Fillet yield and fat content in common carp (Cyprinus carpio) produced in three Austrian carp farms with different culture methodologies

Summary The fat content and fillet yield of common carp (Cyprinus carpio) from three carp farms were investigated with respect to pond management and overall climatic conditions. The mean fat content of the trimmed fillet varied from 2.7 to 6.9% and differed significantly among the three facilities (P < 0.001). It is suggested that differences in the fillet fat content may be based on the climatic conditions as well as the different management strategies (feeding, stocking biomass). The mean fillet yield was calculated as 34.0,39.0%. No significant correlation between fish weight and fillet yield (, = ,0.16) was found. Results suggest that producing smaller fish for fillets of serving size may lead to a higher production and conclude with lower production risks and costs. [source]


ISTA13-catecholamine toxicity and metabolism in the ciliated protozoan, Tetrahymena pyriformis

Abstract A high throughput culture methodology of unicellular eukaryote Tetrahymena pyriformis, strain GL were used for the determination of catecholamines toxicity and their metabolism. Catecholamines exhibited acute toxicity to Tetrahymena cells where dopamine and L -DOPA showed higher toxic potential of EC10 (0.39 and 0.63 mg/L, respectively) and EC20 (1.1 and 1.0 mg/L, respectively). All the testing catecholamines were highly degradable in the PPY-medium due to the oxidizing environment during incubation. They were also naturally synthesized and released by Tetrahymena cells into the culture medium and increasingly accumulated with time where as noradrenalin demonstrated significant results. Cells were exposed with physiological concentration (0.12 mg/L) and one higher concentration (8.0 mg/L) of catecholamines, resulting noradrenalin depletion and in vivo generation of a metabolite in response to dopamine with higher concentration treatment. This dopamine metabolite was relatively nonpolar compared with the catecholamines and was eluted later from the reverse phase C-18 column. © 2008 Wiley Periodicals, Inc. Environ Toxicol, 2009. Evidence of occult HCV genotypes in haemophilic individuals with unapparent HCV mixed infections

Summary., Individuals with haemophilia who received non heat-treated factor concentrates were likely to undergo multiple exposures to the hepatitis C virus (HCV). Therefore, HCV mixed-genotype infections might be more frequent in these patients than in the general population. Their prevalence is extremely variable in similar groups of patients tested by different assays due to the fact that currently available genotyping techniques are not suitable to detect multiple HCV genotypes in a viral population. As an HCV viral reservoir, the peripheral blood mononuclear cell (PBMC) might harbor viral variants distinct from the genotypes detected in plasma. We investigated the presence of HCV genotypes in a group of chronically infected haemophilic patients in the PBMC compartment using a non-stimulated cell culture system that allows the detection of the HCV genome in culture supernatants. We compared them to the HCV genotypes found in plasma samples. Cell culture experiments performed with PBMC demonstrated the presence of additional HCV genotypes that were undetected in the corresponding plasma samples with the same genotyping technique. Although mixed infections at HCV genotype level became evident in 5.6% of the patients (16/288), the culture methodology increased the number of HCV infections with multiple genotypes to 62.5% (10/16) (P < 0.0001). Once more, the role of mononuclear cells as HCV viral reservoirs is emphasized. Considering minor strains could influence the outcome of treatment, detection of covert HCV mixed-genotype infections might be essential for choosing the adequate therapeutic regimen. [source]


The intensity of the 1602 cm,1 band in human cells is related to mitochondrial activity

Abstract We report a Raman band at 1602 cm,1 in the spectra of human cells, which previously had only been observed in mitochondria of yeast cells. This band, which has not yet been assigned to a particular molecular species, was found to occur in HeLa cells, peripheral blood lymphocytes, human mesenchymal stem cells and bovine chondrocytes. The band is proposed as an indicator of the activity of mitochondria in cells. Cells were cultured with and without serum or temporarily deprived of serum. The band can be observed for all these variations in cell culture methodology. The band intensity decreases under the influence of an increase of the calcium ion concentration in the surrounding medium. Copyright © 2009 John Wiley & Sons, Ltd. [source]
