    Relation between Developmental Stage, Sensory Properties, and Volatile Content of Organically and Conventionally Grown Pac Choi (Brassica rapa,var.

    ABSTRACT:, This study was conducted to identify and quantify the sensory characteristics and chemical profile of organically and conventionally grown pac choi (Brassica rapa,var. Mei Qing Choi), also called bok choy, at 3 stages of growth (2.5, 4.5, and 6.5 wk). Sensory and instrumental data were correlated using partial least squares regression. Pac choi was grown in late spring. Descriptive sensory analysis was conducted by a highly trained panel and compounds were identified and quantified using a gas chromatograph/mass spectrometer. The findings of the study indicate that the differences in sensory characteristics and chemical profiles among stages of growth are more substantial than the differences between organic and conventional production. Green-unripe, musty/earthy, lettuce, and sweet flavors are representative in pac choi at early stages of growth. When older, pac choi has higher intensities of green-grassy/leafy, bitter, cabbage, and sulfur flavors that are associated with the increase of (Z)-3-hexen-1-ol, octyl acetate, 1-nonanol, 2-decanone, 1-penten-3-ol, linalool, camphor, menthol, isobornyl acetate, geranylacetone, and cedrol compounds. Conventional pac choi was higher than organic pac choi in green overall, bitter, and soapy flavors only at 2.5 wk of age. This may be associated with the presence of (Z)-3-hexenal, 2-hexyn-1-ol, and (E)-2-hexenal compounds. Practical Application: The increased popularity of organic production has amplified the need for research that will help in understanding how this production system affects the final quality of food products. This study suggests that the stage of development has a much larger impact on sensory quality than organic or conventional growing of pac choi. Findings from this study promote consumer choice by showing that comparable sensory quality can be obtained using either production system, making the ultimate choice not only based on sensory quality but consumer choice related to environmental beliefs or economics.     Revisiting the Developmental Stage and Age-at-Death of the "Mrs.

    Ples" (Sts 5), Sts 14 Specimens from Sterkfontein (South Africa): Do They Belong to the Same Individual?
    Abstract During the 1947 fieldwork season, Member 4 (2-3 My) of the South African Sterkfontein site yielded two important Australopithecus africanus fossils: a cranium popularly nicknamed "Mrs. Ples" (Sts 5), and a partial skeleton (Sts 14). Previous reports have proposed that Sts 5 was a nonfully grown adolescent individual (Thackeray et al., S Afr J Sci 2002a;98:21,22), and that Sts 14 was a sub-adult specimen (according to various signs of immaturity in the skeleton) (Berge and Gommery, C R Acad Sci Paris, Sciences de la terre et des planètes 1999;329:227,232; Häusler and Berger, J Hum Evol, 2001;40:411,417; Thackeray et al., S Afr J Sci, 2002b;98:211,212). It was subsequently proposed that these fossils actually belonged to the same individual (Thackeray et al., S Afr J Sci, 2002b;98:211,212), a proposition supported by their spatial positions within the site. The present work attempts to revise these different assertions. The results obtained: (i) show that the Sts 5 fossil represents a fully grown adult cranium; (ii) provide new evidence of immaturity in the Sts 14 skeletal elements (sustaining the proposed young adult age of this specimen), and (iii) suggest that although the revised ages-at-death for these fossils are partially compatible, there is no evidence to support the idea that they represent a single individual. Finally, the encephalization quotient associated with a hypothetical union of Sts 5 and Sts 14 (calculated using data from both specimens) lies between the upper and lower limits of the currently estimated range for this species and H. habilis, respectively.     Developmental Stages of Age and Moral Reasoning as Predictors of Juvenile Delinquents' Behavioral Intention to Steal Clothing

    This study explored juvenile delinquents' moral reasoning and behavioral intention to steal clothing using age group and the aligned theories of planned behavior and cognitive developmental moralization. Participants (n = 100) were preteen and teen first-time theft/shoplifting offenders participating in a diversion program aimed at preventing future offenses. Factor analysis revealed three strongly correlated dimensions of moral reasoning: risk and need (preconventional ethics), and peers (conventional ethics). Multiple regressions predicted positive moral reasoning that justified stealing clothing for risk, need, and peers by preteens but not teens. Risk and need, but not peers or age group, predicted behavioral intention to steal clothing. Adevelopmental shift existed from preteens to teens in their moral reasoning to steal clothing. Juvenile delinquents that identified with preconventional ethics exhibited behavioral intention to steal clothing. [source]


    Estimating causal effects from observational data with a model for multiple bias

    Abstract Conventional analyses of observational data may be biased due to confounding, sampling and measurement, and may yield interval estimates that are much too narrow because they do not take into account uncertainty about unknown bias parameters, such as misclassification probabilities. We used a simple, multiple bias adjustment method to estimate the causal effect of social anxiety disorder (SAD) on subsequent depression. A Monte Carlo sensitivity analysis was applied to data from the Early Developmental Stages of Psychiatry (EDSP) study, and bias due to confounding, sampling and measurement was modelled. With conventional logistic regression analysis, the risk for depression was elevated in the presence of SAD only in the older cohort (age 17,24 years at baseline assessment); odds ratio (OR) = 3.06, 95% confidence interval (CI) 1.64,5.70, adjusted for sex and age. The bias-adjusted estimate was 2.01 with interval limits of 0.61 and 9.71. Thus, given the data and the bias model used, there was considerably more uncertainty about the real effect, but the probability that SAD increases the risk for subsequent depression (OR > 1) was 88.6% anyway. Multiple bias modelling, if properly used, reveals the necessity for a better understanding of bias, suggesting a need to conduct larger and more adequate validation studies on instruments that are used to diagnose mental disorders.     Evaluation of Drought-Related Traits and Screening Methods at Different Developmental Stages in Spring Barley

    Abstract Despite intensive research and breeding efforts, the physiological and quantitative genetic bases of drought tolerance are still poorly understood. The comparison of results obtained from different sources is also complex, because different testing methods may lead to controversial conclusions. This report discusses various drought stress experiments (hydroponics and in soil) in which the plant tolerance was studied at different developmental stages. Tests were performed in the germination, seedling and adult plant stages on the parental lines of five well-known barley-mapping populations. The results suggest that drought tolerance is a stage-specific trait and changes during the life cycle. The effect of drought stress depended not only on the duration and intensity of water deficiency, but also on the developmental phase in which it began. To induce the same type of stress and to obtain comparable tolerance information from the replications, it is recommended that drought stress should be induced at the same growth stage. Correlations between the traits, commonly associated with improved drought resistance (high relative water content under stress, proline accumulation, osmoregulation) with stress tolerance indexes, are also presented, while the advantages and disadvantages of the most frequently used screening methods are discussed.     Fungicide Effectiveness during the Various Developmental Stages of Peronophythora litchii In Vitro

    Abstract Litchi downy blight caused by Peronophythora litchii is one of the most destructive diseases suffered by litchi in China. This study has evaluated the activities of the fungicides dimethomorph (DMM), azoxystrobin (AZB), famoxadone (FMD), metalaxyl (MTL), cymoxanil (CYX) and mancozeb (MCB) on the mycelial growth, sporulation, zoospores release, and germination of sporangia as well as of cystospores of P. litchii. Dimethomorph and MTL inhibited mycelial growth more effectively than the other fungicides tested. Mycelial growth was affected less by MCB, and only weakly by AZB, FMD and CYX. Sporangia production was more strongly reduced by DMM and MTL than by AZB, FMD, CYX or MCB. Zoospore release from the sporangia was most sensitive to AZB and FMD, less sensitive to MCB and insensitive to DMM, MTL and CYX. Direct germination of sporangia of P. litchii was the most strongly inhibited by AZB and FMD, followed by DMM and MCB, whilst the sensitivity of this life stage to MTL and CYX was relatively low. Germination of encysted zoospores of P. litchii was the most sensitive to AZB and FMD, followed by DMM. Mancozeb was moderately active, while MTL and CYX did not inhibit this development stage. This is the first report on the in vitro response of the litchi pathogen, P. litchii, to fungicides. These findings can be valuable tools in setting up efficient disease management programmes to control litchi downy blight. [source]


    Gonadogenesis in early developmental stages of Acipenser naccarii and influence of estrogen immersion on feminization

    Summary Gonad development processes and the effects of a single 8-hour immersion treatment with 17, -estradiol (E2, 400 ,g L,1) on sex differentation in the Adriatic sturgeon, Acipenser naccarii, were investigated. After migration of germ cells, gonadal ridges appeared in 16- to 18-day old larvae and undifferentiated gonads in 55- to 60-day old larvae. Putative ovaries with notches in the germinal epithelium and presumed testes with smooth germinal epithelium appeared in 180,185-day old juveniles. Ovaries with proliferating oogonia and early meiotic oocytes clusters were observed in 292-day old juveniles. Testes did not exhibit germ cell mitosis until 430 days of age. Developmental stages in E2 -treated animals closely followed those of controls up to 430 days. The treatment significantly increased the percentage of ovaries when administered to embryos about 1.5 day before hatching, while did not significantly altered the normal 1/1 sex ratio when administered to 1.5-day old pre-larvae and 10-day old larvae. It is likely that in A. naccarii exogenous E2 administration may act through a feedback mechanism of self-supporting steroid production and that steroids are the physiological inducers of sex differentiation, as in most teleosts. The E2 -immersion treatment, easier than time-consuming administration through food, could be a good approach to control sex differentiation and caviar production. [source]


    Developmental stages, larval and post-larval growth of angelwing clam Pholas orientalis

    Abstract Angelwing clam were induced to spawn by thermal stimulation. Mature eggs measured 50 ,m in diameter. Cell division occurred within 36 min after fertilization. Mobile trochophore larvae were seen after 12 h and larvae developed within 18 h. Reared on a diet of Isochrysis galbana the larvae reached the umbo stage in 6,7 days. On day 10 the foot could be seen and settlement occurred if a suitable substrate was present. The larvae completed metamorphosis into juveniles within 20 days after settling. [source]


    Characterization of the developmental stages of sucking in preterm infants during bottle feeding

    It is acknowledged that the difficulty many preterm infants have in feeding orally results from their immature sucking skills. However, little is known regarding the development of sucking in these infants. The aim of this study was to demonstrate that the bottle-feeding performance of preterm infants is positively correlated with the developmental stage of their sucking. Infants' oral-motor skills were followed longitudinally using a special nipple/bottle system which monitored the suction and expression/compression component of sucking. The maturational process was rated into five primary stages based on the presence/absence of suction and the rhythmicity of the two components of sucking, suction and expression/compression. This five-point scale was used to characterize the developmental stage of sucking of each infant. Outcomes of feeding performance consisted of overall transfer (percent total volume transfered/volume to be taken) and rate of transfer (ml/min). Assessments were conducted when infants were taking 1-2, 3-5 and 6-8 oral feedings per day. Significant positive correlations were observed between the five stages of sucking and postmenstrual age, the defined feeding outcomes, and the number of daily oral feedings. Overall transfer and rate of transfer were enhanced when infants reached the more mature stages of sucking. We have demonstrated that oral feeding performance improves as infants' sucking skills mature. In addition, we propose that the present five-point sucking scale may be used to assess the developmental stages of sucking of preterm infants. Such knowledge would facilitate the management of oral feeding in these infants. [source]


    Programmed cell death of the ovarian nurse cells during oogenesis of the silkmoth Bombyx mori

    In the present study, we describe the features of programmed cell death of the ovarian nurse cells occurring during vitellogenesis of the silkmoth Bombyx mori. At developmental stage 5, the nurse cells occupy one-half of the follicular volume and obtain a rather spherical shape, while the nurse cell nuclei appear large and elongated, forming impressive projections. At the following stage, stage 6, the nurse cells decrease in size and their shape becomes elliptic. The nuclei remain elongated, being also characterized by large lobes. The lobes of the ramified nurse cell nuclei seem to retain the nucleus in the center of the cell during the dumping of the nurse cell cytoplasm into the growing oocyte. At stage 7, membrane enclosed vacuoles can be easily detected into the nurse cells cytoplasm. Ultrastructural analysis and fluorescent microscopy using mono-dansyl-cadaverine staining of these vacuoles also reveal that they represent autolysosomes. Caspase activity is detected during stage 7, as it is demonstrated by using the Red-VAD-FMK staining reagent. At developmental stages 8 and 9, the nurse cells exhibit chromatin condensation, DNA fragmentation and caspase activity. Finally, during the following stage 10, the nuclear remnants are assembled into apoptotic vesicles, which, after being phagocytosed, are observed in the cytoplasm of adjacent follicle cells. We propose that apoptosis and autophagy operate synergistically during vitellogenesis of B. mori, in order to achieve an efficient and rapid clearance of the degenerated nurse cell cluster.     Development of murine hepatic sinusoidal endothelial cells characterized by the expression of hyaluronan receptors

    Abstract Endothelial cells (ECs) display distinct structural and functional characteristics depending on the tissue and developmental stage; however, the development of tissue-specific ECs remains poorly understood. Here, we describe the development of hepatic sinusoids in mice based on the expression of hyaluronan receptors Stab2 and Lyve-1. Flk-1+ cells in and around the liver bud begin to express Stab2 at embryonic day (E) 9.5, before the formation of vascular lumen. Hepatic sinusoidal endothelial cells (HSECs) begin to express Lyve-1 at E10.5, and both markers continue to be expressed in HSECs thereafter. Although HSECs and lymphatic ECs (LECs) are known to share functional and phenotypic characteristics, we clearly show that HSECs can be distinguished from LECs by the expression of molecular markers and higher endocytotic activity. Our results provide new insight into the development of tissue-specific ECs and phenotypic criteria to distinguish HSECs from other types of ECs, including LECs.     Lineage-independent mosaic expression and regulation of the Ciona multidom gene in the ancestral notochord

    Abstract The transcription factor Ciona Brachyury (Ci-Bra) plays an essential role in notochord development in the ascidian Ciona intestinalis. We characterized a putative Ci-Bra target gene, which we named Ci - multidom, and analyzed in detail its expression pattern in normal embryos and in embryos where Ci - Bra was misexpressed. Ci - multidom encodes a novel protein, which contains eight CCP domains and a partial VWFA domain. We show that an EGFP-multidom fusion protein localizes preferentially to the endoplasmic reticulum (ER), and is excluded from the nucleus. In situ hybridization experiments demonstrate that Ci - multidom is expressed in the notochord and in the anterior neural boundary (ANB). We found that the expression in the ANB is fully recapitulated by an enhancer element located upstream of Ci - multidom. By means of misexpression experiments, we provide evidence that Ci-Bra controls transcription of Ci - multidom in the notochord; however, while Ci-Bra is homogeneously expressed throughout this structure, Ci - multidom is transcribed at detectable levels only in a random subset of notochord cells. The number of notochord cells expressing Ci - multidom varies among different embryos and is independent of developmental stage, lineage, and position along the anterior,posterior axis. These results suggest that despite its morphological simplicity and invariant cell-lineage, the ancestral notochord is a mosaic of cells in which the gene cascade downstream of Brachyury is differentially modulated.     Graphic and movie illustrations of human prenatal development and their application to embryological education based on the human embryo specimens in the Kyoto collection

    Abstract Morphogenesis in the developing embryo takes place in three dimensions, and in addition, the dimension of time is another important factor in development. Therefore, the presentation of sequential morphological changes occurring in the embryo (4D visualization) is essential for understanding the complex morphogenetic events and the underlying mechanisms. Until recently, 3D visualization of embryonic structures was possible only by reconstruction from serial histological sections, which was tedious and time-consuming. During the past two decades, 3D imaging techniques have made significant advances thanks to the progress in imaging and computer technologies, computer graphics, and other related techniques. Such novel tools have enabled precise visualization of the 3D topology of embryonic structures and to demonstrate spatiotemporal 4D sequences of organogenesis. Here, we describe a project in which staged human embryos are imaged by the magnetic resonance (MR) microscope, and 3D images of embryos and their organs at each developmental stage were reconstructed based on the MR data, with the aid of computer graphics techniques. On the basis of the 3D models of staged human embryos, we constructed a data set of 3D images of human embryos and made movies to illustrate the sequential process of human morphogenesis. Furthermore, a computer-based self-learning program of human embryology is being developed for educational purposes, using the photographs, histological sections, MR images, and 3D models of staged human embryos.     Circadian clock and cell cycle gene expression in mouse mammary epithelial cells and in the developing mouse mammary gland

    Abstract Mouse mammary epithelial cells (HC-11) and mammary tissues were analyzed for developmental changes in circadian clock, cellular proliferation, and differentiation marker genes. Expression of the clock genes Per1 and Bmal1 were elevated in differentiated HC-11 cells, whereas Per2 mRNA levels were higher in undifferentiated cells. This differentiation-dependent profile of clock gene expression was consistent with that observed in mouse mammary glands, as Per1 and Bmal1 mRNA levels were elevated in late pregnant and lactating mammary tissues, whereas Per2 expression was higher in proliferating virgin and early pregnant glands. In both HC-11 cells and mammary glands, elevated Per2 expression was positively correlated with c-Myc and Cyclin D1 mRNA levels, whereas Per1 and Bmal1 expression changed in conjunction with ,- casein mRNA levels. Interestingly, developmental stage had differential effects on rhythms of clock gene expression in the mammary gland.     Conditional expression of a myocardium-specific transgene in zebrafish transgenic lines

    Abstract To develop the first heart-specific tetracycline (Tet)-On system in zebrafish, we constructed plasmids in which the cardiac myosin light chain 2 promoter of zebrafish was used to drive the reverse Tet-controlled transactivator (rtTA) and the green fluorescent protein (GFP) reporter gene was preceded by an rtTA-responsive element. In the zebrafish fibroblast cell-line, rtTA-M2, one of rtTA's derivatives, demonstrated the highest increase in luciferase activity upon doxycycline (Dox) induction. We then generated two germ lines of transgenic zebrafish: line T03 was derived from microinjection of a plasmid containing rtTA-M2 and a plasmid containing a responsive reporter gene, whereas line T21 was derived from microinjection of a single dual plasmid. Results showed that line T21 was superior to line T03 in terms of greater GFP intensity after induction and with of minimal leakiness before induction. The photographic images of induced GFP in the heart of F2 larvae showed that the fluorescent level of GFP was dose-responsive. The level of GFP expressed in the F3 3 days postfertilization larvae that were treated with Dox for 1 hr decreased gradually after the withdrawal of the inducer; and the fluorescent signal disappeared after 5 days. The GFP induction and reduction were also tightly controlled by Dox in the F3 adult fish from line T21. This Tet-On system developed in zebrafish shows much promise for the study of the gene function in a specific tissue at the later developmental stage.     RhoA/ROCK and Cdc42 regulate cell-cell contact and N-cadherin protein level during neurodetermination of P19 embryonal stem cells

    Abstract RhoGTPases regulate actin-based signaling cascades and cellular contacts. In neurogenesis, their action modulates cell migration, neuritogenesis, and synaptogenesis. Murine P19 embryonal stem cells differentiate to neurons upon aggregation in the presence of retinoic acid, and we previously showed that RhoA and Cdc42 RhoGTPases are sequentially up-regulated during neuroinduction, suggesting a role at this very early developmental stage. In this work, incubation of differentiating P19 cells with C3 toxin resulted in decreased aggregate cohesion and cadherin protein level. In contrast, C3 effects were not observed in cells overexpressing recombinant dominant active RhoA. On the other hand, C3 did not affect cadherin in uninduced cells and their postmitotic neuronal derivatives, respectively expressing E- and N-cadherin. RhoA is thus influential on cell aggregation and cadherin expression during a sensitive time window that corresponds to the switch of E- to N-cadherin. Cell treatment with Y27632 inhibitor of Rho-associated-kinase ROCK, or advanced overexpression of Cdc42 by gene transfer of a constitutively active form of the protein reproduced C3 effects. RhoA-antisense RNA also reduced cadherin level and the size of cell aggregates, and increased the generation of fibroblast-like cells relative to neurons following neuroinduction. Colchicin, a microtubule disrupter, but not cytochalasin B actin poison, importantly decreased cadherin in neurodifferentiating cells. Overall, our results indicate that the RhoA/ROCK pathway regulates cadherin protein level and cell-cell interactions during neurodetermination, with an impact on the efficiency of the process. The effect on cadherin seems to involve microtubules. The importance of correct timing of RhoA and Cdc42 functional expression in neurogenesis is also raised. © 2004 Wiley Periodicals, Inc.     Developmental shift in bidirectional functions of taurine-sensitive chloride channels during cortical circuit formation in postnatal mouse brain

    Abstract Taurine (2-aminoethanesulfonic acid) is the most abundant free amino acid in the developing mammalian cerebral cortex, however, few studies have reported its neurobiological functions during development. In this study, by means of whole-cell patch-clamp recordings, we examined the effects of taurine on chloride channel receptors in neocortical neurons from early to late postnatal stages, which cover a critical period in cortical circuit formation. We show here that taurine activates chloride channels in cortical neurons throughout the postnatal stages examined (from postnatal day 2 to day 36). The physiological effects of taurine changed from excitatory to inhibitory due to variations in the intracellular Cl, concentration during development. An antagonist blocking analysis also demonstrated a developmental shift in the receptor target of taurine, from glycine receptors to GABAA receptors. Taken together, these results may reflect genetically programmed, bidirectional functions of taurine. At the early developmental stage, taurine acting on glycine receptors would serve to promote cortical circuit formation.     Olfactory preference for own mother and litter in 1-day-old rabbits and its impairment by thermotaxis

    Abstract We investigated the ability of rabbit pups to display preferences towards various elements of their postnatal environment during the stage of confinement in the nest. Subjects were submitted to a two-choice test during the first week after birth to assess if they could detect and discriminate between does, litters of pups, or nesting materials of the same developmental stage. On D1 and D7, pups were attracted to any lactating doe, litter, or nest when compared to an empty compartment. When two stimuli were opposed, pups preferred their own nest to an alien one on D1 and D7 but not their mother nor their siblings when opposed to alien does or pups. However, additional tests indicated that this lack of preference for kin conspecifics resulted from a predominant attraction to thermal cues over individual odors. Indeed, pups were strongly attracted to a warm compartment (37°C) than to a cooler one (20°C) and once thermal cues were controlled for in the testing situation, the pups were specifically attracted to odors of their own mother's hair and of their siblings. No preference was observed towards the mother's uterine secretions. In conclusion, pups can recognize olfactory cues emanating from their mother and their siblings the day after birth. The preference for nesting materials would reflect in major part the combined attraction to maternal and sibling odors present in the nest. © 2008 Wiley Periodicals, Inc.     Light and scanning microscopic studies of integument differentiation in the grass snake Natrix natrix L. (Lepidosauria, Serpentes) during embryogenesis

    Abstract We analysed the differentiation of body cover in the grass snake (Natrix natrix L.) over the full length of the embryo's body at each developmental stage. Based on investigations using both light and scanning electron microscopes, we divided the embryonic development of the grass snake integument into four phases. The shape of the epidermal cells changes first on the caudal and ventral parts of the embryo, then gradually towards the rostral and dorsal areas. In stage V on the ventral side of the embryo the gastrosteges are formed from single primordia, but on the dorsal side the epidermis forms the scale primordia in stage VII. This indicates that scalation begins on the ventral body surface, and spreads dorsally. The appearance of melanocytes between the cells of the stratum germinativum in stage VII coincides with changes in embryo colouration. The first dermal melanocytes were detected in stage XI so in this stage the definitive skin pattern is formed. In the same stage the epidermis forms the first embryonic shedding complex and the periderm layer begins to detach in small, individual flakes. This process coincides with rapid growth of the embryos. [source]


    Being a generalist herbivore in a diverse world: how do diets from different grasslands influence food plant selection and fitness of the grasshopper Chorthippus parallelus?

    1. Generalist insect herbivores occupy a variety of habitats that differ in food plant composition. Dietary mixing has been proposed as a possibility for generalists to overcome nutritional deficiencies of single plant species, but only a few studies have investigated herbivore feeding and fitness for diets that resemble natural scenarios. We studied feeding behaviour, survival, and reproduction of the generalist grasshopper Chorthippus parallelus raised on food plants of four typical habitats. 2. Grasshopper diet consisted of grasses (92.5%), legumes (6.7%) and, in small quantities, other forbs (0.8%). Diet selection differed between the four food plant mixtures, and depended on grasshopper sex and developmental stage. There was no correlation between the relative abundance of plant species in the field and the fraction of these species in the grasshopper diet. 3. Grasshoppers survived on average for 40.4 ± 1.0 days before maturity, grew 106.8 mg until maturity moult, and females laid 4.1 ± 0.4 egg pods, each of which contained 8.5 ± 0.4 eggs. However, despite the differences in feeding behaviour, grasshopper fitness was the same in all of the four food plant mixtures. While the digestibility of ingested food was similar in the four different treatments, indices indicated differences in the conversion efficiency to body mass. 4. Our results show that C. parallelus is a plastic feeder with no fixed preferences in diet composition. The results emphasise that generalist herbivores can counteract putative quality deficiencies of single food plants by selective dietary mixing. [source]


    The effect of host developmental stage at parasitism on sex-related size differentiation in a larval endoparasitoid

    Abstract. 1For their larval development, parasitoids depend on the quality and quantity of resources provided by a single host. Therefore, a close relationship is predicted between the size of the host at parasitism and the size of the emerging adult wasp. This relationship is less clear for koinobiont than for idiobiont parasitoids. 2As size differentiation in host species exhibiting sexual size dimorphism (SSD) is likely to occur already during larval development, in koinobiont larval endoparasitoids the size of the emerging adult may also be constrained based on the sex of the host caterpillar. 3Sex-specific growth trajectories were compared in unparasitised Plutella xylostella caterpillars and in second and fourth instar hosts that were parasitised by the solitary larval koinobiont endoparasitoid Diadegma semiclausum. Both species exhibit SSD, where females are significantly larger than males. 4Healthy female P. xylostella caterpillars developed significantly faster than their male conspecifics. Host regulation induced by D. semiclausum parasitism depended on the instar attacked. Parasitism in second-instar caterpillars reduced growth compared to healthy unparasitised caterpillars, whereas parasitism in fourth-instar caterpillars arrested development. The reduction in growth was most pronounced in hosts producing male D. semiclausum. 5Parasitism itself had the largest impact on host growth. SSD in the parasitoid is mainly the result of differences in growth rate of the parasitoid,host complex producing male and female wasps and differences in exploitation of the host resources. Female wasps converted host biomass more efficiently into adult biomass than males. [source]


    Parental care in response to natural variation in nest predation pressure in six sunfish (Centrarchidae: Teleostei) species

    Abstract,,, Parental care is an important, energetically costly component of the life history of many fishes. Despite this importance, little is known about how different species of fish vary parental care in response to natural nest predator burdens. In this study, underwater videography was used to quantify parental care activity of six species of syntopic nesting male centrarchid fishes in Lake Opinicon, Ontario, in response to natural predators. This approach was used to test the hypothesis that as offspring develop from eggs to wrigglers, parental care activity should decrease or remain static for fish guarding nests with low predator burden and increase for those with high predator burden, reflecting different external risks. Principal components analysis (PCA) was used to derive common aeration and nest defence variables. Aeration and predator defence activity of the fish varied extensively among species. Parental care behaviours indicative of defence and vigilance (e.g., turning, departures, time away from nest, displays) tended to be highest for species that had the most predation attempts, although this was not entirely consistent. There was also a positive relationship between the defence PCA metric and attempted predation. Defence did not vary with stage of offspring development, although interactions between defence and developmental stage were noted for several species. A trade-off between aeration and defence was not observed. In fact, species that provide high levels of aeration also simultaneously provide high levels of defence. Stage-specific patterns of defence in this study were less apparent than those documented by studies using responses to staged predator intrusions making it unclear as to the extent that fish were responding to the level of the risk to offspring than to the value of the brood. Therefore, combined use of observational and experimental assessments of parental care investment may be most appropriate for refining current theoretical paradigms. [source]


    Laboratory-based reproduction success of ruffe, Gymnocephalus cernuus (L.), in brackish water is determined by maternal properties

    Abstract,, Body length, age, egg size, embryo salinity tolerance and length at hatching of the freshwater (salinity <0.1 ppt, Lake Peipsi) and brackish-water (salinity 2,6 ppt, Pärnu and Matsalu Bay) ruffe, Gymnocephalus cernuus (L.), were examined to reveal their reproductive success in moderate salinity. Eggs of females originating from brackish water were significantly larger than eggs of freshwater females. No correlation between egg size and female size and age was found in brackish-water populations. In the freshwater population there was a small negative correlation between egg size and female size, but no correlation with female age. Fertilisation by sperm of males of different origin (brackish water or freshwater) produced no significant differences at any critical developmental stage (fertilisation, gastrulation, hatching) in the development of eggs from brackish-water or freshwater females at 3.3, 5.5, 7.7 and 9.9 ppt salinity. Survival rates in different salinity depended only on female origin; embryonic salinity tolerance was higher in ruffe inhabiting brackish water. Obviously, embryo salinity tolerance in ruffe is determined by egg qualities. [source]


    Proposed diagnostic criteria for internet addiction

    ABSTRACT Objective The objective of this study was to develop diagnostic criteria for internet addiction disorder (IAD) and to evaluate the validity of our proposed diagnostic criteria for discriminating non-dependent from dependent internet use in the general population. Methods This study was conducted in three stages: the developmental stage (110 subjects in the survey group; 408 subjects in the training group), where items of the proposed diagnostic criteria were developed and tested; the validation stage (n = 405), where the proposed criteria were evaluated for criterion-related validity; and the clinical stage (n = 150), where the criteria and the global clinical impression of IAD were evaluated by more than one psychiatrist to determine inter-rater reliability. Results The proposed internet addiction diagnostic criteria consisted of symptom criterion (seven clinical symptoms of IAD), clinically significant impairment criterion (functional and psychosocial impairments), course criterion (duration of addiction lasting at least 3 months, with at least 6 hours of non-essential internet usage per day) and exclusion criterion (exclusion of dependency attributed to psychotic disorders). A diagnostic score of 2 + 1, where the first two symptoms (preoccupation and withdrawal symptoms) and at least one of the five other symptoms (tolerance, lack of control, continued excessive use despite knowledge of negative effects/affects, loss of interests excluding internet, and use of the internet to escape or relieve a dysphoric mood) was established. Inter-rater reliability was 98%. Conclusion Our findings suggest that the proposed diagnostic criteria may be useful for the standardization of diagnostic criteria for IAD. [source]


    Nutrigenomics: a case study in the measurement of insect response to nutritional quality

    Abstract Recent developments in the area of nutrigenomics hold the promise of providing valuable information about the impact of nutrition on a wide range of biochemical parameters by investigating how nutrition alters global gene expression patterns. Our goal is to use a nutrigenomics approach to identify insect molecular markers that could be used as early indicators of insect responses to different nutritional sources. Such molecular markers could be chosen for degree of expression and evaluated for suitability as nutritional biomarkers by examining developmental and generational expression. Ideal markers would be highly expressed, manifested in the first generation within one developmental stage, and consistent over many generations. Our objective for the current study was to illustrate the potential discovery of molecular markers using Perillus bioculatus (F.) (Heteroptera: Pentatomidae), when reared on an optimal vs. suboptimal diet, and analyze the presence of differentially expressed genes resulting from those treatments. In this paper we present preliminary results of microarray and subtractive hybridization experiments that represent the feasibility of using nutrigenomics to assist in analyzing insect responses to nutritional changes and dietary quality with the intent of stimulating further studies in this area.     Moths boring into Ficus syconia on Iriomote Island, south-western Japan

    Abstract Herbivory in the syconia of six Ficus (Moraceae) species (F. superba, F. varieagata, F. virgata, F. irisana, F. bengutensis and F. septica) was examined in March 2002 on Iriomote Island, south-western Japan. Larvae of two lepidopteran species, Pachybotys spissalis (Guenée) (Pyralidae: Pyraustinae) and Stathmopoda sp. (Stathmopodidae) were observed to bore into the Ficus syconia. The attack rate by the moths varied from 0 to 38.5% across Ficus trees. The interiors of the syconia were heavily grazed by the moth larvae. Because figs (syconia) can be regarded as galls and seeds, according to sex and developmental stage, the moth larvae could be considered as gall or seed herbivores, and predators of fig wasps. Moth attack in the Ficus syconia could cause the destruction of fig wasp populations, as fig wasps develop in the syconia. [source]


    Influence of developmental stage on sensitivity to ammonium nitrate of aquatic stages of amphibians

    Abstract In static renewal experiments, we studied how developmental stage influences the effect of ammonium nitrate on embryonic and larval stages of anuran amphibians. The observed lethal effects caused by ammonium nitrate increased with both concentration and duration of exposure. Significant differences were observed in sensitivity to ammonium nitrate as a function of developmental stage in Discoglossus galganoi, Pelobates cultripes, and Bufo calamita. In D. galganoi and P. cultripes, younger individuals displayed greater acute effects from the chemical fertilizer compared with older individuals. For example, 100% of P. cultripes hatchlings died after 4 d of exposure to a nominal concentration of 225.8 mg N-NO3NH4/L, whereas less than 40% of individuals from older larval stages died when exposed to this concentration. A delay of 4 d in the beginning of the exposure to the chemical was enough to cause significant differences in sensitivity. Bufo calamita showed a higher sensitivity in later larval stages after 12 d of exposure. Hyla meridionalis and B. calamita were less sensitive than the other two species. Peak ammonium nitrate concentrations usually occur when amphibians are breeding and, thus, when the most sensitive aquatic stage is in the water. The developmental stage of the test animals should be considered when evaluating the risk of ammonium nitrate to amphibians. [source]


    Effects of carbaryl on green frog (Rana clamitans) tadpoles: Timing of exposure versus multiple exposures

    Abstract The majority of studies on pesticide impacts have evaluated the effects of single exposures. However, multiple exposures to a pesticide may be more prevalent. The objective of our study was to determine how multiple exposures versus single exposure at different times during development affected survival to metamorphosis, tadpole survival, tadpole mass, and tadpole developmental stage of green frog (Rana clamitans) tadpoles reared at low and high density in outdoor cattle tank ponds. Tadpoles were exposed to carbaryl zero, one, two, or three times at 14-d intervals. We applied single doses of carbaryl at one of three times, specifically during early, mid, or late development. Overall, we found that multiple exposures had a greater impact than single exposures during development. More individuals reached metamorphosis in ponds exposed to multiple doses of carbaryl compared with controls, indicating that the presence of carbaryl stimulated metamorphosis. The presence of carbaryl in the aquatic environment also resulted in more developed tadpoles compared with controls. Tadpoles in control ponds did not reach metamorphosis and were less developed than individuals exposed to carbaryl; this effect indicates that, under ideal conditions, green frogs could overwinter in ponds so that greater size could be attained before metamorphosis in the following spring or summer. Our study demonstrated the importance of including realistic application procedures when evaluating the effects of a pesticide and that multiple exposures to a short-lived pesticide are more likely to affect an amphibian population.     New plant protection information system in Hungary,

    Data on the occurrence and spread of pests has been supplied by the Hungarian Plant Protection Organization for over 40 years. Since the 1970s, this service has been operated by the central station and the 20 county stations of the Plant Protection Organization. Data recorded on pest occurrence and developmental stage of pests and plants has been used to run a forecasting system at local and national levels. However, because of significant staff reductions in the Plant Protection Organization and the disappearance of independent forecasting groups, the reliability of the system, which was based on very specific data, has decreased. It has become necessary to develop a more reliable and flexible computer-aided system (Plant Protection Information System, PPIS), better adjusted to the political and economic changes that have occurred. A major element of the new system, introduced in 1997, is that excessively detailed recording was replaced by a more practical general approach with fewer subjective errors. Specialists from the county stations of the Plant Protection Organization monitor infestation levels of 73 pests in 20 crops using five qualification categories. The results obtained are input into the PPIS program, which processes them into user-friendly charts and maps showing the plant health situation in each county and in the country as a whole, as a basis for taking decisions on plant protection measures.     Laminar xanthine oxidase, superoxide dismutase and catalase activities in the prodromal stage of black-walnut induced equine laminitis

    Summary Reasons for study: Xanthine oxidase (XO)-dependent production of superoxide anion and hydrogen peroxide, a characteristic of ischaemia-reperfusion injury, may contribute to the development of equine laminitis. Objective: To determine the levels of XO and antioxidant enzymes (catalase, superoxide dismutase [SOD]) in the digital laminae of normal horses (CON) and horses in the developmental stage of laminitis using the black walnut extract (BWE) model. Methods: Healthy horses (n = 12) were administered BWE (BWE group, n = 6), or water (CON group, n = 6) through a nasogastric tube. At the onset of leucopenia in the BWE-treated animals, all horses were anaesthetised, digital laminae and other samples collected rapidly and flash frozen, and the animals subjected to euthanasia. Extracts of the frozen tissues were assayed for the 2 conformational forms of xanthine: oxygen oxidoreductase (XOR), namely, xanthine dehydrogenase (XDH) and xanthine oxidase (XO), as well as the antioxidant enzymes, SOD and catalase. Results: Extracts of liver, lungs and skin, but not digital laminae, from either CON or BWE-treated horses had endogenous SOD, whereas all had endogenous XO and catalase. The levels of XDH, XO and catalase were similar in extracts of laminae from CON and BWE-treated horses as was the ratio of XDH to XO in extracts. Conclusions and potential relevance: The absence of increased XO activity suggest against the involvement of this reactive oxygen intermediate-generating system in the development of laminar pathology in BWE-treated horses. Conversely, the absence of SOD from extracts of equine digital laminae, but not other tissues, suggests that the equine digital laminae are highly susceptible to damage by superoxide anion, produced, for example, by emigrant inflammatory leucocytes.
