Culture-based fisheries in non-perennial reservoirs in Sri Lanka: production and relative performance of stocked species

Abstract, In Sri Lanka, there is a great potential for the development of culture-based fisheries because of the availability of around 12 000 non-perennial reservoirs in the dry zone (<187 cm annual rainfall) of the island. These reservoirs fill during the north-east monsoonal period in October to December and almost completely dry up during August to October. As these non-perennial reservoirs are highly productive, hatchery-reared fish fingerlings can be stocked to develop culture-based fisheries during the water retention period of 7,9 months. The present study was conducted in 32 non-perennial reservoirs in five administrative districts in Sri Lanka. These reservoirs were stocked with fingerlings of Indian (catla Catla catla Hamilton and rohu Labeo rohita Hamilton) and Chinese (bighead carp Aristichthys nobilis Richardson) major carps, common carp Cyprinus carpio L., genetically improved farmed tilapia (GIFT) strain of Nile tilapia, Oreochromis niloticus (L.) and post-larvae of giant freshwater prawn, Macrobrachium rosenbergii De Man, at three different species combinations and overall stocking densities (SD) ranging from 218 to 3902 fingerlings ha,1, during the 2002,2003 culture cycle. Of the 32 reservoirs stocked, reliable data on harvest were obtained from 25 reservoirs. Fish yield ranged from 53 to 1801 kg ha,1 and the yields of non-perennial reservoirs in southern region were significantly (P < 0.05) higher than those in the northern region. Naturally-recruited snakehead species contributed the catches in northern reservoirs. Fish yield was curvilinearly related to reservoir area (P < 0.05), and a negative second order relationship was evident between SD and yield (P < 0.05). Chlorophyll- a and fish yield exhibited a positive second order relationship (P < 0.01). Bighead carp yield impacted positively on the total yield (P < 0.05), whereas snakehead yield impact was negative. Bighead carp, common carp and rohu appear suitable for poly-culture in non-perennial reservoirs. GIFT strain O. niloticus had the lowest specific growth rate among stocked species and freshwater prawn had a low return. [source]


Bacteriotherapy with Lactobacillus plantarum in burns

Abstract Bacterial colonisation and infection remain the major causes of delayed healing and graft rejection following burns. Topical treatment is necessary to reduce the incidence of burn wound infection. Silver sulphadiazine (SD-Ag) is an often used microbicidal agent. However, this treatment produces adverse reactions and side-effects. On the basis of experimental data and clinical application of lactobacilli as probiotics, we performed this exploratory study to establish the effectiveness of bacteriotherapy with topical application of the innocuous bacteria Lactobacillus plantarum cultured in De Man, Rogosa and Sharpe medium to provide an alternative method for burn treatment using SD-Ag as a reference. These innocuous bacteria would compete with other bacteria that are wound pathogens and would modify the wound environment and promote tissue repair. Eighty burned patients from the Plastic Surgery and Burns Unit were grouped into infected (delayed) second- and third-degree and non infected (early) third-degree burns and treated with L. plantarum or SD-Ag. The proportion of patients with delayed second-degree burns was 0·71 for L. plantarum and 0·73 for SD-Ag (relative rate: ,2·72%) with respect to the decrease in bacterial load (<105 bacteria/g of tissue), promotion of granulating tissue wound bed and healing. In early third-degree burns, the values were 0·75 for L. plantarum and 0·84 for SD-Ag (relative rate: ,1·07%) in preventing wound infection and promotion of granulation tissue, 0·90 in graft taking for both treatments (relative rate: 0%) and 0·75 for L. plantarum and 0·77 for SD-Ag (relative rate: ,2·60%) in healing. In delayed third-degree burns, values were 0·83 for L. plantarum and 0·71 for SD-Ag (relative rate: +16·90%) with respect to the decrease in the bacterial load (<105 bacteria/g of tissue) and providing a granulating tissue wound bed, 0·90 in graft taking for both treatments (relative rate: 0%) and 0·75 for L. plantarum and 0·64 for SD-Ag (relative rate: + 17·19%) in healing. Although the number of patients (between 12 and 15 per group) did not enable the application of a power statistical test, these results suggest that the L. plantarum treatment should be studied in greater depth and could be used as a valid alternative for the topical treatment of burns. Liver oil of pharaoh cuttlefish Sepia pharaonis Ehrenberg, 1831 as a lipid source in the feed of giant freshwater prawn, Macrobrachium rosenbergii (De Man 1879)

Abstract The efficacy of pharaoh cuttlefish (Sepia pharaonis Ehrenberg, 1831) liver lipid in the feed for juveniles of giant freshwater prawn Macrobrachium rosenbergii (De Man 1879) was tested by feeding five experimental diets prepared using clam meat, groundnut oil cake, wheat bran, tapioca powder, vitamin,mineral mixture and cellulose powder as the major ingredients. The test diets T1, T2, T3, T4 and T5, containing five levels (1, 2, 3, 4 and 5%, respectively) of lipid, extracted from the liver of pharaoh cuttlefish, were fed to 30-day-old prawn juveniles for 60 days, in triplicates and compared against a control. Analysis of variance of the growth parameters showed that the juveniles fed with T2, containing 2% cuttlefish liver lipid with a total lipid content of 9.85% and lipid,carbohydrate ratio of 1 : 3.8, showed significantly better growth (P < 0.05) with the highest weight gain of 118%, greatest food conversion and protein utilization efficiency, and the highest body protein content, although the survival rates remained unaffected among treatments. Growth of the juveniles, however, declined at lipid levels >2%. Addition of cuttlefish liver lipid led to an increase of ,-3 and ,-6 fatty acids in T2, the proportion of ,-6 being higher than the former. The crude lipid content of the body of test animals also increased with the increase in dietary lipid from 1.51% in T1 to 3.27% in T5. The present study indicates that cuttlefish liver lipid can be successfully used as a substitute for conventional lipid sources in rearing juveniles of giant freshwater prawn, an eco-friendly practice in recycling cuttlefish waste , a discarded fishery by-product. Use of Cyclop-eeze as a substitute for Artemia nauplii in larval rearing of giant freshwater prawn Macrobrachium rosenbergii (De Man 1879)

Abstract Four feeding experiments, replacing 25% (T1), 50% (T2), 75% (T3) and 100% (T4), by dry weight, of the live feed Artemia nauplii for Cyclop-eeze, a new larval feed that was claimed to contain the highest known levels of astaxanthin and omega-3 polyunsaturated fatty acids, were compared against a control that was fed with Artemia and egg custard alone, to the larvae of giant freshwater prawn Macrobrachium rosenbergii (De Man 1879). Analysis of different production characteristics of the larvae revealed that the highest survival up to postlarvae (PL) stage was obtained for T2 in which 50% of the Artemia nauplii were replaced by Cyclop-eeze [freeze-dried (FD) deep frozen (DF)], and the highest astaxanthin content of the larval tissue obtained in T4 in which the larvae were fed 100% Cyclop-eeze, although the survival rate was the lowest in this treatment. The costs of different treatments were also compared. The Artemia consumption million,1 larvae was the highest in control (11490 g), followed by T1 (8240 g), T2 (4990 g), T3 (3730 g) and T4, which completely replaced Artemia from stage 5 onwards (1830 g). The highest consumption of Cyclop-eeze million,1 larvae was in T4 (1670 and 10 880 g), followed by T3 (850 and 5560 g), T2 (410 and 2690 g) and T1 (230 and 1490 g) of FD and DF, respectively. The astaxanthin contents of the late-stage larvae fed under the four treatments were 24.90, 27.40, 28.60 and 35.60 ,g g,1 tissue for T1, T2, T3 and T4, respectively, while that of the control was 23.70 ,g g,1. The lowest cost of live feeds million,1 PL was obtained for T2 (US$ 428.60), followed by T1 (US$ 490.46), control (US$ 529.07) and T3 (US$ 583.26), while it was the highest for T4 (US$ 890.93). The results indicated that Cyclop-eeze could economically replace Artemia nauplii at 50% level that could significantly improve the survival and carotenoid composition of the larvae of M. rosenbergii. Effects of different dosages of astaxanthin on giant freshwater prawn Macrobrachium rosenbergii (De Man) challenged with Lactococcus garvieae

Abstract The effects of astaxanthin (AX) injected at 0.67 and 1.34 nmol g,1 BW,1 on the survival, antioxidant capacity, total haemocyte count (THC) and hepatopancreas astaxanthin content of giant freshwater prawn, Macrobrachium rosenbergii, challenged with Lactococcus garvieae were evaluated. AX-injected M. rosenbergii at 1.34 nmol g,1 BW,1 had significantly (P,0.05) higher survival rates. However, AX showed no significant effects on the capacity of certain antioxidant indicators (superoxide dismutase, glutathione peroxidase and glutathione reductase). This implies that L. garvieae infection suppressed the activity of the haemolymph antioxidant system of infected M. rosenbergii. This result suggests that the two different dosages used in this study could not exert significant effects on the tested antioxidant capacity of L. garvieae -infected M. rosenbergii. On the other hand, AX-injected M. rosenbergii at either dose showed a significant increase in the THC and hepatopancreas AX content when compared with the challenged control group. Overall, the results of this study indicate that the injected AX led to an improvement in M. rosenbergii's resistance against L. garvieae infection. [source]


Identification and partial characterization of selected proteolytic enzymes in the digestive system of giant freshwater Prawn Macrobrachium rosenbergii (De Man) Postlarvae

Abstract Biochemical assays and substrate SDS-PAGE were conducted to partially characterize and identify various types of proteases present in the digestive tract of PL15 giant freshwater prawn (Macrobrachium rosenbergii). Casein hydrolytic assay of the enzyme extracts showed major proteolytic activities at pH 3.0, 6.0 and 9.0, while assay of preincubated enzyme extracts with phenylmethylsulphonyl fluoride (PMSF), a serine protease inhibitor produced a 33.17% reduction in alkaline protease activity. When specific inhibitors tosyl-lysine chloromethyl ketone and tosyl-phenylalanine chloromethyl ketone were used, they resulted in a reduction in activity of proteases in the enzyme extracts by 82.41% and 55.03%, respectively, confirming the presence of trypsin and chymotrypsin, while ethylenediamine tetraacetic acid produced protease activity reduction in 33.92% showing the presence of metalloproteases in the digestive tract of the prawn. Further characterization of the alkaline proteases using SDS-PAGE technique, after incubating the extract in the presence or absence of specific inhibitors, produced six bands corresponding to molecular masses of between 13.48 and 136.1 kDa; two trypsin bands of 13.48 and 36.4 kDa, three chymotrypsin bands in the range of 23.0,73.4 kDa and one for metalloprotease of 136.1 kDa, all of which were identified from a zymogram. This study suggests that protein digestion in M. rosenbergii is initiated by an acid protease followed by a combination of action of alkaline proteases: trypsin, chymotrypsin and metalloproteases. Growth and carcass composition of giant freshwater prawn, Macrobrachium rosenbergii (De Man), fed different isonitrogenous and isocaloric diets

Abstract A feeding experiment was conducted for 135 days to observe the effect of different isonitrogenous (35% crude protein) and isocaloric (385 kcal) diets on the growth and carcass composition of giant freshwater prawn, Macrobrachium rosenbergii (De Man 1879). Three experimental diets (ED1, ED2 and ED3) were prepared using locally available ingredients. These diets differed mainly in terms of percent contribution of major protein sources such as fish meal, soybean meal, groundnut oil cake and mustard oil cake. Experimental diets were evaluated against a commercial diet, which served as the control (CD). Juveniles 1.87,2.44 g in size were stocked at a population density of 40 000 ha,1 and fed thrice daily at 10% in the beginning and reducing gradually to 7% and 5% of the body weight during the experimental period. No significant differences (P>0.05) in the growth performance were observed; however, a significantly (P<0.05) higher yield (721.9 kg ha,1 135 days,1) was recorded for prawn fed with control diet, followed by experimental diet 2 (676.5 kg ha,1 135 days,1, having soybean meal as a major protein source). The survival ranged between 63.8% and 77.7%, with different diets showing significantly higher survival. The apparent feed conversion ratio values of diets ranged between 3.15 and 3.49, with experimental and control diets showing non-significantly lower AFCR values. At the end of the experiment, representative specimens from each treatment were collected and their carcass composition was measured. Analysis of variance showed that carcass protein and total carbohydrate contents were significantly (P<0.05) higher in prawns fed on a fish,soybean meal-based diet (ED3) and a control diet. The total lipid contents of prawns, however, did not differ significantly among the various dietary treatments. The results of our study suggest that the experimental diets could be used effectively for M. rosenbergii without compromising growth and flesh quality. [source]


Economic analysis of monosex culture of giant freshwater prawn (Macrobrachium rosenbergii De Man): a case study

Abstract All-male monosex culture of Macrobrachium rosenbergii (De Man) has emerged as a popular practice in India, especially in the state of Andhra Pradesh. A study was conducted to compare the economics of all-male, mixed and all-female culture in 15 adjacent, rectangular ponds of 4000 m2 each by stocking juveniles previously reared in a nursery for 60 days. The experiment was conducted using a completely randomized design with three treatments; T1 (all male), T2 (mixed) and T3 (all female), and five replicates for a period of 5 months after the nursery phase. Statistical analysis showed highly significant (P<0.01) differences among the three types of culture. The cost of production was estimated and the economic feasibility of the culture methods was evaluated by cost-return and partial budgeting analysis. The average weight, productivity and specific growth rate were the highest for all male culture, being 80.92±2.41 g, 1532 kg ha,1 and 1.97±0.02 respectively. All-female culture registered significantly higher survival (89.16±0.77%) and the best apparent feed conversion ratio of 1.26±0.02. The economic analysis revealed that all-male monosex culture of M. rosenbergii was 63.13% and 60.20% more profitable than mixed and all-female cultures respectively. [source]


The androgenic gland and monosex culture of freshwater prawn Macrobrachium rosenbergii (De Man): a biotechnological perspective

Abstract Males of the freshwater prawn Macrobrachium rosenbergii (De Man) grow faster and reach a larger size at harvest than females of the species. It is thus obvious that culture of monosex all-male populations would be economically advantageous. Sexual differentiation in crustaceans is regulated by the androgenic gland (AG), which plays a pivotal role in the regulation of male differentiation and in the inhibition of female differentiation. In M. rosenbergii, AG removal from immature males resulted in sex reversal, with complete female differentiation. Similarly, AG implantations into immature females lead to the development of the male reproductive system. Sex-reversed M. rosenbergii animals were capable of mating with normal specimens to produce offspring. Early attempts in Israel and more recently, attempts in other countries to establish all-male populations through manual segregation showed that for the production of monosex prawn populations to be economically feasible, intervention via the AG is probably required. However, a suitable biotechnology is still to be developed, and an androgenic hormone has yet to be identified in decapods. Three lines of aquacultural and biotechnological research and development are proposed for the future: (1) Establishment of monosex cultures through manual segregation, together with the application of selective harvesting and claw ablation, as well as examination of different monosex culture strategies under a variety of economic conditions. (2) Microsurgical intervention in the AG, leading to the development of functional neo-females, which would subsequently be mated with normal males to produce all-male progeny. (3) Elucidation of AG bioactive products to enable biochemical or molecular manipulation of sex differentiation. [source]


Teaching & Learning Guide for: Victorian Life Writing

Author's Introduction The Victorian period was one of the great ages for life-writing. Though traditionally renowned for its monumental ,lives and letters', mainly of great men, this was also a time of self-conscious anxiety about the genre. Critics and practitioners alike were unsure who should be writing autobiography, and whether its inherent assertiveness ruled out all but public men as appropriate subjects. It was also a period of experimentation in the different genres of life-writing , whether autobiography, journals, letters, autobiographical novels, and narratives of lives combined with extracts from correspondence and diaries. Victorian life-writing therefore provides rich and complex insights into the relationship between narrative, identity, and the definition of the self. Recent advances in criticism have highlighted the more radical and non-canonical aspects of life-writing. Already a latecomer to the literary-critical tradition (life-writing was for a long time the ,poor relation' of critical theory), auto/biography stresses the hidden and silent as much as the mainstream and vocal. For that reason, study of Victorian life-writing appeals to those with an interest in gender issues, postcolonialism, ethnicity, working-class culture, the history of religion, and family and childhood studies , to name but a few of the fields with which the genre has a natural connection. Author Recommends A good place to start is the two canonical texts for Victorian life-writing: George P. Landow's edited collection, Approaches to Victorian Autobiography (Athens, OH: Ohio University Press, 1979) and Avrom Fleishman's Figures of Autobiography: The Language of Self-Writing in Victorian and Modern England (Berkeley and Los Angeles, CA: University of California Press, 1983). These two re-ignited interest in Victorian life-writing and in effect opened the debate about extending the canon, though both focus on the firmly canonical Ruskin and Newman, among others. By contrast, David Amigoni's recently edited collection of essays, Life-Writing and Victorian Culture (Aldershot: Ashgate 2006) shows how far the canon has exploded and expanded: it begins with a useful overview of the relationship between lives, life-writing, and literary genres, while subsequent chapters by different authors focus on a particular individual or family and their cultural interaction with the tensions of life-writing. As this volume is fairly male-dominated, readers with an interest in women's life-writing might prefer to start with Linda Peterson's chapter, ,Women Writers and Self-Writing' in Women and Literature in Britain 1800,1900, ed. Joanne Shattock (Cambridge: Cambridge University Press, 2001), 209,230. This examines the shift from the eighteenth-century tradition of the chroniques scandaleuses to the professional artist's life, domestic memoir, and spiritual autobiography. Mary Jean Corbett's Representing Femininity: Middle-Class Subjectivity in Victorian and Edwardian Women's Autobiographies (New York, NY: Oxford University Press, 1992) begins with material on Wordsworth and Carlyle, but ,aims to contest the boundaries of genre, gender, and the autobiographical tradition by piecing together a partial history of middle-class women's subjectivities in the nineteenth and early twentieth centuries' (3). Corbett is particularly interested in the life-writing of actresses and suffragettes as well as Martineau and Oliphant, the first two women autobiographers to be welcomed into the canon in the 1980s and 90s. Laura Marcus's Auto/biographical Discourses, Theory, Criticism, Practice (Manchester and New York, NY: Manchester University Press, 1994) revises and updates the theoretical approaches to the study of life-writing, stressing both the genre's hybrid qualities, and its inherent instability: in her view, it ,comes into being as a category to be questioned' (37). Another of her fruitful suggestions is that autobiography functions as a ,site of struggle' (9), an idea that can be applied to aesthetic or ideological issues. Her book is divided between specific textual examples (such as the debate about autobiography in Victorian periodicals), and an overview of developments in critical approaches to life-writing. Her second chapter includes material on Leslie Stephen, who is also the first subject of Trev Lynn Broughton's Men of Letters, Writing Lives: Masculinity and Literary Auto/biography in the Late Victorian Period (London: Routledge, 1999) , her other being Froude's controversial Life of Carlyle. With the advent of gender studies and masculinities, there is now a return to male forms of life-writing, of which Martin A. Danahay's A Community of One: Masculine Autobiography and Autonomy in Nineteenth-Century Britain (Albany, NY: State University of New York Press, 1993) is a good example. Danahay argues that nineteenth-century male autobiographers present themselves as ,autonomous individuals' free of the constraints of social and familial contexts, thus emphasizing the autonomy of the self at the expense of family and community. Online Materials My impression is that Victorian life-writing is currently better served by books than by online resources. There seem to be few general Web sites other than University module outlines and reading lists; for specific authors, on the other hand, there are too many to list here. So the only site I'd recommend is The Victorian Web: http://.victorianweb.org/genre/autobioov.html This Web site has a section called ,Autobiography Overview', which begins with an essay, ,Autobiography, Autobiographicality and Self-Representation', by George P. Landow. There are sections on other aspects of Victorian autobiography, including ,Childhood as a Personal Myth', autobiography in Dickens and Elizabeth Barrett Browning, and a list of ,Suggested Readings'. Each section is quite short, but summarizes the core issues succinctly. Sample Syllabus This sample syllabus takes students through the landmarks of Victorian life-writing, and demonstrates the development of a counter-culture away from the mainstream ,classic male life' (if there ever was such a thing) , culminating in the paired diaries of Arthur Munby (civil servant) and Hannah Cullwick (servant). Numerous other examples could have been chosen, but for those new to the genre, this is a fairly classic syllabus. One week only could be spent on the ,classic male texts' if students are more interested in pursuing other areas. Opening Session Open debate about the definition of Victorian ,life-writing' and its many varieties; differences between autobiography, autobiographical fiction, diary, letters, biography, collective biography, and memoir; the class could discuss samples of selected types, such as David Copperfield, Father and Son, Ruskin's Praeterita, and Gaskell's Life of Charlotte Brontė. Alternatively, why not just begin with Stave Two of Dickens's A Christmas Carol (1843), in which the First Spirit takes Scrooge back through his childhood and youth? This is a pretty unique type of life-writing, with Scrooge ,laughing and crying' as his childhood and youth are revealed to him in a series of flashbacks (a Victorian version of ,This is Your Life?'). The dual emotions are important to note at this stage and will prompt subsequent discussions of sentimentality and writing for comic effect later in the course. Week 2 Critical landmarks: discussion of important stages in the evolution of critical approaches to life-writing, including classics such as Georges Gusdorf's ,Conditions and Limits of Autobiography', in Autobiography: Essays Theoretical and Critical, ed. James Olney (Princeton, NJ: Princeton University Press, 1980), 28,47; Philippe Lejeune's ,The Autobiographical Pact', in On Autobiography, ed. Paul John Eakin, trans. Katherine Leary (original essay 1973; Minneapolis: University of Minnesota Press, 1989), 3,30; and Paul De Man's ,Autobiography as De-Facement', Modern Language Notes 94 (1979): 919,30. This will provide a critical framework for the rest of the course. Weeks 3,4 Extracts from the ,male classics' of Victorian life-writing: J. S. Mill's Autobiography (1873), Ruskin's Praeterita (1885,89), and Newman's Apologia pro Vita Sua (1864). What do they think is important and what do they miss out? How open or otherwise are they about their family and personal lives? Are these essentially ,lives of the mind'? How self-aware are they of autobiographical structures? Are there already signs that the ,classic male life' is fissured and unconventional? An option here would be to spend the first week focusing on male childhoods, and the second on career trajectories. Perhaps use Martin Danahay's theory of the ,autonomous individual' (see above) to provide a critical framework here: how is the ,Other' (parents, Harriet Taylor) treated in these texts? Weeks 5,6 Victorian women's autobiography: Harriet Martineau's Autobiography (1877) and Margaret Oliphant's Autobiography (1899): in many ways these are completely unalike, Martineau's being ordered around the idea of steady mental growth and public recognition, while Oliphant's is deeply emotional and disordered. Can we therefore generalize about ,women's autobiography'? What impact did they have on Victorian theories of life-writing? Students might like to reconsider Jane Eyre as an ,autobiography' alongside these and compare scenes of outright rebellion. The way each text handles time and chronology is also fascinating: Martineau's arranged to highlight stages of philosophical development, while Oliphant's switches back and forth in a series of ,flashbacks' to her happier youth as her surviving two sons die ,in the text', interrupting her story. Week 7 Black women's autobiography: how does Wonderful Adventures of Mrs Seacole in Many Lands (1857) differ from the Martineau and Oliphant autobiographies? What new issues and genre influences are introduced by a Caribbean/travelogue perspective? Another key text would be Harriet Jacobs's Incidents in the Life of a Slave-Girl (1861). How representative and how individual are these texts? Do these authors see themselves as representing their race as well as their class and sex? Comparison of the performances of different fermentation strategies on cell growth and bacteriocin production by Lactobacillus curvatus CWBI-B28

Abstract The dynamics of cell growth and bacteriocin production by Lactobacillus curvatus CWBI-B28 in modified De Man/Rogosa/Sharp (mMRS) broth with various concentrations of glucose and complex nitrogen source (CNS; peptone, yeast extract and meat extract) was investigated in flask fermentations and in a laboratory fermentor using batch and fed-batch cultivations. In fed-batch fermentation the rate of feeding of the reactor with the substrates was either maintained constant (0.12 L h,1) or varied exponentially as a function of time. The results showed that both cell growth and bacteriocin activity were influenced by changes in the concentrations of glucose and CNS. Optimal growth and bacteriocin activity were obtained in mMRS broth containing 40 g L,1 glucose and 40 g L,1 CNS (mMRS40/40). A bacteriocin titre of 4266 AU mL,1 and a cell count of 8.7 log colony-forming units (cfu) mL,1 were recorded when this medium was used for cultivation. In batch fermentation using the same medium, a higher cell count (9.5 log cfu mL,1) and twice as much bacteriocin as in flask fermentation were produced. The highest bacteriocin titre (8533 AU mL,1) was obtained with fed-batch fermentation at an exponentially varying rate of feeding. Bacteriocin activity and cell dry mass did not always correlate. Bycatch in a tropical schooling , penaeid fishery and comparisons with a related, specialised trawl regime

Abstract, The bycatch in a tropical Australian banana prawn, Penaeus merguiensis (de Man), fishery is described and contrasted with the closely related tiger prawn, Penaeus semisulcatus De Haan and Penaeus esculentus Haswell, fishery. Most of the similarity between banana prawn fishery catches was accounted for by three teleost species constituting 51.4% of the total bycatch weight. The bycatch assemblage structure of this fishery was significantly different (P < 0.05) from the adjoining tiger prawn fishery. The banana prawn fishery had a higher mean bycatch catch rate from shorter duration trawls, but lower estimated total annual bycatch (1502 t yr,1) than the longer duration trawls of the tiger prawn fishery (20 073 t yr,1). This study provides new data for quantifying bycatch and improving the accuracy of quantitative risk assessments currently being used to demonstrate sustainability of bycatch populations. The information will be incorporated into collaborative development of a long-term monitoring programme. [source]


Macrobrachium rosenbergii nodavirus infection in M. rosenbergii (de Man) with white tail disease cultured in Taiwan

Abstract White tail disease (WTD) is a serious problem in Macrobrachium rosenbergii hatcheries and nursery ponds in Asia. The causative agents have been identified as M. rosenbergii nodavirus (MrNV) and its associated extra small virus. This is the first report demonstrating MrNV virus in M. rosenbergii displaying WTD signs in Taiwan by reverse transcriptase-polymerase chain reaction (RT-PCR). Amplified fragments of 850 and 425 bp for RNA-1 and RNA-2 of MrNV, respectively, were obtained by RT-PCR. RT-PCR products of about 850 and 1121 bp for RNA-1 and RNA-2 of MrNV were also obtained using different primer pairs. The amplicons were individually cloned into pGEM-T vector and sequenced. Using this recombinant plasmid of MrNV RNA-2 as DNA template, the non-radioactive DNA probes were prepared by PCR amplification with DIG,11-dUTP. The probes were used to successfully detect MrNV infection in the striated muscle tissues of WTD-diseased prawns using in situ hybridization. The 1121 bp genomic fragment of RNA-2 of MrNV consisted of a unique open reading frame with 1116 nucleotides, and it encoded a structural protein with 371 amino acids. The nucleotide sequence of the partial genome of MrNV RNA-2 revealed a 97% identity with an Indian isolate. A phylogenetic tree constructed using the nucleotide sequence of the viral capsid gene from insect and fish nodaviruses revealed that the MrNV Taiwan isolate could be interpreted as a new genus within the family Nodaviridae. However, its position showed more affinity with Alphanodavirus than with Betanodavirus. The study confirmed the presence of MrNV infection in freshwater prawns cultured in Taiwan suffering from WTD. A sandwich enzyme linked immunosorbent assay (S-ELISA) for detection of MrNV in the giant freshwater prawn, Macrobrachium rosenbergii (de Man)

Abstract A sandwich enzyme-linked immunosorbent assay (S-ELISA) was developed to improve diagnosis of white tail disease of the giant freshwater prawn, Macrobrachium rosenbergii, caused by the nodavirus, Mr NV. Polyclonal antibodies were produced by immunization of Balb/C mice using a purified suspension of the virus and IgG anti- MrNV were purified from ascitic fluid. A sandwich method was successfully developed, coating first with unlabelled antibody and detecting trapped antigens with a second biotinylated antibody. Reaction was demonstrated using an avidin,peroxidase conjugate. Tissue extracts from M. rosenbergii infected with MrNV or purified viral extracts (control) were successfully identified in an individual ELISA, thus confirming the validity of the method. This S-ELISA should be the technique of choice for epidemiological studies of this disease and is a rapid and inexpensive assay with high specificity and sensitivity. [source]


Experimental susceptibility of different life-stages of the giant freshwater prawn, Macrobrachium rosenbergii (de Man), to white spot syndrome virus (WSSV)

Studies were conducted by injecting/feeding white spot syndrome virus (WSSV) derived from infected shrimp, Penaeus monodon (Fabricius), to different life-stages, namely post-larvae, juveniles, sub-adults and adults of Macrobrachium rosenbergii (de Man). The disease was also induced in brood stock, and the eggs and larvae derived from these animals were subsequently tested for WSSV infection. All the stages except egg used for the experiment were found WSSV positive in histopathology, cross infection bioassay and polymerase chain reaction (PCR) analysis. Experimentally infected post-larvae and juveniles showed a high percentage of mortality and an increased rate of cannibalism. The cumulative mortality in post-larvae was up to 28%; with 28,40% cannibalism resulting in a maximum loss of up to 68%. In juveniles, observed mortality and cannibalism were 10,20% and 6.7,30.0%, respectively, and the maximum loss recorded was 50%. In sub-adults, mortality ranged from 2.8 to 6.7%, cannibalism was up to 20% and the total loss was up to 26.7%. Sub-adults and adults were found to be more tolerant to the infection as evidenced by the mortality pattern. A nested (two-step) PCR resulted in a 570-bp product specific to WSSV in all stages, except the eggs. [source]


MICROBIOLOGICAL AND PHYSICOCHEMICAL CHARACTERIZATION OF NATURAL FERMENTED CAMEL MEAT SAUSAGE

ABSTRACT In this study, fermentations of camel meat were followed by analyzing the microbiological and physicochemical aspects of this product. The sausages were characterized by an important microbial activity of lactic acid bacteria that resulted in a product with a final pH of about 5.06. No Listeria monocytogenes, Salmonella spp. and sulfite-reducing clostridia were ever isolated from the raw materials or the fermented sausages during the maturation, underlining the safety of this product. The final water activity of the product was 0.91. Identification showed that the majority of lactobacilli isolated from de Man,Rogosa,Sharpe agar strains were assigned to the species of Lactobacillus plantarum. PRACTICAL APPLICATIONS The production of fermented foods is based on the use of starter cultures, for instance lactic acid bacteria that initiate rapid acidification of the raw material. They contribute to the microbial safety or offer one or more organoleptic, technological, nutritional, or health advantages. Also, their production of acetic acid, ethanol, aroma compounds, bacteriocins, exopolysaccharides, and several enzymes is of importance. In this way they enhance shelf life and microbial safety, improve texture, and contribute to the pleasant sensory profile of the end product. [source]


Derrida's Defence of Paul de Man's Wartime Writings: A Deconstructionist Dilemma

Derrida's attempt at a defence of Paul de Man's wartime writings put him in a difficult position. Had he remained loyal to his usual deconstructionist practice of interpretation, he would have been unable to defend de Man in a politically effective way. Derrida therefore chose a hybrid form of interpretation that is neither purely deconstructionist nor easily classifiable in any other way. Faced with a case in which a purely deconstructionist reading would not have achieved his aim of minimising the political damage caused by the discovery of de Man's wartime writings, Derrida opted for an interpretive approach which allowed him to read into de Man's texts what he wanted to get out of them, ignoring what seems obvious to less biased readers. Effect of different diets on larval production, quality and fatty acid profile of the marine ornamental shrimp Lysmata amboinensis (de Man, 1888), using wild larvae as a standard

Abstract The present work evaluates the influence of broodstock diets [Marine Cuisine®, MC, MC supplemented with highly unsaturated fatty acid- (HUFA) enriched Artemia biomass , MC + AB, and MC supplemented with squid , MC + S] on larval production, newly hatched and early zoeal stage survival and fatty acid profile of newly hatched larvae of Lysmata amboinensis. These parameters are compared with those from larvae hatched from embryos spawned in the wild. The number (±SE) of larvae produced with MC and MC + S (1077 ± 219 and 1103 ± 184, respectively) was similar to that in broodstock carrying embryos spawned in the wild (1224 ± 111), while those fed MC + AB displayed significantly lower values (1044 ± 161). Larvae produced with MC + AB displayed lower survival for all starvation periods, while larvae spawned in the wild displayed the highest survival. No larvae resisted 144 h of starvation and none moulted to zoea II. The fatty acid comparison revealed that larvae from embryos spawned in the wild displayed the highest levels of DHA, as well as higher DHA/EPA and n -3/n -6 ratios. These results suggest that broodstock diets commonly used to promote ornamental shrimp's maturation (based on mixed frozen components) are far from being optimal. [source]


Probiotic effect of Bacillus NL110 and Vibrio NE17 on the survival, growth performance and immune response of Macrobrachium rosenbergii (de Man)

Abstract Eight hundred and eighty-five strains of bacterial isolates from various samples associated with the natural habitat of Macrobrachium rosenbergii were screened for their probiotic potential. Two putative probionts namely Bacillus NL110 and Vibrio NE17 isolated from the larvae and egg samples, respectively, were selected for experimental studies and were introduced to the juveniles of M. rosenbergii (0.080±0.001 g) through different modes such as through feed, water and both. The probiotic potential of the above bacteria in terms of improvements in water quality, growth, survival, specific growth rate (SGR), feed conversion ratio and immune parameters was evaluated. The treatment groups showed a significant improvement in SGR and weight gain (P<0.001). Survival among different treatment groups was better than that in the control group. There were also significant improvements in the water quality parameters such as the concentration of nitrate and ammonia in the treatment groups (P<0.05). Improvements in immune parameters such as the total haemocyte count (P<0.05), phenoloxidase activity and respiratory burst were also significant (P<0.001). It is concluded that screening of the natural microflora of cultured fish and shellfish for putative probionts might yield probiotic strains of bacteria that could be utilized for an environment-friendly and organic mode of aquaculture. Dynamic changes in gene expression during vitellogenic stages of the white shrimp: Fenneropenaeus merguiensis de Man

Abstract Ovarian maturation is a crucial step for shrimp brood stock. A suppressive subtractive hybridization was used to identify differentially expressed genes in the ovaries during vitellogenesis of Fenneropenaeus merguiensis. Three- to sevenfold up-regulated genes were selected. A blast search identified nine unique genes. The genes that may be involved in ovarian maturation, namely translationally controlled tumour protein (TCTP), heat shock protein 70 (HSP70), H-L(3)MBT-LIKE, shrimp ovarian peritrophin (SOP), vitellin (Vn), thrombospondin (TSP) and ribosomal protein L10a (RPL10a), were further studied. The transcripts of HSP70, TCTP, SOP and RPL10a in the ovary showed their highest expression in the early stage and declined in the later stages. In contrast, the transcripts of the H-L(3)MBT-LIKE, TSP and Vn genes increased from the early stage to be significantly up-regulated during the late stage. A comparison of gene expression among organs during the vitellogenesis showed that the transcripts of HSP70, SOP, H-L(3)MBT-LIKE and TSP were down-regulated in the brain, intestine, hepatopancreas and lymphoid (except for TSP) when compared with their expression in shrimp with non-developed ovaries. The mRNA of TCTP and RPL10a was significantly over-expressed in the lymphoid and heart, whereas TCTP transcripts were significantly down-regulated in the brain during the vitellogenesis. The molecular behaviour of the transcripts in this study may, in the future, lead to an ability to stimulate the ovarian development in shrimp. [source]


Low-cost diet for monoculture of giant freshwater prawn (Macrobrachium rosenbergii de Man) in Bangladesh

Abstract An experiment was conducted for 3 months in 12 experimental ponds, each of 30 m2, with a view to develop a low-cost diet for monoculture of Macrobrachium rosenbergii in ponds. Three experimental diets (30% protein) were formulated using fish meal, meat and bone meal, mustard oilcake, sesame meal and rice bran in different combinations partially replacing fish meal by meat and bone meal and sesame meal and assigned to treatments T1, T2 and T3 respectively. A commercial golda feed (Starter-III) was assigned to T4 (reference diet). Each treatment had three replicates. Juveniles of M. rosenbergii (2.90±0.21 g) were stocked at the rate of 40 000 ha,1. Prawns were fed three times daily at the rate of 10% and 5% of their body weight at the beginning and for the last 2 months respectively. The ponds were provided with aeration during the night using air pumps. The ranges of water quality parameters recorded in different ponds were: temperature 28.9,32.5°C, dissolved oxygen 5.1,8.1 mg L,1 and pH 6.4,7.7. The results showed that the weight gain of prawns fed diet 1 was significantly higher (P<0.05) than those fed diets 2 and 3, but was not significantly different from those fed diet 4 (reference diet). The feed conversion ratio (FCR) values of diets ranged between 2.21 and 2.96 with diets 1 and 4 showing significantly lower (P<0.05) FCR values. The survivals (%) ranged between 68% and 78% with prawns fed diets 1 and 4 showing significantly higher survival. The production of prawn ranged between 921 and 1428 kg ha,1 and diet 1 resulted in a significantly high (P<0.05) production. A simple economic analysis showed that diet 1 generated the maximum net profit of Tk 159 178 ha,1. The results of the study showed that a diet containing 20% fish meal, 10% meat and bone meal, 15% mustard oilcake, 15% sesame meal, 35% rice bran, 4% molasses and 1% vitamin,mineral premixes may be recommended to the farmers for monoculture of M. rosenbergii in ponds. Management strategies associating batch-graded and size-graded postlarvae can reduce heterogeneous individual growth in Macrobrachium rosenbergii (de Man)

Abstract Two sets of adaptive trials were performed to determine the effects of size grading and batch grading on size heterogeneity in cultured Macrobrachium rosenbergii raised in the Coconut Garden channels of Kuttanad, Southern India. In the first set of trials, postlarvae were batch graded on the basis of their hatching order, segregated and grown separately as first-hatched and second-hatched groups. In the second set of trials, postlarvae were size graded as jumpers and laggards and were grown in separate channels. The average weight attained by prawns after 10 months of culture was highest for jumpers (83.11 g) and lowest for the prawns from the first-hatched group (43.76 g). The percentage of males was highest in the population of jumpers (58.23%). Highest production was recorded in the channel stocked with postlarvae from the second-hatched group (103.4 kg ha,1) and lowest production was obtained from the first-hatched group (63.74 kg ha,1). The proportions of undesirable small males were highest among laggards and the first-hatched group: 24.8% and 15.1% respectively. The level of heterozygosity within morphotypes was also high in these groups. Jumpers attained good growth by the end of culture but, because of their low survival rate, this approach was not economically feasible. However, higher production and survival in the second-hatched group improved economic viability. Thus, for better results, stocking with later-hatched groups would be more appropriate than stocking with the first-hatched group. [source]


Cold tolerance of the banana prawn Penaeus merguiensis de Man and its growth at different temperatures

Abstract Critical thermal minima (CTMin) were determined for subadults of Penaeus merguiensis acclimated at 15, 18, 21 and 24 °C for 3 weeks. The effect of cooling rate on CTMin was also investigated. The CTMin of prawns from these respective acclimation groups were 5.3, 6.0, 7.4, 9.0 and 4.7, 5.4, 6.0, 7.3 °C at the cooling rate of 1 °C h,1 and 3 °C h,1 respectively. Both acclimation temperature and cooling rate had a significant effect (P < 0.01) on the CTMin. Observation during the acclimation period showed that the growth rate of prawns acclimated at 24 °C was much higher than those acclimated at 21, 18 and 15 °C. The results indicate that P. merguiensis can overwinter in aquaculture ponds in south-east Queensland, Australia, or other subtropical areas and attain good growth if simple overwintering facilities are available.
