduces polymerase chain reaction bias. We showcase this concept in a computation that uses clone library information from travertine-forming hot springs in Yellowstone National Park to provide estimates of relative covers at different locations within the spring system. Samples were used from two media: the water column and the travertine substrate. The cover distribution is found to approximate a power law for samples within the water column. Significant commonality of species with the highest cover is observed in the water column for all locations, but not for species present in the substrate at different locations or between media at the same location. Frequent genetic recombination in natural populations of the marine cyanobacterium Microcoleus chthonoplastes

Summary A culture-independent method for multilocus sequence typing of Microcoleus chthonoplastes was developed based on mechanical separation of individual cyanobacterial filaments from natural microbial mat populations through micromanipulation, subsequent polymerase chain reaction (PCR) amplification and sequence analysis of three genetic loci (kaiC, petB/D, rDNA-ITS). Among 81 individuals sampled from intertidal sand flats of the North Sea and Baltic Sea, we found 8,14 different sequences (alleles) per genetic locus, resulting in 36 distinct genotypes with unique allele profiles. Non-congruent phylogenetic gene trees for the three loci analysed and split decomposition analysis indicated the occurrence of horizontal genetic exchange. The index of association determined for the entire population was 0.096, indicating that recombination occurs frequently enough to cause almost random association (linkage equilibrium) among alleles. Analysing individuals from three different locations in the North Sea and Baltic Sea, we did not find evidence for geographic subdivisions between populations. [source]


Bacterial community structure and function in a metal-working fluid

Summary The diversity of bacterial populations colonizing spatially and temporally separated samples of the same metal-working fluid (MWF) formulation was investigated. Analyses were performed with a view to improve strategies for bioaugmentation of waste MWF in bioreactor systems and prevention of in-use MWF biodeterioration in engineering workshops. Significantly, complementary phenotypic, genotypic and in situ methods revealed that the bacterial communities in operationally exhausted MWFs had low diversity and were similar in species composition from different locations and uses. Of the 179 bacterial isolates studied, only 11 genera and 15 species were identified using fatty acid methyl ester (FAME) analysis, with culture independent analyses by 16S rDNA denaturing gradient gel electrophoresis (DGGE) and fluorescent in situ hybridization being congruent with these FAME data. In order to gain some insight into functional role of detected populations, we correlated the MWF chemical composition and potential pollution load with bacterial abundance and community composition detected within samples. Effects of metal and organophosphate mixtures on Ceriodaphnia dubia survival and reproduction

Abstract The toxicity of mixtures of copper, zinc, and diazinon were determined for Ceriodaphnia dubia using 7-d survival and reproduction tests. Fifteen treatments, including combinations of the chemicals at 0, 25, 50, 75, and 100% of their individual median lethal concentrations, adding up to one toxic unit (TU) were tested. The TU was then used to classify each mixture response as additive, greater than additive, or less than additive. For survival, additive responses occurred in the 75% zinc plus 25% diazinon and the 50% copper plus 25% zinc plus 25% diazinon treatments. For reproduction, additive responses occurred in the 75% copper plus 25% zinc, 75% copper plus 25% diazinon, and 75% zinc plus 25% diazinon treatments. Copper and zinc played a greater role in toxicity than diazinon did. Less-than-additive interactions were found in all remaining mixtures, perhaps because of differences in mode of action between diazinon and metals. Consideration of dose-response curves can help to explain inconsistencies regarding toxic response in treatments with different ratios of the same chemicals. As TU percentages changed, mixture components were taken from different locations on differently shaped dose-response curves. Because most responses were less than additive, however, water-quality criteria based on individual concentrations probably are protective for most metal-organophosphate mixtures. [source]


A Bayesian hierarchical extreme value model for lichenometry

Abstract Currently, there is a tremendous scientific research effort in the area of climate change. In this paper, our motivation is to improve the understanding of historical climatic events such as the Little Ice Age (LIA), a period of relatively cold weather around 1450,1850 AD. Although the LIA is well documented in Europe, its extent and timing are not known in areas of the globe where climatological records were not kept during this period. To study the climate, which predates historical records, proxy climate records must be used. A proxy record for the timing of climatic cooling events are the ages of the moraines left behind by glacial advances. Unfortunately, to determine the ages of these moraines in alpine environments there is little material available but lichens. Hence, lichenometry was developed to determine the ages of glacial landforms by using lichen measurements. To our knowledge, this article provides the first attempt at deriving a comprehensive statistical model for lichenometry. Our model foundation is based on extreme value theory because only the largest lichens are measured in lichenometry studies. This application is novel to extreme value theory because the quantities of interest (the ages of climatic events) are not the measured quantities (lichen diameters), i.e., it is a inverse problem. We model the lichen measurements with the generalized extreme value (GEV) distribution, upon which a Bayesian hierarchical model is built. The hierarchical model enables estimation of the hidden covariate ages of the moraines. The model also allows for pooling of data from different locations and evaluation of spatial differences in lichen growth. Parameter inference is obtained using a straightforward Markov Chain Monte Carlo method. Our procedure is applied to data gathered from the Cordillera Real region in Bolivia. Oceanic influence on the precipitation of the south-east of Venezuela

Abstract The Caroní catchment located in the south-east of Venezuela accounts for 70 per cent of the total hydropower energy of the country. On a year to year basis, it has been shown that low frequency large scale ocean-atmosphere phenomena are highly coupled to the hydroclimatology of the region, El Niño-Southern Oscillation (ENSO) being a major forcing mechanism of climatic and hydrological anomalies. Regional differences in amplitude and timing are due to complex orographic interactions, land surface-atmosphere feedback mechanisms and the evolution of dominant synoptic meteorological conditions. A detailed analysis of the relationship between rainfall and several large scale ocean-atmospheric variables was carried out to determine the potential use of large scale climatic information as predictors of the rainfall anomalies over the region. The problem was tackled in two ways: (a) first a seasonal dynamic rainfall model was fitted to monthly rainfall for different locations. In this case rainfall is assumed as a normal variate w which has been transformed to account for its departure from normality and truncated to account for the positive probability mass of zero values, which corresponds to negative values of the normal variable. The time series of the model parameters and the macroclimatic variables are inspected for their potential relationship with local rainfall via the stochastic model. (b) Second, dynamic linear regression models between the macroclimatic variables as predictors and the rainfall anomalies as predictant were fitted to evaluate and quantify the significance of these dependencies. Consistent patterns are observed with the Tropical Atlantic and Pacific ocean temperature anomalies, in which a significant negative relationship has been present since 1976, indicating an overall decrease (increase) in rainfall when the Pacific and the Tropical Atlantic are warmer (colder) than normal. The influence of strenuous exercise on collagen characteristics of articular cartilage in Thoroughbreds age 2 years

Summary In order to assess the influence of strenuous exercise on collagen characteristics of articularcartilage, the response of the collagen network was studied in seven 2-year-old Thoroughbreds subjected to strenuous exercise compared to 7 nontrained individuals. After 13 weeks, the animals were subjected to euthanasia, fetlock joints of the forelimbs were scored macroscopically after Indian Ink staining, and articular cartilage from different locations of the articular surface of the proximal first phalanx was sampled and analysed for water content, collagen content, hydroxylysine content and amount of hydroxylysylpyridinoline (HP) crosslinks. Gross lesions were significantly more severe in the exercised than in the nonexercised group. In the control animals, the characteristic site-specific differences in collagen parameters were found as described earlier, but in the strenuously exercised animals this physiological biochemical heterogeneity had disappeared. In the exercised animals, an increase in water content and a sharp decrease in HP crosslinking was found that was correlated with the presence of wear lines. It is concluded that the strenuous exercise provoked significant alterations in the characteristics of the collagen network of the articular cartilage of the fetlock joint which were suggestive of microdamage and loosening of the collagen network. The collagen component of cartilage, in contrast to the proteoglycan component, is known to have a very limited capacity for repairand remodelling due to an extremely low turnover rate. Therefore, alterations within the articular collagen network might be expected to play an important role in the pathophysiology of degenerative joint disorders. [source]


Context Dependent Territory Defense: The Importance of Habitat Structure in Anolis sagrei

Territoriality is a potentially costly endeavor, and several mechanisms for mitigating the costs of territoriality have been investigated in the wild. For example, territory owners can reduce the costs of defending territory boundaries by prioritizing defense of the most valuable areas within territories, investing less energy in low quality areas. We staged pairwise encounters between adult male lizards on natural territories in the wild, to test whether male brown anoles, Anolis sagrei, would differentially defend certain regions of their territories over others. Based on our observations that male A. sagrei spend most of their time on elevated perches on tree trunks or branches compared with sites on the ground, we predicted that territory residents would respond more aggressively to territory invasions that took place on elevated perches than to invasions on the ground. We measured significant differences in the behavior of residents following invasion on the ground vs. on the elevated perches, and results partially supported our hypothesis. Males performed more displays and approached intruders more often when territory invasion took place on the ground, but were quicker to attack intruders that entered territories on elevated perches. Our hypothesis was only partially supported, potentially indicating that elevated perches are preferred as outposts to monitor valuable sites on the ground. Our study provides evidence that territory defense varies not just among individuals, but also within individuals at different locations in a territory. [source]


Chemical evaluation of some paprika (Capsicum annuum L.) seed oils

Abstract The oil contents of seeds from paprika (Capsicum annuum L.) collected from different locations in Turkey and Italy varied in a relatively wide range from 8.5,g/100,g to 32.6,g/100,g. The fatty acid, tocopherol and sterol contents of the oils from different paprika seeds were investigated. The main fatty acids in paprika seed oils were linoleic acid (69.5,74.7,g/100,g), oleic acid (8.9,12.5,g/100,g) and palmitic acid (10.7,14.2,g/100,g). The oils contained an appreciable amount of ,-tocopherol (306.6,602.6,mg/kg), followed by ,-tocopherol (7.3,148.7,mg/kg). The major sterols were ,-sitosterol (1571.4,4061.7,mg/kg), campesterol (490.8,1182.7,mg/kg), and ,5 -avenasterol (374.5,899.6,mg/kg). The total concentration of sterols ranged from 3134.0,mg/kg to 7233.7,mg/kg. Remarkable amounts of cholesterol were found in the different samples (164.6,491.0,mg/kg). The present study showed that paprika seeds are a potential source of valuable oil that could be used for edible and industrial applications. [source]


Differential routing of coexisting neuropeptides in vasopressin neurons

Abstract The functional implications of intraneuronal coexistence of different neuropeptides depend on their respective targeting to release sites. In the rat hypothalamic magnocellular neurons, we investigated a possible differential routing of the coexpressed galanin and vasopressin. The respective location of proteins and messengers was assessed with double immunogold and in situ hybridization combining confocal and electron microscope analysis. The various populations of labelled granules were quantitatively compared in three subcellular compartments: perikarya, local processes and posthypophyseal nerve endings. Three subpopulations of granules were detected in all three compartments, but their respective amount showed significant differences. Galanin alone was immunolocalized in some secretory granules, vasopressin alone in others, and both peptides in a third subpopulation of granules. The major part of the granules containing vasopressin, either alone or in association with galanin, is found in neurohypophyseal nerve endings. In contrast, galanin single-labelled granules represent the most abundant population in dendritic processes, while double-labelled granules are more numerous in perikarya. This indicates a preferential distribution of the two peptides in the different compartments of magnocellular neurons. Furthermore, galanin and vasopressin messenger RNAs were detected at different domains of the endoplasmic reticulum, suggesting that translation might also occur at different locations, thus leading to partial segregation of galanin and vasopressin cargoes between two populations of secretory granules. The present study provides, for the first time in mammals, evidence suggesting that galanin and vasopressin are only partly copackaged and undergo a preferential targeting toward dendrites or neurohypophysis, suggesting different functions, autocrine/paracrine and endocrine, respectively. Material stiffness, branching pattern and soil matric potential affect the pullout resistance of model root systems

Summary Understanding of the detailed mechanisms of how roots anchor in and reinforce soil is complicated by the variability and complexity of both materials. This study controlled material stiffness and architecture of root analogues, by using rubber and wood, and also employed real willow root segments, to investigate the effect on pullout resistance in wet and air-dry sand. The architecture of model roots included either no laterals (tap-root) or a single pair at two different locations (herringbone and dichotomous). During pullout tests, data on load and displacement were recorded. These studies were combined with Particle Image Velocimetry (PIV) image analysis of the model root-soil system at a transparent interface during pullout to increase understanding of mechanical interactions along the root. Model rubber roots with small stiffness had increasing pullout resistance as the branching and the depth of the lateral roots increased. Similarly, with the stiff wooden root models, the models with lateral roots embedded deeper showed greatest resistance. PIV showed that rubber model roots mobilized their interface shear strength progressively whilst rigid roots mobilized it equally and more rapidly over the whole root length. Soil water suction increased the pullout resistance of the roots by increasing the effective stress and soil strength. Separate pullout tests conducted on willow root samples embedded in sand showed similar behaviour to the rigid model roots. These tests also demonstrated the effect of the root curvature and rough interface on the maximum pullout resistance. [source]


Poorly crystalline mineral phases protect organic matter in acid subsoil horizons

Summary Soil minerals are known to influence the biological stability of soil organic matter (SOM). Our study aimed to relate properties of the mineral matrix to its ability to protect organic C against decomposition in acid soils. We used the amount of hydroxyl ions released after exposure to NaF solution to establish a reactivity gradient spanning 12 subsoil horizons collected from 10 different locations. The subsoil horizons represent six soil orders and diverse geological parent materials. Phyllosilicates were characterized by X-ray diffraction and pedogenic oxides by selective dissolution procedures. The organic carbon (C) remaining after chemical removal of an oxidizable fraction of SOM with NaOCl solution was taken to represent a stable organic carbon pool. Stable organic carbon was confirmed as older than bulk organic carbon by a smaller radiocarbon (14C) content after oxidation in all 12 soils. The amount of stable organic C did not depend on clay content or the content of dithionite,citrate-extractable Fe. The combination of oxalate-extractable Fe and Al explained the greatest amount of variation in stable organic C (R2 = 0.78). Our results suggest that in acid soils, organic matter is preferentially protected by interaction with poorly crystalline minerals represented by the oxalate-soluble Fe and Al fraction. This evidence suggests that ligand exchange between mineral surface hydroxyl groups and negatively charged organic functional groups is a quantitatively important mechanism in the stabilization of SOM in acid soils. The results imply a finite stabilization capacity of soil minerals for organic matter, limited by the area density of reactive surface sites. [source]


Multi-agent application of substation protection coordination with distributed generators

Abstract This paper presents new explorations into the use of agent technology applied to substation protection coordination of power systems. The impact of distributed generators (DG) on protection coordination in substation with different locations is discussed first. Then a coordination multi-agent system is proposed with the functions of the agents described. In the proposed system, communication will play an important role to provide more information for the relay coordination besides the relay settings. Relay coordination strategy is also discussed and communication simulation has been carried out on the Java agent development framework (JADE) platform. Androgen action on human skin , from basic research to clinical significance

Abstract:, Androgens affect several functions of the human skin, such as sebaceous gland growth and differentiation, hair growth, epidermal barrier homeostasis and wound healing. Their effects are mediated by binding to nuclear androgen receptors. Androgen activation and deactivation are mainly intracellular events. They differ from cell type to cell type and between cells at different locations. The major circulating androgens, dehydroepiandrosterone sulfate and androstenedione, are predominantly produced in the adrenal glands, and testosterone and 5,-dihydrotestosterone are mainly synthesized in the gonads. Testosterone in women and 5,-dihydrotestosterone in both genders are also synthesized in the skin. Skin cells express all androgen metabolizing enzymes required for the independent cutaneous synthesis of androgens and the development of hyperandrogenism-associated conditions and diseases, such as seborrhea, acne, hirsutism and androgenetic alopecia. The major thrust of drug design for the treatment of androgen-associated disorders has been directed against several levels of androgen function and metabolism. Partial effectiveness has only been achieved either by androgen depletion, inhibition of androgen metabolism or blockade of the androgen receptor. [source]


Carboxydipeptidase activities of recombinant cysteine peptidases

CPB of Leishmania mexicana, Cruzain of Trypanosoma cruzi
The recombinant cysteine peptidases, cruzain from Trypanosoma cruzi and CPB2.8,CTE from Leishmania mexicana, are cathepsin L-like and characteristically endopeptidases. In this study, we characterized the carboxydipeptidase activities of these enzymes and compared them with those of human recombinant cathepsin B and cathepsin L. The analysis used the internally quenched fluorescent peptide Abz-FRFK*-OH and some of its analogues, where Abz is ortho -aminobenzoic acid and K* is (2,4-dinitrophenyl)-,-NH2 -lysine. These peptides were demonstrated to be very sensitive substrates, due to the strong quenching effect of K* on the fluorescence of the Abz group. The carboxydipeptidase activity of cruzain was shown to be very similar to that of cathepsin B, while that of CPB2.8,CTE is closer to the carboxydipeptidase activity of cathepsin L. The S2 subsite architecture of cruzain and the nature of the amino acid at the P2 position of the substrates determine its carboxydipeptidase activity and gives further and direct support to the notion that the carboxydipeptidase activity of the papain family cysteine peptidases rely on the S2,P2 interaction [Nägler D. K., Tam, W., Storer, A.C., Krupa, J.C., Mort, J.S. & Menard, R. (1999) Biochemistry38, 4868,4874]. Cruzain and CPB2.8,CTE presented a broad pH-range for both the endo- and exo-peptidase activities, although the later is approximately one order of magnitude lower. This feature, that is not common in related mammalian cysteine peptidases, is consistent with the enzymes being exposed to different environmental conditions and having different locations during parasite development. Bilayer localization of membrane-active peptides studied in biomimetic vesicles by visible and fluorescence spectroscopies

Depth of bilayer penetration and effects on lipid mobility conferred by the membrane-active peptides magainin, melittin, and a hydrophobic helical sequence KKA(LA)7KK (denoted KAL), were investigated by colorimetric and time-resolved fluorescence techniques in biomimetic phospholipid/poly(diacetylene) vesicles. The experiments demonstrated that the extent of bilayer permeation and peptide localization within the membrane was dependent upon the bilayer composition, and that distinct dynamic modifications were induced by each peptide within the head-group environment of the phospholipids. Solvent relaxation, fluorescence correlation spectroscopy and fluorescence quenching analyses, employing probes at different locations within the bilayer, showed that magainin and melittin inserted close to the glycerol residues in bilayers incorporating negatively charged phospholipids, but predominant association at the lipid,water interface occurred in bilayers containing zwitterionic phospholipids. The fluorescence and colorimetric analyses also exposed the different permeation properties and distinct dynamic influence of the peptides: magainin exhibited the most pronounced interfacial attachment onto the vesicles, melittin penetrated more into the bilayers, while the KAL peptide inserted deepest into the hydrophobic core of the lipid assemblies. The solvent relaxation results suggest that decreasing the lipid fluidity might be an important initial factor contributing to the membrane activity of antimicrobial peptides. [source]


Spatial and temporal variability of the bacterial community in different chemotypes of the New Zealand marine sponge Mycale hentscheli

Abstract Molecular fingerprinting of 16S rRNA genes using terminal restriction fragment length polymorphism (T-RFLP) and denaturing gradient gel electrophoresis (DGGE) was used to characterize the temporal and spatial variability among sponge-associated bacteria from Mycale hentscheli having distinct bioactive chemotypes. Cluster analysis of T-RFLP and DGGE profiles from M. hentscheli chemotypes largely grouped sponge microbial diversity to their distinct chemotype pattern. Repeat sampling of individual M. hentscheli at one location over a 21-month period showed that the T-RFLP profiles from individual sponges had similarity indices ranging from 60% to 82% and calculated DGGE similarities between 23% and 95%. However, a portion (>35% from DGGE and >19% from T-RFLP) of the microbial community from M. hentscheli appeared to be spatially conserved through all M. hentscheli populations. Sequence analysis of DGGE band fragments showed a similarity among the bands originating from different individuals, different times, and different locations. The sponge-associated relationship of these bands was confirmed, with sequences having similarity to sponge-associated bacteria reported from global locations. This study highlights the spatial and temporal complexity in the distribution of bacterial communities associated with different chemotypes of the marine sponge M. hentscheli. [source]
