Selected AbstractsOn functional motor adaptations: from the quantification of motor strategies to the prevention of musculoskeletal disorders in the neck,shoulder regionACTA PHYSIOLOGICA, Issue 2010P. Madeleine Abstract Background:, Occupations characterized by a static low load and by repetitive actions show a high prevalence of work-related musculoskeletal disorders (WMSD) in the neck,shoulder region. Moreover, muscle fatigue and discomfort are reported to play a relevant initiating role in WMSD. Aims: To investigate relationships between altered sensory information, i.e. localized muscle fatigue, discomfort and pain and their associations to changes in motor control patterns. Materials & Methods:, In total 101 subjects participated. Questionnaires, subjective assessments of perceived exertion and pain intensity as well as surface electromyography (SEMG), mechanomyography (MMG), force and kinematics recordings were performed. Results:, Multi-channel SEMG and MMG revealed that the degree of heterogeneity of the trapezius muscle activation increased with fatigue. Further, the spatial organization of trapezius muscle activity changed in a dynamic manner during sustained contraction with acute experimental pain. A graduation of the motor changes in relation to the pain stage (acute, subchronic and chronic) and work experience were also found. The duration of the work task was shorter in presence of acute and chronic pain. Acute pain resulted in decreased activity of the painful muscle while in subchronic and chronic pain, a more static muscle activation was found. Posture and movement changed in the presence of neck,shoulder pain. Larger and smaller sizes of arm and trunk movement variability were respectively found in acute pain and subchronic/chronic pain. The size and structure of kinematics variability decreased also in the region of discomfort. Motor variability was higher in workers with high experience. Moreover, the pattern of activation of the upper trapezius muscle changed when receiving SEMG/MMG biofeedback during computer work. Discussion:, SEMG and MMG changes underlie functional mechanisms for the maintenance of force during fatiguing contraction and acute pain that may lead to the widespread pain seen in WMSD. A lack of harmonious muscle recruitment/derecruitment may play a role in pain transition. Motor behavior changed in shoulder pain conditions underlining that motor variability may play a role in the WMSD development as corroborated by the changes in kinematics variability seen with discomfort. This prognostic hypothesis was further, supported by the increased motor variability among workers with high experience. Conclusion:, Quantitative assessments of the functional motor adaptations can be a way to benchmark the pain status and help to indentify signs indicating WMSD development. Motor variability is an important characteristic in ergonomic situations. Future studies will investigate the potential benefit of inducing motor variability in occupational settings. [source] Ci-GATAa, a GATA -class gene from the ascidian Ciona intestinalis: Isolation and developmental expressionDEVELOPMENTAL DYNAMICS, Issue 1 2003Palmira D'Ambrosio Abstract Members of the GATA family of zinc finger transcription factors have been shown to play important roles in controlling gene expression in a variety of cell types in many metazoan. Here, we describe the identification of Ci-GATAa, a member of this gene family, in the ascidian Ciona intestinalis. Whole-mount in situ hybridization showed that Ci-GATAa was expressed in a highly dynamic manner. The maternal transcript was evenly distributed in the embryo during early stages of development; however, the signal gradually decreased until it disappeared at the 64-cell stage. A zygotic transcript was detected at the 110-cell stage in the blastomeres precursors of three different tissues (brain vesicle, mesenchyme, and trunk lateral cells) and the signal was conserved in these territories up to the larval stage, indicating an important role for Ci-GATAa during ascidian differentiation. © 2002 Wiley-Liss, Inc. [source] Hes7: a bHLH-type repressor gene regulated by Notch and expressed in the presomitic mesodermGENES TO CELLS, Issue 2 2001Yasumasa Bessho Background: Whereas Notch signalling is essential for somitogenesis, mice deficient for the basic helix-loop-helix (bHLH) genes Hes1 and Hes5, downstream Notch effectors, display normal somite formation, indicating that there may be an as-yet unidentified Hes1 -related bHLH gene. Results: We identified a novel bHLH gene, designated Hes7, from mouse embryos. Hes7 has a conserved bHLH domain in the amino-terminal region and the WRPW domain at the carboxy-terminal end, like Hes1. The mouse Hes7 gene is located next to Aloxe3, which is mapped to a position 37.0 cM from the centromere on chromosome 11. In a transfection analysis, Hes7 represses transcription from the N box- and E box-containing promoters. In addition, Hes7 suppresses the E47-induced transcriptional activation. Promoter analysis indicated that Hes7 expression is controlled by Notch signalling. Strikingly, Hes7 is specifically expressed in the presomitic mesoderm in a dynamic manner. We also identified two related bHLH genes from human: one is closely related to mouse Hes7 and therefore designated hHes7 and the other designated hHes4. Conclusion: The structure, transcriptional activity and expression pattern in the presomitic mesoderm of Hes7 are very similar to those of Hes1, suggesting that Hes7, together with Hes1, may play a role in somite formation under the control of Notch signalling. Production of the serine protease inhibitors (serpins) plasminogen activator inhibitor-1 (PAI-1) and protease nexin-1 (PN-1) by astrocytes may counterbalance excessive serine protease activity associated with CNS pathologies such as ischemic stroke. Knowledge regarding the regulation of these genes in the brain is limited, so the objective of the present study was to characterize the effects of injury-related factors on serpin expression in human astrocytes. Native human astrocytes were exposed to hypoxia or cytokines, including interleukin-6 (IL-6), IL-1,, tumor necrosis factor-, (TNF-,), IL-10, transforming growth factor-, (TGF-,), and TGF-, for 0,20 hr. Serpin mRNA expression and protein secretion were determined by real-time RT-PCR and ELISA, respectively. Localization of PAI-1 and PN-1 in human brain tissue was examined by immunohistochemistry. Hypoxia and all assayed cytokines induced a significant increase in PAI-1 expression, whereas prolonged treatment with IL-1, or TNF-, resulted in a significant down-regulation. The most pronounced induction of both PAI-1 and PN-1 was observed following early treatment with TGF-,. In contrast to PAI-1, the PN-1 gene did not respond to hypoxia. Positive immunoreactivity for PAI-1 in human brain tissue was demonstrated in reactive astrocytes within gliotic areas of temporal cortex. We show here that human astrocytes express PAI-1 and PN-1 and demonstrate that this astrocytic expression is regulated in a dynamic manner by injury-related factors. © 2010 Wiley-Liss, Inc. [source] Political Economy of the Asia-Pacific Free Trade Area: A Dilemma for ChinaCHINA AND WORLD ECONOMY, Issue 5 2007Bin Sheng F53; F59; O53 Abstract The Asia-Pacific Free Trade Area (FTAAP) has become a topic of focus since the proposal was first raised in 2004. The present paper considers China's policy towards the FTAAP from a political economy perspective by probing the gains, impediments and concerns for China, and makes judgments based on several possible scenarios. The author argues that from an economic perspective, China would benefit from joining the FTAAP both in a static and a dynamic manner because both its main trade partners and trade barriers in export markets are concentrated in the Asia-Pacific region. However, whether the Chinese Government is likely to support the initiative is largely dependent on certain crucial political and diplomatic elements, including the APEC approach, US-Chinese relations, quality of treaty, sensitive sectors, competitive proposal of alternative and membership of Chinese Taipei. Therefore, if the Chinese Government cannot ratify the ideology and terms of the initiative, or issues that are central to China's interests are not addressed, the FTAAP will only remain a proposal possessing economic possibility in the long run, without political feasibility in the near term.
