    COMBINING REVEALED AND STATED PREFERENCE DATA TO ESTIMATE THE NONMARKET VALUE OF ECOLOGICAL SERVICES: AN ASSESSMENT OF THE STATE OF THE SCIENCE

    Abstract This paper reviews the marketing, transportation and environmental economics literature on the joint estimation of revealed preference (RP) and stated preference (SP) data. The RP and SP approaches are first described with a focus on the strengths and weaknesses of each. Recognizing these strengths and weaknesses, the potential gains from combining data are described. A classification system for combined data that emphasizes the type of data combination and the econometric models used is proposed. A methodological review of the literature is pursued based on this classification system. Examples from the environmental economics literature are highlighted. A discussion of the advantages and disadvantages of each type of jointly estimated model is then presented. Suggestions for future research, in particular opportunities for application of these models to environmental quality valuation, are presented. [source]


    On-line motion blending for real-time locomotion generation

    Abstract In this paper, we present an integrated framework of on-line motion blending for locomotion generation. We first provide a novel scheme for incremental timewarping, which always guarantees that the time goes forward. Combining the idea of motion blending with that of posture rearrangement, we introduce a motion transition graph to address on-line motion blending and transition simultaneously. Guided by a stream of motion specifications, our motion synthesis scheme moves from node to node in an on-line manner while blending a motion at a node and generating a transition motion at an edge. For smooth on-line motion transition, we also attach a set of example transition motions to an edge. To represent similar postures consistently, we exploit the inter-frame coherency embedded in the input motion specification. Finally, we provide a comprehensive solution to on-line motion retargeting by integrating existing techniques. Copyright © 2004 John Wiley & Sons, Ltd. [source]


    Guidelines for the Management of Squamous Cell Carcinoma in Organ Transplant Recipients

    Background. Solid-organ transplant recipients have a high incidence of cutaneous squamous cell carcinoma (SCC) and often develop multiple and aggressive tumors. There are few published studies or reviews, which provide guidance to the clinician in the treatment of these patients. Objective. The objective was to develop useful clinical guidelines for the treatment of skin cancer in organ transplant recipients (OTRs). Methods. The members of the Guidelines Committee of the International Transplant,Skin Cancer Collaborative (ITSCC) carried out a computerized search utilizing the databases of the National Library of Medicine for reports in the literature on SCC in OTRs. These reports were collectively examined by the group and combined with experiences from the members' clinical practices in the development of the guidelines. Results. More than 300 articles relating to SCC in OTRs were reviewed. In general, reports concerning the prevention and treatment of SCC in OTRs are of individual cases or small case series. They are retrospective in nature, statistically nonrigorous, and lack the complete epidemiologic data necessary to derive definitive conclusions. Combining these studies and collective clinical experience, however, is at present the best available method for devising guidelines for the treatment of SCC in OTRs. Conclusion. Guidelines developed for the treatment of skin cancer in OTRs, supported by the best available data and collective clinical experience, may assist in the management of OTRs with SCC. The development of clinical pathways and complete documentation with rigorous prospective study is necessary to improve and refine future guideline development. [source]


    Donor Harvesting: A New Approach to Minimize Transection of Hair Follicles

    Background. There are several methods for harvesting donor hairs, including punch excision, single-bladed knife excision, and multibladed knife excision. All of these procedures are blind and thus result in transection of hair follicles. Transection of hair follicles during harvesting results in fewer follicles being available for transplantation, detrimentally affecting the final cosmetic result. Objective. To explore a new method of donor hair harvesting called "donor dissecting." This new procedure is an open technique because hair follicles are directly visualized during the harvesting process. Methods. The technique of donor dissecting utilizes a #15 scalpel blade to excise the donor hair ellipse from the occiput while maintaining meticulous hemostasis. This enables individual hair follicles to be visualized and protected from transection during the harvesting process. Once the donor ellipse is harvested, it is then further divided into individual mini- and micrografts using direct visualization of individual follicles to again prevent transection. Results. The technique of donor dissecting was utilized in 50 consecutive hair transplant patients. Utilizing this new technique, only 1.9% of hair follicles in the donor ellipse were transected during the harvesting process. The dissection of the donor ellipse 1.2% follicles being transected in the graft cutting process. Combining the donor dissection technique with dissection of the individual grafts, we were able to transect 1.59% of hair follicles harvested for transplantation. Conclusion. The technique of donor dissection minimizes the transection of hair follicles in the donor hair harvesting phase of hair transplantation. This technique is superior to the blind methods of donor harvesting which have been plagued by the problem of hair follicle transection. [source]


    Medical treatment of hirsutism

    ABSTRACT:, Hirsutism is usually the result of an underlying adrenal, ovarian, or central endocrine abnormality mainly due to polycystic ovary syndrome but may also be idiopathic or drug induced. The aim of medical treatment of hirsutism is to rectify any causal hormonal balance, slow down or stop excessive hair growth, and improve the aesthetic appearance of hirsutism, thereby positively affecting the patient's quality of life. Today, for the majority of women, a monotherapy with oral contraceptives that have antiandrogenic activity is recommended as a first-line treatment for hirsutism. Combining an oral contraceptive pill with an antiandrogen is recommended if clinical improvement of hirsutism is insufficient after 6,9 months' monotherapy. In women who present with hirsutism, hyperandrogenism, and insulin resistance, insulin sensitizers are effective for the hirsutism as well as the hyperinsulinemia, hyperandrogenism, and infertility but there is no convincing evidence that they are effective for hirsutism alone. Topical eflornithine is a medical therapy that can be a useful adjuvant for hirsutism when used in conjunction with systemic medications or with laser/photoepilation. [source]


    Developmental profiles for multiple object tracking and spatial memory: typically developing preschoolers and people with Williams syndrome

    The ability to track moving objects, a crucial skill for mature performance on everyday spatial tasks, has been hypothesized to require a specialized mechanism that may be available in infancy (i.e. indexes). Consistent with the idea of specialization, our previous work showed that object tracking was more impaired than a matched spatial memory task in individuals with Williams syndrome (WS), a genetic disorder characterized by severe visuo-spatial impairment. We now ask whether this unusual pattern of performance is a reflection of general immaturity or of true abnormality, possibly reflecting the atypical brain development in WS. To examine these two possibilities, we tested typically developing 3- and 4-year-olds and people with WS on multiple object tracking (MOT) and memory for static spatial location. The maximum number of objects that could be correctly tracked or remembered (estimated from the k -statistic) showed similar developmental profiles in typically developing 3- and 4-year-old children, but the WS profile differed from either age group. People with WS could track more objects than 3-year-olds, and the same number as 4-year-olds, but they could remember the locations of more static objects than both 3- and 4-year-olds. Combining these data with those from our previous studies, we found that typically developing children show increases in the number of objects they can track or remember between the ages of 3 and 6, and these increases grow in parallel across the two tasks. In contrast, object tracking in older children and adults with WS remains at the level of 4-year-olds, whereas the ability to remember multiple locations of static objects develops further. As a whole, the evidence suggests that MOT and memory for static location develop in tandem typically, but not in WS. Atypical development of the parietal lobe in people with WS could play a causal role in the abnormal, uneven pattern of performance in WS. This interpretation is consistent with the idea that multiple object tracking engages different mechanisms from those involved in memory for static object location, and that the former can be particularly disrupted by atypical development.     The Econometrics of Ultra-high-frequency Data

    Ultra-high-frequency data is defined to be a full record of transactions and their associated characteristics. The transaction arrival times and accompanying measures can be analyzed as marked point processes. The ACD point process developed by Engle and Russell (1998) is applied to IBM transactions arrival times to develop semiparametric hazard estimates and conditional intensities. Combining these intensities with a GARCH model of prices produces ultra-high-frequency measures of volatility. Both returns and variances are found to be negatively influenced by long durations as suggested by asymmetric information models of market micro-structure. [source]


    The political economy of direct legislation: direct democracy and local decision,making

    Local and regional governments account for an important share of total government spending and, given the decentralization trend in OECD nations, this is likely to increase. How should this spending be governed? This article argues that direct democracy is best suited to organize decision,making at the state and local level. To support this, we present the main theoretical arguments on why and how referenda and initiatives affect fiscal policy outcomes. The basic argument concerns voter control. Under representative democracy, citizens only have direct control at election time. With referenda and initiatives, citizens can selectively control their representatives on specific policies whenever they deviate sufficiently from citizens' preferences. As a result, fiscal policy outcomes are likely to more closely reflect voter preferences. We empirically test this on Swiss data since Switzerland provides a ,natural laboratory' for local governance. The governance structures of Swiss cantons and localities with respect to fiscal issues range from classic parliamentary democracy to pure direct democracy, and an important part of spending and taxation is controlled at these levels. Specifically, we estimate an econometric model of fiscal behaviour using data from 1986 to 1997 for the 26 Swiss cantons, and 1990 data on 134 local communities. It is shown that mandatory referenda on fiscal issues at both levels have a dampening effect on expenditure and revenue, and at the local level also on public debt. Combining this with existing empirical evidence leads to a relatively uncontested result, namely that elements of direct democracy are associated with sounder public finances, better economic performance and higher satisfaction of citizens.     Developing transgenic arabidopsis plants to be metal-specific bioindicators

    Abstract Deoxyribonucleic acid (DNA) microarrays provide a means to assess genome-wide expression patterns after exposure of an organism to different xenobiotics. Potential uses for this technology include identification of unknown toxicants, assessment of toxicity of new compounds, and characterization of the cellular mechanisms of toxicant action. Here we describe another use of DNA microarrays in toxicant-specific gene discovery. Combining results from two DNA microarray experiments, we have identified genes from the model plant Arabidopsis thaliana that are induced in response to one but not other heavy metals. The promoters of these genes should be useful in developing metal-specific transgenic biomonitors. To test this idea, we have fused the promoter of one of the newly identified Ni-inducible genes (AHB1) to the ,-glucuronidase (GUS) reporter gene. Arabidopsis plants containing the AHB1::GUS transgene show reporter gene activity when they are grown on media containing Ni but not when grown on media containing Cd, Cu, Zn, or without added metals. Thus, this approach has resulted in the creation of a transgenic strain of Arabidopsis that can report on the presence and concentration of Ni in plant growth media. Such transgenic models can serve as cheap and efficient biomonitors of bioavailable heavy metal contamination in soils and sediments. [source]


    Selection of Antiepileptic Drug Polytherapy Based on Mechanisms of Action: The Evidence Reviewed

    Summary: Purpose: When monotherapy with antiepileptic drugs (AEDs) fails, combination therapy is tried in an attempt to improve effectiveness by improving efficacy, tolerability, or both. We reviewed the available studies (both animal and human) on AED polytherapy to determine whether AEDs can be selected for combination therapy based on their mechanisms of action, and if so, which combinations are associated with increased effectiveness. Because various designs and methods of analysis were used in these studies, it was also necessary to evaluate the appropriateness of these approaches. Methods: Published papers reporting on AED polytherapy in animals or humans were identified by Medline search and by checking references cited in these papers. Results: Thirty-nine papers were identified reporting on two-drug AED combinations. Several combinations were reported to offer improved effectiveness, but no uniform approach was used in either animal or human studies for the evaluation of pharmacodynamic drug interactions; efficacy was often the only end point. Conclusions: There is evidence that AED polytherapy based on mechanisms of action may enhance effectiveness. In particular, combining a sodium channel blocker with a drug enhancing GABAergic inhibition appears to be advantageous. Combining two GABA mimetic drugs or combining an AMPA antagonist with an NMDA antagonist may enhance efficacy, but tolerability is sometimes reduced. Combining two sodium channel blockers seems less promising. However, given the incomplete knowledge of the pathophysiology of seizures and indeed of the exact mechanisms of action of AEDs, an empirical but rational approach for evaluating AED combinations is of fundamental importance. This would involve appropriate testing of all possible combinations in animal models and subsequent evaluation of advantageous combinations in clinical trials. Testing procedures in animals should include the isobologram method, and the concept of drug load should be the basis of studies in patients with epilepsy. [source]


    A role for nitric oxide in sensory-induced neurogenesis in an adult insect brain

    In the adult cricket, neurogenesis occurs in the mushroom bodies, the main integrative structures of the insect brain. Mushroom body neuroblast proliferation is modulated in response to environmental stimuli. However, the mechanisms underlying these effects remain unspecified. In the present study, we demonstrate that electrical stimulation of the antennal nerve mimics the effects of olfactory activation and increases mushroom body neurogenesis. The putative role of nitric oxide (NO) in this activity-regulated neurogenesis was then explored. In vivo and in vitro experiments demonstrate that NO synthase inhibition decreases, and NO donor application stimulates neuroblast proliferation. NADPH-d activity, anti- l -citrulline immunoreactivity, as well as in situ hybridization with a probe specific for Acheta NO synthase were used to localize NO-producing cells. Combining these three approaches we clearly establish that mushroom body interneurons synthesize NO. Furthermore, we demonstrate that experimental interventions known to upregulate neuroblast proliferation modulate NO production: rearing crickets in an enriched sensory environment induces an upregulation of Acheta NO synthase mRNA, and unilateral electrical stimulation of the antennal nerve results in increased l -citrulline immunoreactivity in the corresponding mushroom body. The present study demonstrates that neural activity modulates progenitor cell proliferation and regulates NO production in brain structures where neurogenesis occurs in the adult insect. Our results also demonstrate the stimulatory effect of NO on mushroom body neuroblast proliferation. Altogether, these data strongly suggest a key role for NO in environmentally induced neurogenesis.     Distribution of polycyclic aromatic hydrocarbons in particle-size separates and density fractions of typical agricultural soils in the Yangtze River Delta, east China

    Summary Soil organic matter can be divided into different organic carbon (C) pools with different turnover rates. The organic pollutants in soils associated with these organic C pools may have different bioavailability and environmental risks during the decomposition of soil organic matter. We studied the distribution patterns of 15 USEPA priority polycyclic aromatic hydrocarbons (PAHs) in different particle-size separates (clay, fine silt, coarse silt, fine sand and coarse sand) and density fractions (light and heavy fractions) of nine agricultural topsoils (0,20 cm depth) from a contaminated area in the Yangtze River Delta region of east China. There was a decreasing trend in PAH concentration in particle-size separates with decreasing particle size. However, the different particle-size separates had similar PAH composition. The concentration of PAHs in the light fraction ranged from 13 037 to 107 299 ,g kg,1, far higher than in the heavy fraction, which ranged from 222 to 298 ,g kg,1. Although the light fraction accounted for only 0.4,2.3% of the soils, it was associated with 31.5,69.5% of soil PAHs. The organic matter in coarse silt had the strongest capacity for enrichment with PAHs. Combining the distributions of PAHs and the turnover rates of organic matter in different soil fractions, the environmental risks of PAH-polluted soils may be due mainly to the PAHs associated with sand and the light fraction.     Abstract: Neural network ensembles (sometimes referred to as committees or classifier ensembles) are effective techniques to improve the generalization of a neural network system. Combining a set of neural network classifiers whose error distributions are diverse can generate better results than any single classifier. In this paper, some methods for creating ensembles are reviewed, including the following approaches: methods of selecting diverse training data from the original source data set, constructing different neural network models, selecting ensemble nets from ensemble candidates and combining ensemble members' results.     Protein engineering of Bacillus megaterium CYP102

    The oxidation of polycyclic aromatic hydrocarbons
    Cytochrome P450 (CYP) enzymes are involved in activating the carcinogenicity of polycyclic aromatic hydrocarbons (PAHs) in mammals, but they are also utilized by microorganisms for the degradation of these hazardous environmental contaminants. Wild-type CYP102 (P450BM-3) from Bacillus megaterium has low activity for the oxidation of the PAHs phenanthrene, fluoranthene and pyrene. The double hydrophobic substitution R47L/Y51F at the entrance of the substrate access channel increased the PAH oxidation activity by up to 40-fold. Combining these mutations with the active site mutations F87A and A264G lead to order of magnitude increases in activity. Both these mutations increased the NADPH turnover rate, but the A264G mutation increased the coupling efficiency while the F87A mutation had dominant effects in product selectivity. Fast NADPH oxidation rates were observed (2250 min,1 for the R47L/Y51F/F87A mutant with phenanthrene) but the coupling efficiencies were relatively low (< 13%), resulting in a highest substrate oxidation rate of 110 min,1 for fluoranthene oxidation by the R47L/Y51F/A264G mutant. Mutation of M354 and L437 inside the substrate access channel reduced PAH oxidation activity. The PAHs were oxidized to a mixture of phenols and quinones. Notably mutants containing the A264G mutation showed some similarity to mammalian CYP enzymes in that some 9,10-phenanthrenequinone, the K -region oxidation product from phenanthrene, was formed. The results suggest that CYP102 mutants could be useful models for PAH oxidation by mammalian CYP enzymes, and also potentially for the preparation of novel PAH bioremediation systems. [source]


    Defect-Related Emissions and Magnetization Properties of ZnO Nanorods

    Abstract A clear correlation between defect-related emissions and the magnetization of ZnO nanorods synthesized by a one-step aqueous chemical method is demonstrated. The relative contribution of the emission bands arising from various types of defects is determined and found to be linked with the size of the nanorods and annealing conditions. When the size of the nanorods and the annealing temperature are increased, the magnetization of pure ZnO nanorods decreases with the reduction of a defect-related band originating from singly charged oxygen vacancies (). With a sufficient increase of annealing temperature (at 900,°C), the nanorods show diamagnetic behavior. Combining with the electron paramagnetic resonance results, a direct link between the magnetization and the relative occupancy of the singly charged oxygen vacancies present on the surface of ZnO nanorods is established.     Abstract The wetting properties of structured self-assembled monolayers are used to fabricate sample target substrates for MALDI-TOF mass spectrometry. Combining the advantages of a hydrophobic-hydrophilic surface pattern and the possibility of obtaining micrometer patterns allows an increase in the sensitivity of MALDI-TOF mass spectrometry analysis and a reduction in the traceable concentration down to fmol µL,1. This easy, cheap and fast patterning process provides substrates that allow sensitive, high-resolution mass spectrometry of dilute solutions. [source]


    New hemocyte-specific enhancer-reporter transgenes for the analysis of hematopoiesis in Drosophila

    Abstract Based on environmental challenges or altered genetic composition, Drosophila larvae can produce up to three types of blood cells that express genetic programs essential for their distinct functions. Using transcriptional enhancers for genes expressed exclusively in plasmatocytes, crystal cells, or lamellocytes, several new hemocyte-specific enhancer-reporter transgenes were generated to facilitate the analysis of Drosophila hematopoiesis. This approach took advantage of fluorescent variants of insulated P-element reporter vectors for multilabeling cell analyses; two additional color variants were generated in these studies. These vectors were successfully used to produce transgenic fly lines that label specific hemocyte lineages with separate colors. Combining three transgene reporters allowed for the unambiguous identification of plasmatocytes, crystal cells, and lamellocytes within a complex hemocyte population. While this work focused on the hematopoietic process, these new vectors can be used to mark multiple cell types or trace complex cell lineages during any chosen aspect of Drosophila development. genesis 47:771,774, 2009. © 2009 Wiley-Liss, Inc.     Crustal structure of central Tibet as derived from project INDEPTH wide-angle seismic data

    Summary In the summer of 1998, project INDEPTH recorded a 400 km long NNW,SSE wide-angle seismic profile in central Tibet, from the Lhasa terrane across the Banggong-Nujiang suture (BNS) at about 89.5°E and into the Qiangtang terrane. Analysis of the P- wave data reveals that (1) the crustal thickness is 65 ± 5 km beneath the line; (2) there is no 20 km step in the Moho in the vicinity of the BNS, as has been suggested to exist along-strike to the east based on prior fan profiling; (3) a thick high-velocity lower crustal layer is evident along the length of the profile (20,35 km thick, 6.5,7.3 km s,1); and (4) in contrast to the southern Lhasa terrane, there is no obvious evidence of a mid-crustal low-velocity layer in the P- wave data, although the data do not negate the possibility of such a layer of modest proportions. Combining the results from the INDEPTH III wide-angle profile with other seismic results allows a cross-section of Moho depths to be constructed across Tibet. This cross-section shows that crustal thickness tends to decrease from south to north, with values of 70,80 km south of the middle of the Lhasa terrane, 60,70 km in the northern part of the Lhasa terrane and the Qiangtang terrane, and less than 60 km in the Qaidam basin. The overall northward thinning of the crust evident in the combined seismic observations, coupled with the essentially uniform surface elevation of the plateau south of the Qaidam basin, is supportive of the inference that northern Tibet until the Qaidam basin is underlain by somewhat thinner crust, which is isostatically supported by relatively low-density, hot upper mantle with respect to southern Tibet.     Noble Metal Nanocrystal-Incorporated Fullerene-Like Polyoxometalate Based Microspheres

    Abstract Monodisperse fullerene-like polyoxometalate-based microspheres are synthesized using a single-phase approach which is quick, easier to control surfactant to polyoxometalate (POM) molar ratios and result in better control over the size-distribution range. Noble-metal nanocrystals are successfully incorporated in the surfactant encapsulated complex multilayered spherical assemblies without any change in their layered morphology. Combining the merits of fullerene-like POMs and monodisperse noble metal nanocrystals, these monodisperse hybrid spheres show interesting composition- and size-dependent photochemical properties.     Loss of forb diversity in relation to nitrogen deposition in the UK: regional trends and potential controls

    Abstract In this study we investigate the impact of nitrogen (N) deposition on the diversity of three different vegetation functional groups , forbs, grasses and mosses , using a field survey of acid grasslands across Great Britain. Our aim is to identify the vegetation types that are most vulnerable to enhanced N deposition, and to shed light on the mechanisms that may be driving N-initiated species changes in the UK. Sixty-eight randomly selected grasslands belonging to the UK National Vegetation Classification group U4 (Festuca ovina,Agrostis capillaris,Galium saxatile grassland) were studied along a gradient of atmospheric N deposition ranging from 6 to 36 kg N ha,1 yr,1. At each site, vegetation was surveyed and samples were taken from the topsoil and subsoil. Aboveground plant material was collected from three species: a forb, grass and moss. Both the species richness and cover of forbs declined strongly with increasing N deposition, from greater than eight species/20% cover per m2 quadrat at low levels of N to fewer than two species/5% cover at the highest N deposition levels. Grasses showed a weak but significant decline in species richness, and a trend toward increasing cover with increasing N input. Mosses showed no trends in either species richness or cover. Most of the decline in plant species richness could be accounted for by the level of ammonium deposition. Soil KCl-extractable ammonium concentration showed a significant positive correlation with N input, but there was no relationship between N deposition and extractable nitrate. In the soil O/A horizon, there was no relationship between N deposition and %N, and only a very weak positive relationship between the level of N deposition and the C : N ratio. Finally, in the vegetation, there was no relationship between N deposition and either shoot tissue N concentration or N : P ratio for any of the three reference species. Combining our regional survey with the results of published N-addition experiments provides compelling evidence that there has been a significant decline in the species richness and cover of forbs across Great Britain, and that the primary cause is competition due to an increase in the cover of grasses in response to enhanced deposition of reactive N, primarily NH4+.     Variation in litterfall-climate relationships between coniferous and broadleaf forests in Eurasia

    ABSTRACT Aim, The objectives of this study were to determine the relationships between climatic factors and litterfall in coniferous and broadleaf forests in Eurasia and to explore the difference in litterfall between coniferous and broadleaf forests as related to climate at a continental scale. Location, We have used data from across Eurasia. Methods, The relationships between litterfall and climatic factors were examined using linear regression analysis of a compilation of published data from coniferous and broadleaf forests in Eurasia. Results, The relationships between litterfall and climatic factors show that in the temperate, subtropical, and tropical areas, broadleaf forests had higher litterfall than coniferous ones, whilst the opposite was found for boreal forests. Combining all climatic zones, a multiple regression analysis using annual mean temperature (T) and annual precipitation (P) as independent variables gave an adjusted R2 () of 0.272 for total litterfall in coniferous forests (n = 199, P < 0.001), 0.498 for broadleaf litterfall (n = 240, P < 0.001), and 0.535 for combined coniferous and broadleaf litterfall (n = 439, P < 0.001). The linear models for broadleaf stands have significantly higher coefficients for T and P than those for coniferous ones but the intercepts were similar. Thus, litterfall in broadleaf forests increased faster with T and P than that in coniferous forests. Further, a transformation of temperature and precipitation to relative units showed that a relative-unit change in T had a larger impact than P on total litterfall in broadleaf forests. The results indicate that at a continental scale, climatic controls over litterfall differ between coniferous and broadleaf forests. Conclusions, A relative unit change in annual mean temperature has a greater effect on litterfall compared to the same change in annual precipitation across the Eurasian forests. Further, the higher response to T for broadleaf forests indicates a difference in climate control between coniferous and broadleaf forests at a continental scale, and consequently different litterfall responses to climate change.     MODFLOW 2000 Head Uncertainty, a First-Order Second Moment Method

    A computationally efficient method to estimate the variance and covariance in piezometric head results computed through MODFLOW 2000 using a first-order second moment (FOSM) approach is presented. This methodology employs a first-order Taylor series expansion to combine model sensitivity with uncertainty in geologic data. MOD-FLOW 2000 is used to calculate both the ground water head and the sensitivity of head to changes in input data. From a limited number of samples, geologic data are extrapolated and their associated uncertainties are computed through a conditional probability calculation. Combining the spatially related sensitivity and input uncertainty produces the variance-covariance matrix, the diagonal of which is used to yield the standard deviation in MODFLOW 2000 head. The variance in piezometric head can be used for calibrating the model, estimating confidence intervals, directing exploration, and evaluating the reliability of a design. A case study illustrates the approach, where aquifer transmis-sivity is the spatially related uncertain geologic input data. The FOSM methodology is shown to be applicable for calculating output uncertainty for (1) spatially related input and output data, and (2) multiple input parameters (trans-missivity and recharge).     Early on-treatment prediction of response to peginterferon alfa-2a for HBeAg-negative chronic hepatitis B using HBsAg and HBV DNA levels,

    Peginterferon alfa-2a results in a sustained response (SR) in a minority of patients with hepatitis B e antigen (HBeAg),negative chronic hepatitis B (CHB). This study investigated the role of early on-treatment serum hepatitis B surface antigen (HBsAg) levels in the prediction of SR in HBeAg-negative patients receiving peginterferon alfa-2a. HBsAg (Architect from Abbott) was quantified at the baseline and during treatment (weeks 4, 8, 12, 24, 36, and 48) and follow-up (weeks 60 and 72) in the sera from 107 patients who participated in an international multicenter trial (peginterferon alfa-2a, n = 53, versus peginterferon alfa-2a and ribavirin, n = 54). Overall, 24 patients (22%) achieved SR [serum hepatitis B virus (HBV) DNA level < 10,000 copies/mL and normal alanine aminotransferase levels at week 72]. Baseline characteristics were comparable between sustained responders and nonresponders. From week 8 onward, serum HBsAg levels markedly decreased in sustained responders, whereas only a modest decline was observed in nonresponders. However, HBsAg declines alone were of limited value in the prediction of SR [area under the receiver operating characteristic curve (AUC) at weeks 4, 8, and 12 = 0.59, 0.56, and 0.69, respectively]. Combining the declines in HBsAg and HBV DNA allowed the best prediction of SR (AUC at week 12 = 0.74). None of the 20 patients (20% of the study population) in whom a decrease in serum HBsAg levels was absent and whose HBV DNA levels declined less than 2 log copies/mL exhibited an SR (negative predictive value = 100%). Conclusion: At week 12 of peginterferon alfa-2a treatment for HBeAg-negative CHB, a solid stopping rule was established with a combination of declines in serum HBV DNA and HBsAg levels from the baseline. Methods and results:, The synovitis score was determined in 666 synovial specimens: normal synovium, n = 33; post-traumatic arthropathy (PtA), n = 29; osteoarthritis (OA), n = 221; psoriatic arthritis (PsA), n = 42; and rheumatoid arthritis (RA), n = 341. The discriminatory abilities of the score and its components were quantified with binary and multicategory receiver operating characteristic (ROC) analysis. The score differentiated all arthropathies accurately from normal tissue (area under the ROC curve, AUC: 0.87,0.98) and RA from OA or PtA (AUC: 0.85 for both), but could not distinguish well within pairs of inflammatory or degenerative arthropathies. AUCs of the intimal hyperplasia and stromal cellularity components correlated with the AUCs of the complete score markedly more strongly (r = 0.94 and 0.91, respectively) than the inflammatory infiltration component (r = 0.60). Multicategory ROC analysis ranked the score several-fold higher than any of its components, and the components in the order stromal cellularity>intimal hyperplasia>infiltration. Conclusion:, Combining three distinct histological parameters into a three-component score produces greatly increased overall diagnostic power. The discriminatory ability of the score stems more from measuring proliferative than infiltrative aspects of synovitis. [source]

    Heterozygous SOX9 Mutations Allowing for Residual DNA-binding and Transcriptional Activation Lead to the Acampomelic Variant of Campomelic Dysplasia,

    Abstract Campomelic dysplasia is a malformation syndrome with multiple symptoms including characteristic shortness and bowing of the long bones (campomelia). CD, often lethal due to airway malformations, is caused by heterozygous mutations in SOX9, an SRY-related gene regulating testis and chondrocyte development including expression of many cartilage genes such as type II collagen. Male to female sex reversal occurs in the majority of affected individuals with an XY karyotype. A mild form without campomelia exists, in which sex-reversal may be also absent. We report here two novel SOX9 missense mutations in a male (c.495C>G; p.His165Gln) and a female (c.337A>G; p.Met113Val) within the DNA-binding domain leading to non-lethal acampomelic CD. Functional analyses of mutant proteins demonstrate residual DNA-binding and transactivation of SOX9-regulated genes. Combining our data and reports from the literature we postulate a genotype-phenotype correlation: SOX9 mutations allowing for residual function lead to a mild form of CD in which campomelia and sex reversal may be absent. © 2010 Wiley-Liss, Inc.     Suspended-sediment sources in an urban watershed, Northeast Branch Anacostia River, Maryland

    Abstract Fine sediment sources were characterized by chemical composition in an urban watershed, the Northeast Branch Anacostia River, which drains to the Chesapeake Bay. Concentrations of 63 elements and two radionuclides were measured in possible land-based sediment sources and suspended sediment collected from the water column at the watershed outlet during storm events. These tracer concentrations were used to determine the relative quantity of suspended sediment contributed by each source. Although this is an urbanized watershed, there was not a distinct urban signature that can be evaluated except for the contributions from road surfaces. We identified the sources of fine sediment by both physiographic province (Piedmont and Coastal Plain) and source locale (streambanks, upland and street residue) by using different sets of elemental tracers. The Piedmont contributed the majority of the fine sediment for seven of the eight measured storms. The streambanks contributed the greatest quantity of fine sediment when evaluated by source locale. Street residue contributed 13% of the total suspended sediment on average and was the source most concentrated in anthropogenically enriched elements. Combining results from the source locale and physiographic province analyses, most fine sediment in the Northeast Branch watershed is derived from streambanks that contain sediment eroded from the Piedmont physiographic province of the watershed. Sediment fingerprinting analyses are most useful when longer term evaluations of sediment erosion and storage are also available from streambank-erosion measurements, sediment budget and other methods.     Recharge, upflux and water table response for shallow water table conditions in southwest Florida

    Abstract A disproportionate increase or decrease in water table in response to minor water input or drainage is observed in shallow water table conditions inside drainage lysimeters. This increase happens because the capillary fringe of the shallow water table reaches up to or near the surface (Wieringermeer effect). The correlations between water table level changes and rainfall, seepage irrigation, drip irrigation, and drainage were analysed. Correlations with rainfall, seepage irrigation, and drainage were high (R2 ranged from 0·46 to 0·97). Drip irrigation had low correlations due to the low rates of application (R2 ranged from 0·26 to 0·44). Conventional methods of calculating recharge, such as multiplying the specific yield with the water table fluctuations, cannot be used for Wieringermeer effect situations. A method using water balance data and soil moisture at different depths in the lysimeters was developed to estimate recharge and upflux. The recharge results were used to develop the apparent specific yield Sya, which could be used to calculate consequent recharge events from water table fluctuation data. Combining the water table fluctuation relationships developed with the Sya value will allow the prediction of recharge from rainfall and irrigation events without the need for soil moisture equipment.     Establishment and recall of CD8+ T-cell memory in a model of localized transient infection

    Summary:, The influenza A virus model of localized, transient respiratory infection provides a well-defined experimental system for dissecting the induction and maintenance of CD8+ T-cell memory. This review focuses on quantitative and qualitative aspects of the prominent DbNP366 - and DbPA224 -specific CD8+ T-cell responses in virus-infected B6 mice. The different virus-specific effector and memory sets are compared by phenotypic [CD62L, interleukin-7 receptor-, (IL-7R,), and IL-15R, expression] and functional [interferon-, (IFN-,), tumor necrosis factor-, (TNF-,), and IL-2 production] analyses. Most clonotypes [defined by T-cell receptor (TCR) CDR3, sequence] generated during the acute phase of infection survive into memory, with those expressing the more consensus ,canonical' TCRs being the major contributors to the recall response. The extent of clonal expansion and the size of memory CD8+ T-cell populations has been characterized for mice challenged with either wildtype or mutant viruses, where broadly equivalent DbNP366 and DbPA224 expression was achieved by disabling the peptides in their native configuration, then expressing them in the viral neuraminidase protein. Combining the clonotypic and antigen dose analyses led to a somewhat mechanistic conclusion that the magnitude of any virus-specific CD8+ T-cell response will be a direct function of antigen dose and the size of the naïve or memory CD8+ T-cell precursor pool.
