Selected AbstractsA multilayer poly(dimethylsiloxane) electrospray ionization emitter for sample injection and online mass spectrometric detectionELECTROPHORESIS, Issue 24 2005Jamie M. Iannacone Abstract An ESI emitter made of poly(dimethylsiloxane) interfaces on-chip sample preparation with MS detection. The unique multilayer design allows both the analyte and the spray solutions to reside on the device simultaneously in discrete microfluidic environments that are spatially separated by a polycarbonate track-etched, nanocapillary array membrane (NCAM). In direct spray mode, voltage is applied to the microchannel containing a spray solution delivered via a syringe pump. For injection, the spray potential is lowered and a voltage is applied that forward biases the membrane and permits the analyte to enter the spray channel. Once the injection is complete, the bias potential is switched off, and the spray voltage is increased to generate the ESI of the injected analyte plug. Consecutive injections of a 10,,M bovine insulin solution are reproducible and produce sample plugs with limited band broadening and high quality mass spectra. Peptide signals are observed following transport through the NCAM, even when the peptide is dissolved in solutions containing up to 20% seawater. The multilayer emitter shows great potential for performing multidimensional chemical manipulations on-chip, followed by direct ESI with negligible dead volume for online MS analysis. [source] A Novel Cyclic Squamosamide Analogue Compound FLZ Improves Memory Impairment in Artificial Senescence Mice Induced by Chronic Injection of D-Galactose and NaNO2BASIC AND CLINICAL PHARMACOLOGY & TOXICOLOGY, Issue 6 2007Fang Fang Artificially senescent mouse model was induced by consecutive injection of D-galactose (120 mg/kg) and NaNO2 (90 mg/kg) once daily for 60 days. Compound FLZ (75 and 150 mg/kg) was orally administered once daily for 30 days after D-galactose and NaNO2 injection for 30 days. The water maze test was used to evaluate the learning and memory function of mice. The content of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in serum were determined using different biochemical kits. The alterations in hippocampus morphology were assessed by light and electronic microscope. Immunoreactive cells of Bcl-2 in the hippocampus were counted by immunohistochemical staining, and Bcl-2 protein expression was analysed by Western blot method. The results indicate that injection of D-galactose and NaNO2 induces memory impairment and neuronal damage in hippocampus of mice. In addition, serum SOD and GSH-Px activities decreased, while MDA level increased. Bcl-2-positive neurons and Bcl-2 protein expression in the hippocampus decreased remarkably. Oral administration of FLZ for 30 days significantly improved the cognitive deficits and the biochemical markers mentioned above, and also reduced the pathological alterations in mouse hippocampus. The results suggest that FLZ ameliorates memory deficits and pathological injury in artificially senescent mice induced by chronic injection of D-galactose and NaNO2, indicating that FLZ is worth further studies for fighting antisenescence and dementia. [source] Quantitative capillary electrophoresis and its application in analysis of alkaloids in tea, coffee, coca cola, and theophylline tabletsJOURNAL OF SEPARATION SCIENCE, JSS, Issue 2 2009Mengjia Li Abstract A quantitative CE (qCE) system with high precision has been developed, in which a 4-port nano-valve was isolated from the electric field and served as sample injector. The accurate amount of sample was introduced into the CE system with high reproducibility. Based on this system, consecutive injections and separations were performed without voltage interruption. Reproducibilities in terms of RSD lower than 0.8% for retention time and 1.7% for peak area were achieved. The effectiveness of the system was demonstrated by the quantitative analysis of caffeine, theobromine, and theophylline in real samples, such as tea leaf, roasted coffee, coca cola, and theophylline tablets. [source] Single run measurements of drug-protein binding by high-performance frontal analysis capillary electrophoresis and mass spectrometryRAPID COMMUNICATIONS IN MASS SPECTROMETRY, Issue 12 2005Hong Wan A novel drug-protein binding measurement method based on high-performance frontal analysis and capillary electrophoresis (HPFA/CE) is presented. A single run measurement approach is proposed to circumvent utilization of a calibration curve that is often performed with HPFA. A sensitive mass spectrometer is applied as a detector enabling the measurement of in vitro protein binding at lower drug concentrations. Unbound free fraction and binding constants can be determined by a single run measurement by consecutive injections of an internal drug standard, a buffer plug and a drug-protein mixture. Effects of injection volumes on peak height and plateau profile were investigated in two different separation systems, non-volatile buffer and volatile buffer, with UV and mass spectrometry detection, respectively. A simplified one-to-one binding model is employed to evaluate the proposed method by using both single and multiple drug concentrations to measure the unbound free fraction and calculate the binding constants of some selected compounds. The method is suitable for rapid and direct screening of the binding of a drug to a specific protein or drug-plasma protein binding. Copyright © 2005 John Wiley & Sons, Ltd. [source] Effects of misty-fan cooling and supplemental rbST on rumen function and milk production of crossbred Holstein cattle during early, mid and late lactation in a tropical environmentANIMAL SCIENCE JOURNAL, Issue 2 2010Wilaiporn CHANCHAI ABSTRACT Two groups of five crossbred 87.5% Holstein cattle were housed in normal shade only (NS) as non-cooled cows and in shaded housing with misty-fan cooling (MF) as cooled cows. The cows were treated with recombinant bovine somatotropin (rbST) in early, mid and late lactation with three consecutive injections of rbST in every 14 days. Ambient temperatures and the temperature humidity index in the NS barn were significantly higher than those of the MF barn, whereas relative humidity in MF was higher than that of NS barn. The DMI of cooled cows were higher than those of non-cooled cows, and cooled cows exhibited more response to rbST treatment. Exogenous rbST significantly increased milk yield throughout lactation. The rbST-treated cows had higher total ruminal fermentation products as volatile fatty acid and ammonia nitrogen than the non-rbST treated cows and associated changes were greater in cooled animals in all stages of lactation. Exogenous rbST increased the concentrations of milk urea nitrogen in both groups. These results suggest that the changes in ruminal fermentation with greater production of total VFA and NH3N in response to rbST in crossbred cows whether under misty-fan cooling or not, is in part through an increase in feed intake, thereby making more substrate available to the mammary gland for milk synthesis. Uijl Objective,,, In the Netherlands, presurgical screening for temporal lobe epilepsy (TLE) includes the intracarotid amobarbital procedure (IAP), consisting of two consecutive injections of amobarbital, ipsilateral and contralateral to the epileptic focus. We studied whether a bilateral IAP has added value to a unilateral, ipsilateral IAP. Methods,,, This population-based study included 183 consecutive patients referred for screening for TLE surgery who underwent bilateral IAP. Using multivariable modeling, we assessed the added value of bilateral IAP on the decision for surgery, resection size, amygdalohippocampectomy, post-operative seizure freedom, memory performance, and IQ change. Results,,, Given the results from the unilateral IAP, the bilateral IAP had added prognostic value for postoperative change in verbal memory (P < 0.01) and verbal IQ (P < 0.01), especially if patients had a left-sided focus. In contrast, information provided by the contralateral IAP was not associated with decision-making or surgical strategy. Conclusions,,, A bilateral IAP has added value in predicting post-operative verbal memory and IQ. A bilateral IAP is currently not used to guide surgical strategy, but may be used for this purpose when verbal capacity is of particular concern in patients with a left-sided focus.
