Fish assemblages as influenced by environmental factors in streams in protected areas of the Czech Republic

Abstract,,, Three streams of comparable size located in different landscape-protected areas were selected for studying the effect of environmental factors on fish assemblages using indirect (detrended correspondence analysis, DCA) and direct (canonical correspondence analysis, CCA) gradient analysis. DCA of species showed well a gradient of assemblage changes in the longitudinal profile. DCA of sites stressed the variability between the fish assemblages of the three streams. This pattern was then confirmed by the highly significant between-stream CCA. In the within-site CCA, environmental factors explained 50.7% variability for presence,absence data and 58.3% for the relative abundance data. The analysis revealed that number of ponds and land use are the most influential factors of the strongest environmental gradient. However, in the partial CCAs, factor substratum type explained the largest proportion of the variability affecting fish in their habitat choice. Generally, presence,absence and relative abundance data of fish gave similar results in both DCA and CCA analyses; the same environmental factors proved to be important in both data type analyses. The environmental factors explain more variability than the regional (between-stream) one. The total proportion of variability explained by the presence,absence data analysis was 71.9% and in the relative abundance analysis even 80.8%. The environmental factors measured during the field survey explain 2.1- and 3.4-times more assemblages' variability than factors measured from a hydrological map. Resumen 1. Tres ríos de tamaño comparable localizados en diferentes áreas de paisaje protegido de la República Checa fueron seleccionados para estudiar el efecto de factores ambientales sobre los ensamblajes de peces. Para ello, utilizando análisis de gradientes indirectos (DCA) y directos (CCA). 2. El DCA para las especies enfatizó la variabilidad entre los ensamblajes de peces de los tres ríos. Este patrón fue confirmado por un CCA altamente significativo. Para la variabilidad dentro de la localidad, un CCA reveló que los factores ambientales explicaron un 50.7% para datos de presencia-ausencia y un 58.3% para las abundancias relativas. 3. Los análisis revelaron que el número de pozas y el uso del suelo fueron los factores de mayor influencia en el gradiente ambiental. Sin embargo, en el CCA parcial, el tipo de sustrato explicó la mayor proporción de la variabilidad que afecta a los peces en la elección de hábitat. 4. Generalmente los datos de presencia-ausencia y abundancia relativa produjeron resultados similares tanto en los análisis DCA como en los CCA; los mismos factores ambientales probaron ser importantes en los análisis de ambos tipos de datos. Los factores ambientales explicaron mas variabilidad que los regionales (entre ríos). La proporción total de variabilidad explicada por el análisis de los datos de presencia-ausencia fue 71.9% mientras que para las abundancias relativas fue de 80.8%. Los factores ambientales medidos durante los muestreos de campo explicaron 2.1 y 3.4 veces mas variabilidad que los factores medidos sobre mapas hidrológicos. [source]


Control of apatite crystal growth by the co-operative effect of a recombinant porcine amelogenin and fluoride

Recently, we used native amelogenins extracted from developing pig enamel to examine the combined effect of fluoride and amelogenins on the growth of octacalcium phosphate (OCP) and apatite crystals. The purpose of the present study was to investigate this combined effect using a highly purified recombinant amelogenin. We applied porcine amelogenin (rP172) and fluoride in a dual-membrane system as a model for tooth enamel formation. The combination of rP172 and fluoride in this system resulted in the formation of rod-like apatite crystals. On the other hand, without fluoride, rod-like OCP crystals of a comparable size were formed, and rather large hexagonal prisms of mixed crystals of OCP and apatite grew without amelogenins. Thus, highly purified and homogeneous recombinant amelogenin, in co-operation with F, regulated the mineral phase, habit, and size of crystals in the same manner as the extracted heterogeneous porcine amelogenins. We suggest that in both cases the control over the crystal phase and morphology was a direct effect of amelogenin protein serving as a scaffold for apatite mineralization. [source]


Time Course of Esophageal Lesions After Catheter Ablation with Cryothermal and Radiofrequency Ablation: Implication for Atrio-Esophageal Fistula Formation After Catheter Ablation for Atrial Fibrillation

Background: Atrio-esophageal fistulas have been described as a consequence of radiofrequency (RF) ablation for the treatment of atrial fibrillation (AF). However, whether cryoablation can avoid this potential fatal complication remains unclear. Methods and Results: We studied the effects of direct application of RF and cryoablation on the cervical esophagus in 16 calves. Cryoablation was performed with a 6.5-mm catheter probe using a single 5-minute freeze at <,80°C, and RF ablation was delivered with an 8-mm catheter electrode at 50 W and 50°C for 45,60 seconds. Histopathologic assessments were performed at 1, 4, 7, and 14 day(s) after completion of the ablation protocol: four animals were examined each day. A total of 85 direct esophageal ablations were performed: 41 with RF and 44 with cryoablation. There were no significant differences in lesion width, depth, or volume between cryoablation and RF ablation at Day 1, 4, and 14 after the procedure (P > 0.05). However, lesion width and volume were significantly larger with RF than with cryoablation at Day 7. Although acute (Day 1) and chronic (Day 14) RF and cryoablation lesions were of comparable size, histologic evidence of partial- to full-wall esophageal lesion ulceration was observed in 0 of 44 (0%) lesions with cryoablation, compared with 9 of 41 (22%) lesions with RF ablation (P = 0.0025). Conclusions: Direct application of cryoablation and RF ablation created similar acute and chronic lesion dimensions on the esophagus. However, cryoablation was associated with a significantly lower risk of esophageal ulceration, compared with RF ablation. [source]


P2Y receptor-activating nucleotides modulate cellular reactive oxygen species production in dissociated hippocampal astrocytes and neurons in culture independent of parallel cytosolic Ca2+ rise and change in mitochondrial potential

Abstract With mixed cultures of hippocampal astrocytes and neurons, we investigated the influence of nucleotides on cytosolic Ca2+ level, generation of reactive oxygen species (ROS), and mitochondrial potential. We employed ATP and four purine/pyrimidine derivates, which are P2Y receptor subtype-preferring agonists. Stimulation with ATP, a P2Y1/2/4 receptor agonist in rat, caused a large cytosolic Ca2+ increase in astrocytes and a considerably smaller Ca2+ response in neighboring neurons. The P2Y1 receptor antagonist MRS2179 completely blocked the ATP-induced Ca2+ response in astrocytes and neurons. Application of ATP significantly reduced the mitochondrial potential in neurons, which was not inhibited by MRS2179. Interestingly, MRS2179 mediated a mitochondrial depolarization without affecting the cytosolic Ca2+ level. Stimulation with UDP, a P2Y6 receptor agonist; UTP, a P2Y2/4 receptor agonist; 2MeSATP, a P2Y1 receptor agonist; or 2MeSADP, a P2Y1/12/13 receptor agonist, evoked significant Ca2+ responses in astrocytes but small Ca2+ responses in neurons. In astrocytes, there was an inverse relationship between the amplitude of the cytosolic Ca2+ peak and the rate of ROS generation in response to nucleotide application. Activation with UDP resulted in the highest ROS generation that we detected, whereas 2MeSADP and 2MeSATP reduced the ROS generation below the basal level. 2MeSADP and UDP caused mitochondrial depolarization of comparable size. Thus, neither in astrocytes nor in neurons did the degree of mitochondrial depolarization correlate with ROS generation. Nucleotides acting via P2Y receptors can modulate ROS generation of hippocampal neurons without acutely changing the cytosolic Ca2+ level. Thus, ROS might function as a signaling molecule upon nucleotide-induced P2Y receptor activation in brain. © 2007 Wiley-Liss, Inc. [source]


Development and characterization of a fusion protein between thermally responsive elastin-like polypeptide and interleukin-1 receptor antagonist: Sustained release of a local antiinflammatory therapeutic

Objective Interleukin-1 receptor antagonist (IL-1Ra) has been evaluated for the intraarticular treatment of osteoarthritis. Such administration of proteins may have limited utility because of their rapid clearance and short half-life in the joint. The fusion of a drug to elastin-like polypeptides (ELPs) promotes the formation of aggregating particles that form a "drug depot" at physiologic temperatures, a phenomenon intended to prolong the presence of the drug. The purpose of this study was to develop an injectable drug depot composed of IL-1Ra and ELP domains and to evaluate the properties and bioactivity of the recombinant ELP-IL-1Ra fusion protein. Methods Fusion proteins between IL-1Ra and 2 distinct sequences and molecular weights of ELP were overexpressed in Escherichia coli. Environmental sensitivity was demonstrated by turbidity and dynamic light scattering as a function of temperature. IL-1Ra domain activity was evaluated by surface plasmon resonance, and in vitro antagonism of IL-1,mediated lymphocyte and thymocyte proliferation, as well as IL-1,induced tumor necrosis factor , (TNF,) expression and matrix metalloproteinase 3 (MMP-3) and ADAMTS-4 messenger RNA expression in human intervertebral disc fibrochondrocytes. IL-1Ra immunoreactivity was assessed before and after proteolytic degradation of the ELP partner. Results Both fusion proteins underwent supramolecular aggregation at subphysiologic temperatures and slowly resolubilized at 37°C. Interaction with IL-1 receptor was slower in association but equivalent in dissociation as compared with the commercial antagonist. Anti,IL-1 activity was demonstrated by inhibition of lymphocyte and thymocyte proliferation and by decreased TNF, expression and ADAMTS-4 and MMP-3 transcription by fibrochondrocytes. ELP domain proteolysis liberated a peptide of comparable size and immunoreactivity as the commercial IL-1Ra. This peptide was more bioactive against lymphocyte proliferation, nearly equivalent to the commercial antagonist. Conclusion The ELP-IL-1Ra fusion protein proved to retain the characteristic ELP inverse phase-transitioning behavior as well as the bioactivity of the IL-1Ra domain. This technology represents a novel drug carrier designed to prolong the presence of bioactive peptides following intraarticular delivery. [source]


Recent observations of AB Dor and interpretation

Abstract We use minimal empirical modelling techniques to interpret recent (2006,2007) photometry and spectroscopy of AB Dor. We compare, in particular, broadband (B and V) maculation effects with emission features in high-resolution Ca II K-line spectroscopy. We also compare emission effects in the Ca II Kand H, lines observed at different rotational phases. We refer to a broader multiwavelength campaign, of which these optical data were a part, involving X-ray and microwave observations to be published later. The broadband light curves are characterized by one outstanding macula, whereas the emission lines suggest 4 possible main chromospheric activity sites (,faculae'). These appear at a similar latitude and with comparable size to the main umbra, but there are significant displacements in longitude. However, one strong facular concentration near phase zero may have a physical relationship to the main macula. The derived longitudes of these features would have been affected by differential rotation operating over the several months between the spectroscopic and photometric observations, but the difference of at least ,30° between facula and umbra appears too great to allow their coincidence. The possibility of a large bipolar surface structure is considered, keeping in mind the bipolar character of solar activity centres: the activity of rapidly rotating cool stars being generally compared with that of the Sun, scaled up by a few orders of magnitude. Observed microwave activity may link to this same main photospheric and chromospheric centre picked up by the optical analysis. Characterization of macular and facular contributions in stellar activity sites would be improved with a closer timing of observations and higher signal to noise ratios in emission line data (© 2009 WILEY-VCH Verlag GmbH & Co.
