Selected AbstractsAn obesity drug sibutramine reduces brain natriuretic peptide (BNP) levels in severely obese patientsINTERNATIONAL JOURNAL OF CLINICAL PRACTICE, Issue 4 2010D. Taner Ertugrul Summary Objectives:, Sibutramine is a selective inhibitor of the reuptake of monoamines. Plasma levels of brain natriuretic peptide (BNP) appear to be inversely associated with body mass index (BMI) in subjects with and without heart failure for reasons that remain unexplained. The aim of this study was to investigate the possible influence of sibutramine treatment on BNP levels in severely obese patients. Methods:, Fifty-two severely obese female patients with BMI > 40 kg/m2 were included to this study. The women were recommended to follow a weight-reducing daily diet of 25 kcal/kg of ideal body weight. During the treatment period, all patients were to receive 15 mg of sibutramine once a day. Blood chemistry tests were performed before the onset of the medication and after 12 weeks of treatment. Results:, None of the subjects was withdrawn from the study because of the adverse effects of sibutramine. Body weight (108.8 ± 13.3 kg vs. 101.7 ± 15.6 kg, p < 0.001), BMI (44.6 ± 4.6 kg/m2 vs. 41.8 ± 5.7 kg/m2, p < 0.001) and BNP [8.6 (0.5,49.5) ng/l vs. 3.1 (0.2,28.6) ng/l, p = 0.018] levels were significantly decreased after 12 weeks of sibutramine treatment. Total cholesterol (5.19 ± 0.90 mmol/l vs. 4.82 ± 1.05 mmol/l respectively; p < 0.001), low-density lipoprotein-cholesterol (3.26 ± 0.86 mmol/l vs. 2.99 ± 0.40 mmol/l respectively; p = 0.008), levels were significantly decreased; however, there was no significant alteration in high-density lipoprotein-cholesterol and triglyceride levels. Conclusion:, This study has shown a decrease in BNP levels which may lead to improvement in cardiac outcome after sibutramine treatment. Further randomised studies are needed to be conducted to clarify the relationship between sibutramine and BNP. [source] Pre-emptive renal transplantation in childrenINTERNATIONAL JOURNAL OF UROLOGY, Issue 5 2001Hiroshi Harada Abstract Background: Renal transplantation is a definitive therapeutic modality in end-stage renal disease (ESRD). Most ESRD patients in Japan experience dialysis prior to renal transplantation. The present study was undertaken to examine the usefulness of pre-emptive renal transplantation (PET). Methods: Between 1987 and 1998, 255 renal transplantations were carried out by the authors. Among those consecutive cases, 10 were cases of PET. In nine pediatric cases, demographics, graft and patient survival, height growth and benefits from successful transplantation were studied and compared with age-matched dialyzed transplantation controls. Results: All transplantation was living-related. There was a disparity of causes of ESRD between the two groups. In PET, acquired renal deterioration due to a congenital lower urinary tract disorder was the major cause. Graft and patient prognosis was favorable in both groups. Growth retardation in PET patients under 15 years of age was significantly less apparent at the time of transplantation and after 3 years compared to the control. The benefits from transplantation were different in the two groups. Most PET patients felt an improvement of their physical condition; however, all of the control patients felt that the major boon was the freedom from the restriction of the daily diet and time for dialysis. Conclusion: In pediatric renal transplantation, short-term preceding dialysis does not have a detrimental effect, but PET could benefit ESRD patients by maintaining their quality of life. Moreover, PET minimizes the production of renal dwarfism in prepubertal children. Thus, PET should be taken into consideration in the choice of renal replacement therapy. [source] Current Opinions on the Functions of Tocopherol Based on the Genetic Manipulation of Tocopherol Biosynthesis in PlantsJOURNAL OF INTEGRATIVE PLANT BIOLOGY, Issue 9 2008Yin Li Abstract As a member of an important group of lipid soluble antioxidants, tocopherols play a paramount role in the daily diet of humans and animals. Recently, genes required for tocochromanol biosynthesis pathway have been identified and cloned with the help of genomics-based approaches and molecular manipulation in the model organisms: Arabidopsis thaliana and Synechocystis sp. PCC 6803. At the basis of these foundations, genetic manipulation of tocochromanol biosynthesis pathway can give rise to strategies that enhance the level of tocochromanol content or convert the constitution of tocochromanol. In addition, genetic manipulations of the tocochromanol biosynthesis pathway provide help for the study of the function of tocopherol in plant systems. The present article summarizes recent advances and pays special attention to the functions of tocopherol in plants. The roles of tocopherol in the network of reactive oxygen species, antioxidants and phytohormones to maintain redox homeostasis and the functions of tocopherol as a signal molecule in chloroplast-to-nucleus signaling to regulate carbohydrate metabolism are also discussed. [source] Proximal composition and in vitro starch digestibility in flaxseed-added corn tortillaJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 3 2009Rodolfo Rendón-Villalobos Abstract BACKGROUND: The effect of addition of flaxseed flour (10:90, 15:85 and 20:80, w/w) on the chemical composition and starch digestibility of corn tortilla was investigated. Tortillas were baked and frozen in liquid nitrogen, freeze-dried, ground and analyzed for fat, protein, ash, total starch (TS), available starch (AS) and resistant starch (RS) contents as well as for starch hydrolysis rate and predicted glycemic index (pGI). Tortillas made from commercial nixtamalized corn flour were used as control sample. RESULTS: Flaxseed flour addition increased the fat and protein content of tortilla, whereas TS and AS decreased. TS was 15.25% lower in the 20% flaxseed-containing tortilla as compared to the control sample. The AS content was 12.65% lower in the composite tortilla. RS content in the samples ranged between 1.92% for the control sample and 5.08% for the tortilla containing 20% flaxseed. The reduced enzymatic starch hydrolysis rate and pGI recorded for the flaxseed-added tortilla, indicated slow digestion features. CONCLUSIONS: Flaxseed-added tortilla might be used to increase the consumption of ,-linolenic acid in the daily diet and modulate starch digestibility of corn tortilla. This kind of product may be used by people with special diet reqirements. Carotenoids are a significant source of provitamin A. The objective of this study was to analyse the carotenoid composition of jackfruit (Artocarpus heterophyllus sinhala: Waraka) kernel using MPLC and visible spectrophotometry and to determine the bioavailability and bioconversion of carotenoids present in jackfruit kernel by monitoring (i) the growth and (ii) levels of retinol and carotenoids in the liver and serum of Wistar rats provided with jackfruit incorporated into a standard daily diet. Carotenoid pigments were extracted using petroleum ether/methanol and saponified using 10% methanolic potassium hydroxide. Six carotenoids were detected in jackfruit kernel. The carotenes ,-carotene, ,-carotene, ,-zeacarotene, ,-zeacarotene and ,-carotene-5,6-epoxide and a dicarboxylic carotenoid, crocetin, were identified, corresponding theoretically to 141.6 retinol equivalents (RE) per 100 g. Our study indicated that jackfruit is a good source of provitamin A carotenoids, though not as good as papaya. Serum retinol concentrations in rats supplemented with jackfruit carotenoids were significantly higher (p = 0.008) compared with the control group. The same was true for liver retinol (p = 0.006). Quantification was carried out by RP-HPLC. These results show that the biological conversion of provitamin A in jackfruit kernel appears satisfactory. Thus increased consumption of ripe jackfruit could be advocated as part of a strategy to prevent and control vitamin A deficiency in Sri Lanka. Copyright © 2004 Society of Chemical Industry [source] Antioxidative and antimutagenic activities and polyphenol content of pesticide-free and organically cultivated green vegetables using water-soluble chitosan as a soil modifier and leaf surface sprayJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 15 2001Huifeng Ren Abstract Five green vegetables (qing-gen-cai, Chinese Cabbage, spinach, Welsh onion and green pepper) commonly used in our daily diet were analysed to determine their antioxidative and antimutagenic activities and chemical content of polyphenols. We obtained pesticide-free and organically cultivated (O) vegetables using water-soluble chitosan as a soil modifier and leaf surface spray (as an alternative natural insecticide) in order to investigate biofunctions induced or enhanced by such specialised cultivation practices. In addition, we purchased the same varieties of vegetables cultivated on an adjacent farm in the conventional manner (C) using pesticides and chemical fertilisers in order to examine the differences in biological activities and distribution of constituents responsible for such activities. The antioxidative activity shown by O vegetables was 120% times higher than that shown by C vegetables in the case of spinach and 20,50% higher in the case of Welsh onion, Chinese cabbage and qing-gen-cai. In comparison with C vegetables, the antimutagenic activity shown by O vegetables was higher against 4-nitroquinoline oxide (4NQO) in qing-gen-cai, Chinese cabbage and Welsh onion, against benzo[a]pyrene (BaP) in all five vegetables, against 2-amino-3-methylimidazo[4,5- f]quinoline (IQ) in qing-gen-cai, Chinese cabbage and green pepper and against 3-amino-1-methyl-5H -pyrido[4,3- b]indole acetate (Trp-P-2) in spinach only. Among all green vegetable juices tested for flavonoid composition, quercitrin, caffeic acid and baicalein in O vegetables were detected in concentrations 1.3,10.4 times higher than those found in C vegetables, suggesting the influence of different cultivation practices. © 2001 Society of Chemical Industry [source] Leucine metabolism in regulation of insulin secretion from pancreatic beta cellsNUTRITION REVIEWS, Issue 5 2010Jichun Yang Leucine, a branched-chain amino acid that must be supplied in the daily diet, plays an important role in controlling protein synthesis and regulating cell metabolism in various cell types. In pancreatic , cells, leucine acutely stimulates insulin secretion by serving as both metabolic fuel and allosteric activator of glutamate dehydrogenase to enhance glutaminolysis. Leucine has also been shown to regulate gene transcription and protein synthesis in pancreatic islet , cells via both mTOR-dependent and -independent pathways at physiological concentrations. Long-term treatment with leucine has been shown to improve insulin secretory dysfunction of human diabetic islets via upregulation of certain key metabolic genes. In vivo, leucine administration improves glycemic control in humans and rodents with type 2 diabetes. This review summarizes and discusses the recent findings regarding the effects of leucine metabolism on pancreatic ,-cell function. [source] Thermoregulatory sympathetic nervous system activity and diet-induced waist-circumference reduction in obese Japanese womenAMERICAN JOURNAL OF HUMAN BIOLOGY, Issue 6 2009Mami Fujibayashi The present study is designed to investigate how and to what extent sympathovagal behavior in a balanced low-calorie diet relates to favorable changes of body mass, waist circumference, and/or metabolic risk factors. The study involved 28 mildly obese women without clinical complications, who underwent an 8-week calorie restriction program using a 1,200-kcal daily diet with an adequate nutrient content; including two regular meals, and one formula meal replacement. All subjects were examined before and after the dietary intervention. We measured anthropometric parameters, blood pressure, and biochemical blood profiles for lipid metabolism. Autonomic nervous system activity was evaluated by heart rate variability power spectral analysis. The dietary intervention induced moderate, but significant reduction of waist circumference (,5.3% ± 0.8%), body fat percentage (,5.8% ± 0.8%), and body mass (,6.6% ± 0.5%). Linear regression analysis showed that ,very low frequency (VLF) power reflecting energy metabolic- and thermoregulatory sympathetic function significantly correlated to ,waist circumference (r = ,0.53, P < 0.01), ,body fat percentage (r = ,0.39, P < 0.05), ,body mass (r = ,0.43, P < 0.05), ,HDL-cholesterol/total cholesterol ratio (HDL-C/TC) (r = 0.62, P < 0.001), and ,nonesterified fatty acids (NEFA) (r = 0.56, P < 0.01). A stepwise multiple regression analysis additionally revealed that ,waist circumference (P = 0.024), ,HDL-C/TC (P = 0.013), and ,NEFA (P = 0.016) were significant and independent factors, which contributing to the variance in ,VLF power (r2 = 0.61). Although causes and consequences of obesity continue to elude researchers, the present study indicates that thermoregulatory sympathetic activity relates to moderate waist-circumference reduction together with favorable changes of blood lipid profiles after short-term dietary modification in mildly obese women. [source] PROTEIN, FIBRE AND BLOOD PRESSURE: POTENTIAL BENEFIT OF LEGUMESCLINICAL AND EXPERIMENTAL PHARMACOLOGY AND PHYSIOLOGY, Issue 4 2008Ya Ping Lee SUMMARY 1Prevention of hypertension and improved blood pressure control can be achieved through dietary modification. In particular, population studies and randomised controlled trials have indicated a beneficial effect of both dietary protein and dietary fibre on level of blood pressure. 2A large population study indicates that an increase in 37 g/day of protein leads to a decrease in mean systolic and diastolic blood pressure by approximately 3 and 2.5 mmHg, respectively. This protective effect is independent of the source of dietary protein. 3Meta-analysis suggests that a fibre increase of approximately 17 g/day will decrease systolic blood pressure by 1.15 mmHg and diastolic blood pressure by 1.65 mmHg, with soluble fibre showing a stronger effect than insoluble fibre. 4Protein and dietary fibre may have additive effects to lower blood pressure. One feasible approach to increasing both protein and fibre in the daily diet could be through the incorporation of legumes, a protein- and fibre-rich food. 5This review assesses the evidence for effects of protein and fibre to reduce blood pressure and the potential of incorporation of legumes into the daily diet as a feasible approach to achieving such benefits for blood pressure. [source] SENSORY DESCRIPTIVE ANALYSIS, SENSORY ACCEPTABILITY AND EXPECTATION STUDIES ON BISCUITS WITH REDUCED ADDED SALT AND INCREASED FIBERJOURNAL OF SENSORY STUDIES, Issue 4 2009MARISA BEATRIZ VÁZQUEZ ABSTRACT The aims of this study were to formulate biscuits with 50% more fiber and 50% less added salt than classic formulations, to describe their sensory characteristics, to measure expectation/sensory acceptability, and to investigate if sensory acceptability for these biscuits was related to the interest in consuming food products with less salt and/or more fiber content. A 2 × 2 factorial design was used to develop four formulations: conventional fiber/conventional salt; conventional fiber/reduced salt; increased fiber/conventional salt; and increased fiber/reduced salt. Differences in the sensory profiles measured by a trained panel were of low magnitude, except for presence and taste of bran. Adolescent and adult consumers evaluated acceptability in three stages: blind with three-digit codes; expectation of the label only; and biscuit + label. The low salt formulations received the lowest scores in the expectation stage, but in the blind and biscuit + label stages acceptability of all formulations was similar. The variables that explained overall acceptance were: measurement stage; formulation salt level; interest in reducing consumption of high salt foods; and interest in consuming bakery products with fiber. PRACTICAL APPLICATIONS Increasing fiber content of biscuits by 40 to 50% helps achieve recommendations to increase fiber intake in daily diets. Also, as biscuits currently on the Argentine and other world markets present two extreme varieties , with or without added salt , formulating a biscuit with 50% less added salt facilitate sodium reduction. In our research we have found that these goals can be achieved without seriously affecting sensory acceptability. We propose the articulation of the necessary strategies with the food industry to market biscuits with less added salt and more fiber for the general population; and the use of these healthier biscuits by institutional food services. Lee ABSTRACT The important roles of reactive oxygen species in diseases related to aging and the necessity and benefits of antioxidative nutraceuticals in the prevention of diseases and promotion of healthy aging have been extensively reported in recent years. Oxygen is an essential component of living organisms. The generation of reactive oxygen species such as superoxide anion, hydrogen peroxide, hydroxyl radicals, and singlet oxygen is inevitable in aerobic metabolism of the body. Reactive oxygen species cause lipid oxidation, protein oxidation, DNA strand break and base modification, and modulation of gene expression. In the past several years, unprecedented progress has been made in the recognition and understanding of roles of reactive oxygen species in many diseases. These include atherosclerosis, vasospasms, cancers, trauma, stroke, asthma, hyperoxia, arthritis, heart attack, age pigments, dermatitis, cataractogenesis, retinal damage, hepatitis, liver injury, and periodontis, which are age-related. The body protects itself from the potential damages of reactive oxygen species. Its first line of defense is superoxide dismutases, glutathione peroxidases, and catalase. Scientists have indicated that antioxidant nutraceuticals supplied from daily diets quench the reactive oxygen species or are required as cofactors for antioxidant enzymes. Nutraceuticals play significant roles in the prevention of a number of age-related diseases and are essential for healthy aging. Epidemiological studies also reported the relevance of antioxidative nutraceuticals to health issues and the prevention of age-related diseases. Health-conscious consumers have made antioxidative nutraceuticals the leading trend in the food industry worldwide in recent years. [source] |
