Regional differences in kelp-associated algal assemblages on temperate limestone reefs in south-western Australia

Abstract.,Ecklonia radiata (C. Agardh) J. Agardh kelp beds , a characteristic feature of the nearshore environment along the south-west Australian coastline , contribute significantly to the coastal biodiversity in temperate Australia, yet, little is known about the organization of these macroalgal assemblages. By compiling existing and new data sets from habitat surveys, we have characterized and compared the structure of kelp-associated macroalgal assemblages in three regions (Marmion Lagoon, Hamelin Bay and the marine environment neighbouring the Fitzgerald River National Park) across more than 1000 kilometres of the south-west Australian coastline. 152 macroalgal taxa had been recognized within the three regions and this is in the range of species richness reported from other Australian and African kelp beds. The kelp-associated algal assemblages were regionally distinct, 66% of all taxa were only found in one region and only 17 taxa were found in all three regions. Adjacent regions shared an additional 13,15 taxa. The regional shifts in assemblage structure were evident in species composition of both canopy and understorey. The organization of assemblages followed a spatial hierarchy where differences in assemblage structure were larger among regions (hundreds of kilometres apart) than among sites within regions (kilometres apart) and differences among sites within region were larger than differences among quadrats within sites (metres apart). Despite this hierarchy each level of nesting contributed approximately the same to total variation in assemblage structure and these spatial patterns were stronger than temporal differences from seasons to 2,3 years. Our results suggest that local and small-scale processes contribute considerably to heterogeneity in macroalgal assemblages throughout south-western Australia, and, in particular, our results are consistent with E. radiata exerting a strong influence on macroalgal assemblage structure. Further, our study contradicts the existence of a general south-west Australian kelp assemblage, although a few species may form the core of E. radiata associations across regions. [source]


Loss of steroidogenic factor 1 alters cellular topography in the mouse ventromedial nucleus of the hypothalamus

Abstract Knockout (KO) mice lacking the orphan nuclear receptor steroidogenic factor 1 (SF-1) exhibit marked structural abnormalities of the ventromedial nucleus of the hypothalamus (VMH). In this study, we sought to determine the molecular mechanisms underlying the VMH abnormalities. To trace SF-1-expressing neurons, we used a SF-1/enhanced green fluorescent protein (eGFP) transgene. Although the total numbers of eGFP-positive cells in wild-type (WT) and SF-1 KO mice were indistinguishable, cells that normally localize precisely within the VMH were scattered more diffusely in adjacent regions in SF-1 KO mice. This abnormal distribution is likely due to the loss of SF-1 expression in VMH neurons rather than secondary effects of deficient steroidogenesis, as redistribution also was seen in mice with a CNS-specific KO of SF-1. Thus, the absence of SF-1 alters the distribution of cells that normally form the VMH within the mediobasal hypothalamus. Consistent with this model, the hypothalamic expression patterns of the transcription factors islet-1 and nkx2.1 also were displaced in SF-1 KO mice. Independent of gene expression, birthdate analyses further suggested that cells with earlier birthdates were affected more severely by the loss of SF-1 than were later born cells. We conclude that the absence of SF-1 causes major changes in cellular arrangement within and around the developing VMH that result from altered cell migration. © 2004 Wiley Periodicals, Inc. Phytogeographical evidence for post-glacial dispersal limitation of European beech forest species

The post-glacial migration of European beech Fagus sylvatica has been addressed by many studies using either genetic or fossil data or a combination of both. In contrast to this, only little is known about the migration history of beech forest understorey species. In a review of phytosociological literature, we identified 110 plant species which are closely associated with beech forest. We divided the distribution range of European beech forests into 40 geographical regions, and the presence or absence of each species was recorded for each region. We compared overall species numbers per region and numbers of narrow-range species (species present in <10 regions). A multiple regression model was used to test for the explanatory value of three potential diversity controls: range in elevation, soil type diversity, and distance to the nearest potential refuge area. A hierarchical cluster analysis of the narrow-range species was performed. The frequency of range sizes shows a U-shaped distribution, with 42 species occurring in <10 regions. The highest number of beech forest species is found in the southern Alps and adjacent regions, and species numbers decrease with increasing distance from these regions. With only narrow-range species taken into consideration, secondary maxima are found in Spain, the southern Apennines, the Carpathians, and Greece. Distance to the nearest potential refuge area is the strongest predictor of beech forest species richness, while altitudinal range and soil type diversity had little or no predictive value. The clusters of narrow-range species are in good concordance with the glacial refuge areas of beech and other temperate tree species as estimated in recent studies. These findings support the hypothesis that the distribution of many beech forest species is limited by post-glacial dispersal rather than by their environmental requirements. [source]


Local and regional-scale responses of ant diversity to a semiarid biome transition

The locations of biome transitions and ecotones are frequently defined by the rapid shift from one form of dominant vegetation to another. The composition of animal taxa is predicted to shift in parallel with that of dominant plants and species diversity is predicted to he greater in transitional zones than in adjacent areas. We asked whether ant species diversity and composition supported these predictions across a biome transition between shortgrass steppe and Chihuahuan desert vegetation. Neither species richness nor diversity was highest at the biome transition region as a whole, or within habitats in the biome transition. The biome transition region was not intermediate in ant species composition or in the representation of different faunal complexes. The community similarity between matched habitats shared between the biome transition zone and adjacent regions was less than that between distinct habitats occurring within regions. A zoogeographic transition for ants may occur to the north of the phytogeographic transition and may be coincident with the northern limits of monsoonal precipitation patterns. In contrast, the phytogeographic transition may be related to less extreme climatic variation within the monsoonal region occurring further south. [source]


The neuroanatomy of grapheme,color synesthesia

Abstract Grapheme,color synesthetes perceive particular colors when seeing a letter, word or number (grapheme). Functional neuroimaging studies have provided some evidence in favor of a neural basis for this type of synesthesia. Most of these studies have reported extra activations in the fusiform gyrus, which is known to be involved in color, letter and word processing. The present study examined different neuroanatomical features (i.e. cortical thickness, cortical volume and cortical surface area) in a sample of 48 subjects (24 grapheme,color synesthetes and 24 control subjects), and revealed increased cortical thickness, volume and surface area in the right and left fusiform gyrus and in adjacent regions, such as the lingual gyrus and the calcarine cortex, in grapheme,color synesthetes. In addition, we set out to analyze structural connectivity based on fractional anisotropy (FA) measurements in a subsample of 28 subjects (14 synesthetes and 14 control subjects). In contrast to the findings of a recent neuroanatomical study using modern diffusion tensor imaging measurement techniques, we did not detect any statistically significant difference in FA between synesthetes and non-synesthetes in the fusiform gyri. Our study thus supports the hypothesis of local anatomical differences in cortical characteristics in the vicinity of the V4 complex. The observed altered brain anatomy in grapheme,color synesthetes might be the anatomical basis for this particular form of synesthesia but it is also possible that the detected effects are a consequence (rather than the primary cause) of the life-long experience of grapheme,color synesthesia. Spontaneous recurrent network activity in organotypic rat hippocampal slices

Abstract Organotypic hippocampal slices were prepared from postnatal day 4 rats and maintained in culture for >6 weeks. Cultured slices exhibited from 12 days in vitro spontaneous events which closely resembled giant depolarizing potentials (GDPs) recorded in neonatal hippocampal slices. GDP-like events occurred over the entire hippocampus with a delay of 30,60 ms between two adjacent regions as demonstrated by pair recordings from CA3,CA3, CA3,CA1 and interneurone,CA3 pyramidal cells. As in acute slices, spontaneous recurrent events were generated by the interplay of GABA and glutamate acting on AMPA receptors as they were reversibly blocked by bicuculline and 6,7-dinitroquinoxaline-2,3-dione but not by dl -2-amino-5-phosphonopentaoic acid. The equilibrium potentials for GABA measured in whole cell and gramicidin-perforated patch from interconnected interneurones,CA3 pyramidal cells were ,70 and ,56 mV, respectively. The resting membrane potential estimated from the reversal of N -methyl- d -aspartate-induced single-channel currents in cell-attach experiments was ,75 mV. In spite of its depolarizing action, in the majority of cases GABA was still inhibitory as it blocked the firing of principal cells. The increased level of glutamatergic connectivity certainly contributed to network synchronization and to the development of interictal discharges after prolonged exposure to bicuculline. In spite of its inhibitory action, in a minority of cells GABA was still depolarizing and excitatory as it was able to bring principal cells to fire, suggesting that a certain degree of immaturity is still present in cultured slices. This was in line with the transient bicuculline-induced block of GDPs and with the isoguvacine-induced increase of GDP frequency. [source]


An efficient methodology for security assessment of power systems based on distributed optimal power flow

This paper presents an algorithm for the parallel solution of the security constrained optimal power flow (SCOPF) problem in a decentralized framework, consisting of regions, using a price-based mechanism that models each region as an economic unit. We first solve the distributed optimal power flow (OPF) problem to determine the maximum secure simultaneous transfer capability of each tie-line between adjacent regions by taking only the security constraints imposed on the tie-lines into account. In this paper, the line outage distribution factors (LODF) calculated at the current state are used to formulate the appended constraints. Once the secure transfer capability of each tie-line is determined, the intra-regional SCOPF is performed using the conventional linear programming (LP) approach. A description on the inclusion of security constraints with distributed OPF algorithm will be given, followed by the case study for Korea Electric Power System. [source]


Molecular physiology of SLC4 anion exchangers

Plasmalemmal Cl,,HCO3, exchangers regulate intracellular pH and [Cl,] and cell volume. In polarized epithelial cells, they contribute also to transepithelial secretion and reabsorption of acid,base equivalents and of Cl,. Members of both the SLC4 and SLC26 mammalian gene families encode Na+ -independent Cl,,HCO3, exchangers. Human SLC4A1/AE1 mutations cause either the erythroid disorders spherocytic haemolytic anaemia or ovalocytosis, or distal renal tubular acidosis. SLC4A2/AE2 knockout mice die at weaning. Human SLC4A3/AE3 polymorphisms have been associated with seizure disorder. Although mammalian SLC4/AE polypeptides mediate only electroneutral Cl,,anion exchange, trout erythroid AE1 also promotes osmolyte transport and increased anion conductance. Mouse AE1 is required for DIDS-sensitive erythroid Cl, conductance, but definitive evidence for mediation of Cl, conductance is lacking. However, a single missense mutation allows AE1 to mediate both electrogenic SO42,,Cl, exchange or electroneutral, H+ -independent SO42,,SO42, exchange. In the Xenopus oocyte, the AE1 C-terminal cytoplasmic tail residues reported to bind carbonic anhydrase II are dispensable for Cl,,Cl, exchange, but required for Cl,,HCO3, exchange. AE2 is acutely and independently inhibited by intracellular and extracellular H+, and this regulation requires integrity of the most highly conserved sequence of the AE2 N-terminal cytoplasmic domain. Individual missense mutations within this and adjacent regions identify additional residues which acid-shift pHo sensitivity. These regions together are modelled to form contiguous surface patches on the AE2 cytoplasmic domain. In contrast, the N-terminal variant AE2c polypeptide exhibits an alkaline-shifted pHo sensitivity, as do certain transmembrane domain His mutants. AE2-mediated anion exchange is also stimulated by ammonium and by hypertonicity by a mechanism sensitive to inhibition by chelation of intracellular Ca2+ and by calmidazolium. This growing body of structure,function data, together with increased structural information, will advance mechanistic understanding of SLC4 anion exchangers. [source]


An Approximation for the Rank Adjacency Statistic for Spatial Clustering with Sparse Data

The rank adjacency statistic D provides a simple method to assess regional clustering. It is defined as the weighted average absolute difference in ranks of the data, taken over all possible pairs of adjacent regions. In this paper the usual normal approximation to the D statistic is found to give inaccurate results if the data are sparse and some regions have tied ranks. Adjusted formulae for the moments of D that allow for the existence of ties are derived. An example of analyses of sparse mortality data (with many regions having no deaths, and hence tied ranks) showed satisfactory agreement between the adjusted formulae and the empirical distribution of the D statistic. We conclude that the D statistic, when used with adjusted moments, provides a valid approximate method to evaluate spatial clustering, even in sparse data situations. [source]


A preliminary study of crustal structure in Taiwan region using receiver function analysis

SUMMARY Selected teleseismic data observed at temporary and permanent broad-band stations have been analysed using the receiver function method in order to investigate the very complex crustal structure in Taiwan region. Very significant azimuthal variations of radial and transverse receiver function responses from broad-band stations could be attributed to, among other things, the sampling of incoming seismic waves across the nearby subduction zone, a subsurface dipping interface, or a localized anisotropic region. A mid-crust discontinuity, interpreted as the Conrad discontinuity, can be identified at 18,20 km depth beneath TATO and TPUB stations in the Western Foothills, but is absent beneath the two nearby stations SSLB and TDCB in the Central Mountain Range. The separation of upper and lower crust beneath the Western Foothills and the steady increase in crustal velocity as a function of depth across the entire thicker crust beneath the Central Mountain Range suggest that the tectonic evolution of the crust may be significantly different for these two adjacent regions. Although a ,thin-skinned' model may be associated with the tectonic evolution of the upper crust of the Western Foothills and Western Coastal Plain, a ,thick-skinned' or ,lithospheric deformation' model can probably be applied to explain the crustal evolution of the Central Mountain Range. A trend of crustal thinning from east (50,52 km) to west (28,32 km) is in very good agreement with the results from two east,west-trending deep seismic profiles obtained using airgun sources. The thinner crust (20,30 km) beneath TWB1 station in northeastern Taiwan can be associated with the high-heat-flow backarc opening at the western terminus of the Okinawa trough behind the subduction of the Philippine Sea plate. The relatively simple crustal structure beneath KMNB station, offshore southeastern China, depicts typical continental crust, with the Moho depth at 28,32 km. An apparent offset of the thickest Moho beneath NACB station from the topographic high in the central Central Mountain Range suggests that the Taiwan orogeny has probably not reached its isostatic status. Spatial Effects in Website Adoption by Firms in European Regions

ABSTRACT The purpose of this paper is to provide empirical evidence on the neighboring effects of Internet adoption as measured by the percentage of firms with their own website in the European regions. This is the first study that explicitly analyzes the role played by spatial effects to explain website adoption for the European case. A set of instruments and techniques commonly used in the spatial econometrics framework is employed to test the hypothesis that proximity matters when explaining Internet adoption by firms. Results show that firms in physically adjacent regions register a similar degree of Internet adoption, confirming the presence in this context of positive spatial dependence. Nevertheless, the spatial effects detected are mainly constrained by national borders. Gross domestic product (GDP) per capita, population density, sectoral composition, and education are positively related to geographic distribution of Internet adoption in the enlarged European Union. In addition, regional disparities in Internet adoption were found to be less important than territorial inequalities in GDP per capita. [source]


Temporal Elements in the Spatial Extension of Production Networks

ABSTRACT The spatial extension of production networks presents a significant challenge to managers accustomed to reducing lead times by geographically contracting supply chains. This paper extends the theory on time in transportation by defining the elements of transport time, order time, timing, punctuality, and frequency and elaborating on their characteristics. Structured along these elements, it analyses the consequences of extending production networks from within a mature economic region, mainly the EU-15, U.S., and Japan, first to adjacent and then to nearby and finally distant low-cost regions. Distance obviously affects the transport quality in all time dimensions. Except for air parcel services that globally match what road transport offers within an economic region, the longer the distance, the lower the time-related performance. Distant, low-cost regions, meaning China and India, also imply a polarisation between air and sea transport at opposite ends of the time, cost, and capacity scales. This supply gap restricts the types of products traded. The conceptual framework is illustrated in the setting of a global vehicle manufacturer spatially extending its sourcing. It demands that sequenced sub-assemblies and small, cheap, and generic components are delivered from the vicinity of each assembly plant. Batched components can be sourced from adjacent regions, but deliveries from longer distances imply storage at pick-up points to fulfil their time requirements. Hence, the suppliers must offer the manufacturing firm deliveries as if they produce relatively close to the assembly plants. [source]


CO2 laser surgery in the treatment of glottic cancer

Abstract Background. The aim of the study was to assess the effectiveness of CO2 laser endoscopic surgery in the treatment of glottic carcinoma limited to the true vocal cords or involving the adjacent regions. Methods. Seven hundred nineteen patients (687 men and 32 women; mean age, 60.4 years; range, 33,86 years) with glottic carcinoma (432 T1N0M0, 236 T2N0M0, 51 T3N0M0) underwent CO2 laser surgery (mean follow-up, 5 years; range, 2,17 years). Statistical comparison was carried out with Wilcoxon test, considering p < .05 the minimum significance value. Results. Overall actuarial survival, adjusted actuarial survival, and percentage of patients with no evidence of disease at 5 years were 85%, 97%, and 85%, respectively, in patients with T1a disease; 84%, 96%, and 83% in those with T1b disease; 77%, 86%, and 61% in those with T2 unilateral tumors; 77%, 88%, and 55% in those with T2 bilateral tumors; and 64%, 72%, and 60% in those with T3 disease. The statistical analysis showed the following: significant differences in the comparison of T1 versus T2 and T2 versus T3 tumors (p < .01), with the exception of no evidence of disease in the comparison of T2 versus T3 (p > .05); and no significant differences in the comparison of unilateral and bilateral tumors (p > .05). Actuarial local control, actuarial nodal control, and actuarial distant metastasis control at 5 years were 85%, 98%, and 99%, respectively, in patients with T1 disease; and 66%, 82%, and 91% in patients with T2 disease; and 66%, 83%, and 95% in patients with T3 disease. The laryngeal preservation rate was 97.3% in the T1 group, 82.5% in the T2 group, and 80.5% in T3 group. Conclusions. CO2 laser endoscopic surgery is effective in the treatment of glottic carcinoma not infiltrating the cartilaginous skeleton; the results achieved are competitive with those of open conservative operations, if we take into account the possibilities afforded by salvage surgery and the rate of laryngeal preservation achieved in the study patients. © 2004 Wiley Periodicals, Inc. 3D Mapping of brain differences in native signing congenitally and prelingually deaf subjects

Abstract In the prelingual and congenital deaf, functional reorganization is known to occur throughout brain regions normally associated with hearing. However, the anatomical correlates of these changes are not yet well understood. Here, we perform the first tensor-based morphometric analysis of voxel-wise volumetric differences in native signing prelingual and congenitally deaf subjects when compared with hearing controls. We obtained T1-weighted scans for 14 native signing prelingual and congenitally deaf subjects and 16 age- and gender-matched controls. We used linear and fluid registration to align each image to a common template. Using the voxel-wise determinant of the Jacobian of the fluid deformation, significant volume increases, of up to 20%, were found in frontal lobe white matter regions including Broca's area, and adjacent regions involved in motor control and language production. A similar analysis was performed on hand-traced corpora callosa. A strong trend for group differences was found in the area of the splenium considered to carry fibers connecting the temporal (and occipital) lobes. These anatomical differences may reflect experience-mediated developmental differences in myelination and cortical maturation associated with prolonged monomodal sensory deprivation. Southeast Alaska: oceanographic habitats and linkages

Abstract We provide an overview of the physical oceanographic and geological processes that affect marine biological habitats and production in the marine waters throughout the archipelago and continental shelf of Southeast Alaska. Given the paucity of regional data, our overview summarizes work done in adjacent regions of the Gulf of Alaska shelf and basin, and draws on research carried out in similar settings elsewhere. The geological setting, which critically influences the regional meteorology and oceanography, includes a narrow continental shelf, deep channels that permeate the archipelago, fjords, glaciers and a rugged, mountainous coast. The large-scale meteorology is influenced primarily by seasonal variations in the intensity and position of the Aleutian Low. Winds, freshwater runoff, tides and cross-shelf exchange control the regional oceanography. The large-scale flow field advects mass, heat, salt, nutrients and planktonic organisms northward from British Columbia (and even further south) to the northern Gulf of Alaska along the slope, shelf, and within the channels of Southeast Alaska. The deep channels permeating the island archipelago and narrow continental shelf facilitate communication between basin and interior waters. Water properties and flow fields are subject to large annual variations in response to similarly large variations in winds and coastal freshwater discharge. The complex geological setting leads to large spatial heterogeneity in the physical processes controlling the local circulation fields and mixing, thereby creating numerous and diverse marine biological habitats. These various circulation and mixing processes modify substantially Southeast Alaska water masses and thus influence marine ecosystem processes downstream over the northern and western Gulf of Alaska shelf. Timing and nature of late Quaternary mountain glaciation,

Abstract Compilations of regional- to subcontinental-scale mountain glacier chronologies from five continents, plus Hawaii, examine the temporal and spatial patterns of mountain glaciation. The compilations yield key insights into the nature of past fluctuations in temperature and precipitation in mountains and adjacent regions, and provide baseline data for examining the dynamics of glacier responses to climate change. Key insights from these compilations include the wide variability of glacier responses to fluctuations in insolation, temperature and precipitation and the regionally specific variations in climatic variables through late Quaternary time. The compilations highlight the need to improve the density and quality of geochronological data and to enhance the understanding of the links between climate and glaciation. Fabrication of a novel micron scale Y-structure-based chiral metamaterial: Simulation and experimental analysis of its chiral and negative index properties in the terahertz and microwave regimes

Abstract In this report, we describe the fabrication of a chiral metamaterial based on a periodic array of Y-shaped Al structures on a dielectric Mylar substrate. The unit cell dimensions of the Y-structure are ,100 ,m on a side with 8 ,m linewidths. The fabricated Y-structure elements are characterized using scanning electron microscopy (SEM) and atomic force microscopy (AFM). Quantitative elemental analyses were carried out on both the Y-structure, comprised of Al and its oxide, as well as adjacent regions of the underlying mylar substrate using the energy dispersive X-ray spectroscopy (EDS) capability of the SEM. Finite-Difference Time-Domain (FDTD) calculations of the negative index of refraction for a 3D wedge of multiple layers of the 2D metamaterials showed that these metamaterials possess double negative (,,,,,) electromagnetic bulk properties at THz frequencies. The same negative index of refraction was determined for a wedge comprised of appropriately scaled larger Y-structures simulated in the microwave region. This double negative property was confirmed experimentally by microwave measurements on a 3D wedge comprised of stacked and registered Y-structure sheets. Microsc. Res. Abstract Patterns of genetic variation within a species may be a consequence of historical factors, such as past fragmentation, as well as current barriers to gene flow. Using sequence data from the mitochondrial cytochrome oxidase subunit II region (COII) and the nuclear gene wingless, we conducted a phylogeographical study of the holarctic skipper Hesperia comma to elucidate patterns of genetic diversity and to infer historical and contemporary processes maintaining genetic variation. One hundred and fifty-one individuals were sampled from throughout North America and Eurasia, focusing on California and adjacent regions in the western United States where morphological diversity is highest compared to the rest of the range. Analyses of sequence data obtained from both genes revealed a well-supported division between the Old and New World. Within western North America, wingless shows little geographical structure, while a hierarchical analysis of genetic diversity of COII sequences indicates three major clades: a western clade in Oregon and Northern California, an eastern clade including the Great Basin, Rocky Mountains and British Columbia, and a third clade in southern California. The Sierra Nevada and the Transverse Ranges appear to be the major barriers to gene flow for H. comma in the western United States. Relatively reduced haplotype diversity in Eurasia compared to North America suggests that populations on the two continents have been affected by different historical processes. [source]


Role of the N-terminal Region in the Function of the Photosynthetic Bacterium Transcription Regulator PpsR,

PpsR is a transcription repressor for the gene cluster encoding photosystem genes in Rhodobacter sphaeroides. Repression activity is accomplished by DNA binding on the promoter regions of the photosystem gene clusters, and depends on both the redox potential and the presence of antirepressor protein AppA. To understand DNA repression regulation by PpsR, we investigated the function of PpsR domains in self-association for DNA binding. We constructed domain-deletion mutants and verified DNA-binding activity and dimer formation. Gel shift assay for measuring the DNA-binding activity of three sequential N-terminal deletion mutants revealed that N-terminal deletions (of minimum 121 residues) caused loss of binding activity. Size-exclusion gel chromatography revealed that deletion mutant which lacks the N-terminal 121-amino acid deletion mutant to exist as a dimer, although it was less stable than the intact PpsR. The mutants lacking the adjacent regions, Q-linker region and the first Per-Ant-Sim domain, did not form dimers, suggesting the involvement of the N-terminal region in dimer formation. This region is thus considered to be a functional domain in self-association, although not yet identified as a structural domain. Circular dichroism spectrum of the N-terminal region fragment exhibited a ,/, structure. We conclude that this region is a structural and functional domain, contributing to PpsR repression through dimer stabilization. [source]


Structure of murine angiogenin: features of the substrate- and cell-binding regions and prospects for inhibitor-binding studies

Angiogenin is an unusual member of the pancreatic ribonuclease superfamily that induces blood-vessel formation and is a promising anticancer target. The three-dimensional structure of murine angiogenin (mAng) has been determined by X-ray crystallography. Two structures are presented: one is a complex with sulfate ions (1.5,Å resolution) and the other a complex with phosphate ions (1.6,Å resolution). Residues forming the putative B1, P1 and B2 subsites occupy positions similar to their hAng counterparts and are likely to play similar roles. The anions occupy the P1 subsite, sulfate binding conventionally and phosphate adopting two orientations, one of which is novel. The B1 subsite is obstructed by Glu116 and Phe119, with the latter assuming a less invasive position than its hAng counterpart. Hydrophobic interactions between the C-terminal segment and the main body of the protein are more extensive than in hAng and may underly the lower enzymatic activity of the murine protein. Elsewhere, the structure of the H3,B2 loop supports the view that hAng Asn61 interacts directly with cell-surface molecules and does not merely stabilize adjacent regions of the hAng structure. mAng crystals appear to offer small-molecule inhibitors a clear route to the active site and may even withstand a reorientation of the C-terminal segment that provides access to the cryptic B1 subsite. These features represent considerable advantages over crystalline hAng and bAng. [source]


Island-like radiation of Saussurea (Asteraceae: Cardueae) triggered by uplifts of the Qinghai,Tibetan Plateau

Increasing evidence suggests that geological or climatic events in the past triggered the radiative diversification of both animals and plants on islands as well as continents. The Qinghai,Tibetan Plateau (QTP) has been extensively uplifted since the Miocene, but there is little information on possible links between these events and biological diversification in this and adjacent regions. Partly to explore such links, we have examined the diversification of Saussurea (Asteraceae: Cardueae), a species-rich genus that is mostly endemic to QTP, but also occurs in arid highlands elsewhere in the Northern Hemisphere. The phylogenetic analyses were conducted on the basis of the nuclear (internal transcribed spacer, ITS) and plastid (trnL-F and psbA-trnH) sequences from 55 species, representing 19 sections from all six subgenera of Saussurea, and species from 15 genera of the Cardueae. The results suggest that the currently circumscribed genus Saussurea (s.l.) is a polyphyletic group and that five sections should be excluded from the genus. Samples from the other 14 sections (representing five subgenera) clustered as a monophyletic group (here designated the Saussurea s.s. lineage, SSSL) with high statistical support. However, none of the analyses (nuclear, plastid or combined) resolved SSSL's infrageneric phylogeny, and the parallel clades of the lineage indicate that island-like adaptive radiation occurred. Furthermore, this radiation appears to have occurred 14,7 Mya, during the period of the major uplift events of QTP.
