Two (3+2) annulation processes allow the synthesis of homochiral 4-alkyl-4-hydroxy-2-cyclopentenones 4,5, 10,11, and 17. The first process involves a domino aza-Claisen/Mannich cyclization reaction, resulting from the alkylation of an ,-alkoxy-enamine, derived from chiral ,-alkoxy aldehydes 1, 9, or 16 with 3-iodo-2-(methoxymethoxy)prop-1-ene (3) as the acetonyl equivalent. The second process is based on the sequential alkylation of esters 21, 39, or nitrile 20 with acetonyl equivalents 3 or 25, followed by an intramolecular Wittig reaction. As an application, the synthesis of the naturally occurring alkylidene-cyclopentenone prostaglandin clavulone II from the spiro[cyclopentene-furan]one 5 and the formal total synthesis of (+/-)-untenone 19 has been carried out. KGaA, 69451 Weinheim, Germany, 2003) [source] Silver-Catalyzed One-Pot Cyclization Reaction of Electron- Deficient Alkynes and 2-Yn-1-ols: An Efficient Domino Process to Polysubstituted FuransADVANCED SYNTHESIS & CATALYSIS (PREVIOUSLY: JOURNAL FUER PRAKTISCHE CHEMIE), Issue 1 2010Hua Cao Abstract Transition metal-catalyzed domino reactions have been used as powerful tools for the preparation of polysubstituted furans in a one-pot manner. In this paper, an efficient synthetic method was developed for the construction of tri- or tetrasubstituted furans from electron-deficient alkynes and 2-yn-1-ols by a silver-catalyzed domino reaction. It is especially noteworthy that a 2,3,5-trisubstituted 4-ynyl-furan was formally obtained in an extremely direct manner without tedious stepwise synthesis. In addition, regio-isomeric furans were observed when substituted aryl alkynyl ketones were employed. This methodology represents a highly efficient synthetic route to electron-deficient furans for which catalytic approaches are scarce. The reaction proceeds efficiently under mild conditions with commercially available catalysts and materials. The key step is the lithiation of the 1,1,-disubstituted ferrocene 11 bearing (S)-2-(methoxymethyl)pyrrolidines as the chiral ortho -directing groups. The diastereoselectivity of these reactions has been mastered by an appropriate choice of the metallating agent, so as to afford a suitable access to C2 -symmetrical, tetrasubstituted ferrocenes. These compounds have been converted into the enantiomerically pure 2-phospha[3]-ferrocenophanes 16, via the corresponding acetates and their reactions with primary phosphines. Phosphines 16 have been used as nucleophilic catalysts in model cyclization reactions. Unlike 2-phospha[3]-ferrocenophanes with stereogenic ,-carbons, the planar chiral derivatives 16 proved to be suitable catalysts for these processes. Thus, for instance, phosphine 16c successfully promotes the enantioselective [3+2],annulations of allenes and enones into functionalized cyclopentenes (ees up to 96%). Among others, spirocyclic derivatives have been obtained in good yields and ees in the range 77,85%. The robustness of this catalyst has been demonstrated by recycling experiments.
