Selected AbstractsCD4 count and viral load time-courses in patients treated with highly active antiretroviral therapy and association with the CDC staging systemHIV MEDICINE, Issue 8 2006J Collazos Objectives The aim of the study was to analyse CD4 cell count and viral load dynamics in patients undergoing antiretroviral therapy and their association with the Centers for Disease Control and Prevention (CDC) classification system. Methods CD4 cell count and viral load were determined in 2982 patients who were classified according to clinical and immunological CDC stages. Measurements were carried out at baseline and at the 3rd, 6th and 12th months. Results Clear differences in the immunological and virological responses to therapy were observed depending on the CDC stage, with better results associated with less advanced stages. There was a marked parallelism in the CD4 cell count curves of the different CDC stages over the year of follow up, in both naïve and experienced patients, indicating that the increase in CD4 cell count at each time-point was similar for all clinical and immunological CDC stages. However, as the baseline values were closely associated with CDC stage, the CD4 cell counts finally reached were clearly dependent on CDC stage. The highest virological responses were observed during the initial 3 months, particularly in naïve patients, but whereas naïve patients showed additional increases up to the 6th month experienced patients reached a plateau at the 3rd month. The CD4 increases were also higher during the initial 3 months but persisted during the year of follow-up. Conclusion Both clinical and immunological CDC stages at baseline are highly predictive of the immunological and virological response to therapy, a finding that could have clinical implications. Dissolution of root canal sealer cements in volatile solvents. International Endodontic Journal, 33, 19,24, 2000. Aim There are few published data on the solubility profiles of endodontic sealers in solvents commonly employed in root canal retreatment. This study tested the hypothesis that root canal sealer cements are insoluble in the volatile solvents chloroform and halothane. Methodology Standardized samples (n=5) of glass ionomer (Ketac Endo), zinc oxide-eugenol (Tubli-Seal EWT), calcium hydroxide (Apexit) and epoxy resin (AH Plus) based sealers were immersed in chloroform or halothane for 30 s, 1 min, 5 min and 10 min. Mean loss of weight was plotted against time of exposure, and differences in behaviour assessed by multiple paired t-tests (P <0.01). Results Clear differences were shown in the solubility profiles of major classes of root canal sealer cements in two common volatile solvents. In comparison with other classes of material, Ketac Endo was the least soluble in chloroform and halothane (P <0.01), with less than 1% weight loss after 10 min exposure to either solvent. Apexit had low solubility with 11.6% and 14.19% weight loss after 10 min exposure to chloroform and halothane, respectively. The difference between solvents was not significant (P >0.01). Tubli-Seal EWT was significantly less soluble in halothane than chloroform (5.19% and 62.5% weight loss after 10 min exposure, respectively (P <0.01)). Its solubility in halothane was not significantly different from that of Apexit. AH Plus was significantly more soluble than all other materials in both chloroform and halothane (96% and 68% weight loss after 10 min exposure, respectively (P <0.01)). Conclusions There are significant differences in the solubility profiles of major classes of root canal sealer in common organic solvents. Efforts should continue to find a more universally effective solvent for use in root canal retreatment. [source] Eating fast food: attitudes of high-school studentsINTERNATIONAL JOURNAL OF CONSUMER STUDIES, Issue 1 2007Jan Mattsson Abstract Alarmingly consistent recent research shows that industrially produced foods such as fast food contain compounds that add to obesity and high cholesterol among young people. Less physical activity and a higher propensity to eat ready-made food (in Sweden and internationally) have aggravated the health situation for the young generation. They also have become ,addicted' to sugar by the consumption of lemonade and other sweet drinks that are often served in conjunction with fast food. Food consumption patterns are highly cultural, and, once formed in early years, they become difficult to change. The findings of this study, which was based on a small sample of written accounts and transcribed interviews, indicate that high-school students in Sweden are well aware of the good and bad attributes of fast food, such as: speed, convenience, fat and sugar. Clear differences in attitude were found between male and female students: female students view fast food in a broad food chain context, whereas male students concentrate on fast eating and satiety. [source] Prolonged vastus lateralis denervation after botulinum toxin type A injectionMOVEMENT DISORDERS, Issue 3 2010FRACP, John W Dunne MBBS (Hons) Abstract Intramuscular injection of botulinum toxin (BoNT) produces reversible blockade of neuromuscular transmission. In animal experimental models, recovery begins within four weeks and is usually complete by twelve weeks. We present evidence of prolonged denervation following BoNT injection of the vastus lateralis (VL) muscle to correct quadriceps muscle imbalance in patients with chronic anterior knee pain. Needle electromyography data were obtained from 10 subjects who had received a single BoNT treatment 5 to 19 months earlier as part of a clinical trial. Insertional and spontaneous activity, recruitment, and motor unit action potentials were examined. Clear differences between the injected and non-injected VL muscles, which correlated with the time since injection, were identified in all subjects. All 10 subjects studied with needle EMG showed evidence of persisting denervation in the BoNT-A injected VL muscle beyond the period of neuromotor recovery expected from animal experimental studies. © 2010 Movement Disorder Society [source] Symptom experience associated with immunosuppressive drugs after liver transplantation in adults: possible relationship with medication non-compliance?CLINICAL TRANSPLANTATION, Issue 6 2008G. Drent Abstract:, Symptom experience (occurrence and perceived distress) associated with side effects of immunosuppressive medications in organ transplant patients may well be associated with poorer quality of life and medication non-compliance. The aims of this study were: first, to assess symptom experience in clinically stable adult patients during long-term follow-up after liver transplantation; and second, to study the relationship between symptom experience and medication non-compliance. This cross-sectional study included 123 liver transplant patients. Symptom experience was assessed using the "Modified Transplant Symptom Occurrence and Symptom Distress Scale" (29-item version) at the annual evaluation. According to the duration of follow-up, patients were divided into a short-term (1,4 yr) and a long-term (5,18 yr) cohort. Medication non-compliance was measured using electronic monitoring. Results showed that increased hair growth was the most frequent symptom in both sexes. Symptom distress was more serious in women than in men. The most distressing symptom in women was excessive and/or painful periods, while in men this was impotence. Clear differences were revealed at item level between symptom occurrence and symptom distress in relationship with the two time cohorts and between sexes. No relationship was found between symptom experience and prednisolone non-compliance. [source] Soil detachment and transport on field- and laboratory-scale interrill areas: erosion processes and the size-selectivity of eroded sedimentEARTH SURFACE PROCESSES AND LANDFORMS, Issue 8 2006O. Malam Issa Abstract Field- and laboratory-scale rainfall simulation experiments were carried out in an investigation of the temporal variability of erosion processes on interrill areas, and the effects of such variation upon sediment size characteristics. Poorly aggregated sandy soils from the semi-arid environment of Senegal, West Africa, were used on both a 40 m2 field plot and a 0·25 m2 laboratory plot; rainfall intensity for all experiments was 70 mm h,1 with a duration of 1 to 2 hours. Time-series measurements were made of the quantity and the size distribution of eroded material: these permitted an estimate of the changing temporal balance between the main erosion processes (splash and wash). Results from both spatial scales showed a similar temporal pattern of runoff generation and sediment concentration. For both spatial scales, the dominant erosional process was detachment by raindrops; this resulted in a dynamic evolution of the soil surface under raindrop impact, with the rapid formation of a sieving crust followed by an erosion crust. However, a clear difference was observed between the two scales regarding the size of particles detached by both splash and wash. While all measured values were lower than the mean weight diameter (MWD) value of the original soil (mean 0·32 mm), demonstrating the size-selective nature of wash and splash processes, the MWD values of washed and splashed particles at the field scale ranged from 0·08 to 0·16 mm and from 0·12 to 0·30 mm respectively, whereas the MWD values of washed and splashed particles at the laboratory scale ranged from 0·13 to 0·29 mm and from 0·21 to 0·32 mm respectively. Thus only at the field scale were the soil particles detached by splash notably coarser than those transported by wash. This suggests a transport-limited erosion process at the field scale. Differences were also observed between the dynamics of the soil loss by wash at the two scales, since results showed wider scatter in the field compared to the laboratory experiments. [source] Effect of environmental variables on eukaryotic microbial community structure of land-fast Arctic sea iceENVIRONMENTAL MICROBIOLOGY, Issue 3 2010Brian Eddie Summary Sea ice microbial community structure affects carbon and nutrient cycling in polar seas, but its susceptibility to changing environmental conditions is not well understood. We studied the eukaryotic microbial community in sea ice cores recovered near Point Barrow, AK in May 2006 by documenting the composition of the community in relation to vertical depth within the cores, as well as light availability (mainly as variable snow cover) and nutrient concentrations. We applied a combination of epifluorescence microscopy, denaturing gradient gel electrophoresis and clone libraries of a section of the 18S rRNA gene in order to compare the community structure of the major eukaryotic microbial phylotypes in the ice. We find that the community composition of the sea ice is more affected by the depth horizon in the ice than by light availability, although there are significant differences in the abundance of some groups between light regimes. Epifluorescence microscopy shows a shift from predominantly heterotrophic life styles in the upper ice to autotrophy prevailing in the bottom ice. This is supported by the statistical analysis of the similarity between the samples based on the denaturing gradient gel electrophoresis banding patterns, which shows a clear difference between upper and lower ice sections with respect to phylotypes and their proportional abundance. Clone libraries constructed using diatom-specific primers confirm the high diversity of diatoms in the sea ice, and support the microscopic counts. Evidence of protistan grazing upon diatoms was also found in lower sections of the core, with implications for carbon and nutrient recycling in the ice. [source] Plasma-soluble Fas (APO-1, CD95) and soluble Fas ligand in immune thrombocytopenic purpuraEUROPEAN JOURNAL OF HAEMATOLOGY, Issue 4 2000Chie Yoshimura Abstract: We investigated the levels of various cytokines and soluble factors in ITP patients, in order to determine the influence of these factors on the pathogenesis of ITP. We found increases in IL-2, IL-6, IFN-,, and M-CSF levels in ITP patients compared with those in healthy individuals. On lymphocyte phenotype analysis, we found no clear difference in total T cell population (CD2+ CD19, cells) or cytotoxic T cell frequency (CD8+ CD11b, cells) between these two groups. The frequency of helper/inducer T cells (CD4+ CD8, cells) was decreased in ITP patients. There was a significant increase in activated T cells (CD3+ HLA-DR+ cells) in ITP patients. Furthermore, frequencies of NK cells of potent activity (CD16+ CD56+ cells) were significantly elevated in ITP patients. Seventeen of the 54 ITP patients (31.5%) had elevated levels of sFas, and 11 of the 54 patients (20.4%) of sFasL. In addition, a significant increase of sFasL was observed in sFas-positive ITP patients, and in these patients the sFasL level was correlated with that of sFas (r=0.687, p<0.01). We found significant increases in IL-2 and sIL-2R levels in sFas-positive ITP patients. For other factors examined, however, there were no differences in level between sFas-positive and-negative ITP patients. Percentages of activated T cells (CD3+ and HLA-DR+ cells) and NK cells (CD16+ and CD56+ cells) were significantly higher in sFas-positive ITP patients than in sFas-negative ITP patients. These findings suggests that the pathogenesis of ITP includes alteration of the Fas/FasL pathway. [source] CLINICAL STUDY: Prolactin response to fenfluramine in abstinent, alcohol-dependent patientsADDICTION BIOLOGY, Issue 3-4 2008Richard J. Porter ABSTRACT It has been suggested that serotonin (5HT) function is abnormal in alcoholics even during abstinence. The prolactin response to fenfluramine (PRF) is generally believed to reflect the activity of the 5HT system and has been previously used to investigate 5HT activity in a variety of conditions, including alcoholism. The origin of the cortisol (CORT) response to fenfluramine is less clear. The objectives of this paper are to examine the prolactin (PRL) and CORT response to dl -fenfluramine in a large cohort of males with alcohol dependence who had been abstinent for 3 weeks, and to compare this with an age-matched control group. Ninety-four subjects with a DSM-III-R diagnosis of moderate to severe alcohol dependence who had been abstinent for 3 weeks, and 23 control subjects underwent neuroendocrine challenge with dl -fenfluramine (10 mg per 10 kg body weight). PRL and CORT responses were measured. No significant difference was found in PRF between abstinent, alcoholic patients and controls (F = 2.7, d.f. = 1.115, P = 0.10). CORT response was significantly lower in abstinent alcoholics than in controls (F = 10.0, d.f. = 1.116, P = 0.002). The results suggest no clear difference in 5HT function between abstinent alcoholics and healthy controls. The reduced CORT response in abstinent alcoholics further supports evidence of hypofunction of the adrenocortical system in this group. [source] RM2 antigen (,1,4-GalNAc-disialyl-Lc4) as a new marker for prostate cancerINTERNATIONAL JOURNAL OF CANCER, Issue 1 2005Seiichi Saito Abstract Although prostate-specific antigen (PSA) has been widely used for early detection of prostate cancer, PSA has problems with specificity and prediction of pathological stage. Therefore, a new marker for prostate cancer is urgently required. We examined expression of a novel carbohydrate antigen, ,1,4-GalNAc-disialyl-Lc4, defined by the monoclonal antibody RM2, in prostate cancer using 75 cases of radical prostatectomy specimens. RM2 immunoreactivity was negative to weak in all benign glands, and weak to moderate in high-grade prostatic intraepithelial neoplasia. In prostatic adenocarcinoma, RM2 immunoreactivity was negative to weak (lower expression) in 20 cases, and moderate to strong (higher expression) in 55 cases. A clear difference of RM2 expression level was observed between Gleason patterns 3 and ,4. Higher expression of RM2 antigen was significantly associated with primary Gleason pattern ,4, high Gleason score (,8), larger tumor volume and advanced tumor stage. Furthermore, 5-year PSA failure-free survival was significantly lower in the higher expression group. However, no significant relationship was observed between RM2 expression level and preoperative serum PSA. Western blot analysis in prostate cancer cell lines PC3 and LNCap revealed that major 49-kDa and minor 39-kDa glycoproteins were common to both cells, but there was an increase of 59- and 125-kDa glycoproteins unique to LNCap and an increase of 88- and 98-kDa glycoproteins unique to PC3. RM2 antigen is a new histological marker for prostate cancer that may reflect the Gleason grading system. Identification of the glycoproteins carrying the RM2 antigen will provide new insights into the properties of prostate cancer. © 2005 Wiley-Liss, Inc. [source] The spatial pattern of summertime subtropical anticyclones over Asia and Africa: A climatological reviewINTERNATIONAL JOURNAL OF CLIMATOLOGY, Issue 2 2010Azar Zarrin Abstract The National Centers for Environmental Prediction-National Center for Atmospheric Research (NCEP-NCAR) monthly mean reanalysis dataset has been used to analyze spatial variations of summertime subtropical anticyclones over the Asia,Africa region. The geopotential height and zonal wind components of 1000, 500, 200, and 100 hPa in a 30-year period (1971,2000) have been used to determine the spatial and temporal variations of the anticyclone centres, their monthly frequency and latitudinal axis variations during April,October. The results revealed that there is a clear difference in the location of the summer anticyclone centres in lower, middle and upper levels of the troposphere. In the lower levels, the Azores subtropical anticyclone is located at the east of North Atlantic. In the middle levels, the frequencies of anticyclone centre are concentrated over the northwest of Africa, Arabian Peninsula and Iranian Plateau. In the upper troposphere, the geographical location of the anticyclone centres and their frequencies in the summer season exhibit a scattered pattern from south of China up to western Iran at 200 hPa, and a bimodal pattern over the Tibetan and the Iranian Plateaus at 100 hPa. In fact, in the entire study domain, the Iranian Plateau is a preferable location of the middle and upper troposphere anticyclones. The highest observed latitude of the subtropical anticyclone at 100, 200 and 500 hPa levels have been seen over north of Tibetan plateau, a large area from east to west of Asia and Iran during August, July,August and July, respectively. The maximum monthly variation in the latitude of the ridgeline is seen at 500, 200, and 100 hPa from June to July which goes even up to 10 degrees at some longitudes. Despite these benefits there is strong evidence that participation in traditional cardiac rehabilitation programs remains low. Various models of cardiac rehabilitation have been implemented including the use of brief structured interventions to enable modification of coronary risk factors. Objectives, The objective of this review was to determine the effect of brief structured interventions on risk factor modification in patients with coronary heart disease. Search strategy, A literature search was performed using the following databases MEDLINE (1966,2006), CINAHL (1982,2006), EMBASE (1980,current) and up to the Cochrane Controlled Trials Register (Issue 2, 2006 of Cochrane Library). In addition, the reference lists of relevant trials and conference proceedings were also scrutinised. Company representatives, experts and investigators were contacted to elicit further information. Selection criteria, All randomised and quasi-randomised controlled trials that compared the effects of brief structured interventions on risk factor modification in patients with coronary heart disease were considered for inclusion in the review. Data collection and analysis, Eligibility of the trials for inclusion in the review, details of eligible trials and the methodological quality of the trials were assessed independently by two reviewers. Relative risks for dichotomous data and a weighted mean difference for continuous data were calculated with 95% confidence intervals. Where synthesis was inappropriate, trials were considered separately. Main results, Seventeen trials involving a total of 4725 participants were included in the final review: three trials compared the effects of brief structured interventions on diet modification; seven on smoking cessation; and seven on multiple risk factors. Two trials involving 76 patients compared brief structured intervention versus usual care for dietary modification. Although there was a tendency for more participants in the intervention arm to lose weight at the 12-week follow up and achieve target cholesterol levels at the 6-month follow up, these results were not statistically significant. Only one small trial involving 36 patients compared brief structured intervention and extensive intervention for dietary modification and demonstrated a significant reduction in the percentage of energy obtained from fat and saturated fat intake among participants receiving extensive intervention. However, no difference in fish, fruit and vegetable intake between the groups was evident. Six trials involving 2020 patients compared brief structured intervention versus usual care for smoking cessation. There was no difference in the smoking cessation rates at the 3- and 6-week follow up, however, there was evidence of a benefit of brief structured interventions for smoking cessation at the 3-, 6- and 12-month follow up. In the only trial that and compared brief structured intervention and extensive intervention for smoking cessation in 254 participants there was no clear difference of a likelihood of smoking cessation between the two groups. In the seven trials that compared brief structured intervention and usual care for multiple risk factor modification there was evidence of a benefit of the intervention on behavioural changes such as fat intake, weight loss and consequently on reduction in the body mass index, smoking cessation and physical activity among the participants. The findings concerning the effect on blood pressure, blood glucose levels and the lipid profile, however, remain inconclusive. Conclusions, There is suggestive but inconclusive evidence from the trials of a benefit in the use of brief interventions for risk factor modification in patients with coronary heart disease. This review, however, supports the concept that brief interventions for patients with coronary heart disease can have beneficial effects on risk factor modification and consequently on progression of coronary heart disease. Further trials using larger sample sizes need to be undertaken to demonstrate the benefits of brief structured intervention targeted at the modification of single or multiple risk factors. [source] Genetic diversity in pollen beetles (Meligethes aeneus) in Sweden: role of spatial, temporal and insecticide resistance factorsAGRICULTURAL AND FOREST ENTOMOLOGY, Issue 4 2007Nadiya Kazachkova Abstract 1,Pollen beetles Meligethes aeneus are pests of oilseed Brassica crops that are subject to intensive chemical control. Resistance to pyrethroids has been reported. Although this insect is of great economic importance, little is known about its genetic properties and population structure. 2,Amplified fragment length polymorphism (AFLP) analysis with the restriction endonuclease combination EcoRI and PstI was performed on 133 samples of groups of three pollen beetles collected during 2001,04 from five different provinces of Sweden. Both susceptible and resistant insects were studied. Using one primer combination, more than 450 polymorphic DNA fragments were obtained and, in total, four primer combinations were used for analysis. A subsample of 59 single beetles was analysed using one primer combination. 3,AFLP profiles were analysed by similarity measures using the Nei and Li coefficient and Neighbour-joining dendrograms were generated. The dendrogram built using 133 samples showed three distinct groups, each containing beetles representing one generation. Statistical analysis using analysis of molecular variance of single beetle samples showed no evidence of significant genetic difference between resistant and susceptible beetles. Instead, a clear difference between samples, depending on time of collection and generation, was observed. 4,The expected regional population structure, although statistically significant, explained little of the variation. The levels of genetic variation within populations were very high. There appears to be a high rate of gene flow between pollen beetle populations. The implications of this in the context of insecticide resistance are discussed. [source] Effects of soil degradation and management practices on the surface water dynamics in the Talgua River Watershed in HondurasLAND DEGRADATION AND DEVELOPMENT, Issue 4 2004D. L. Hanson Abstract When tropical forests are felled, subsequent land uses affect surface runoff, soil erosion, and soil compaction. In some cases, they can markedly change the hydrology of a region with disastrous effects on human life. The objective of this paper is to investigate the effect of rainfall on stream hydrology due to conversion of primary forests to agriculture. Near surface water dynamics were compared for three land uses on the steep hillsides in the Talgua River Watershed in Honduras: degraded grass-covered field; traditional coffee plantation; and primary forest. Infiltration and surface runoff rates were measured using several methods. A clear difference was observed in hydraulic conductivity between the degraded and non-degraded lands. The degraded grass-covered hillslopes developed a surface restrictive layer with a low saturated hydraulic conductivity of 8 to 11,mm/hr, resulting in more frequent overland flow than traditional coffee plantation and primary forest. Soils under the latter two land-use types maintained high infiltration capacities and readily conducted water vertically at rates of 109 and 840,mm/hr, respectively. Dye tests confirmed that the coffee plantation and primary forest both maintained well-connected macropores through which water flowed readily. In contrast, macropores in the degraded soil profile were filled by fine soil particles. Soils in the degraded grass-covered field also showed more compaction than soils in the coffee plantation. [source] Differential expression of toll-like receptor mRNA in treatment non-responders and sustained virologic responders at baseline in patients with chronic hepatitis CLIVER INTERNATIONAL, Issue 9 2006Qi He Abstract: Background/Aims: The contribution of the host immune response to sustained virologic response is not clear in patients with chronic hepatitis C (CHC). The aim of this study was to explore the relationship of the toll-like receptor (TLR) expression with the outcome of antiviral therapy in hepatitis C viral infection. Methods: Peripheral blood mononuclear cells (PBMC) were obtained from 15 CHC patients before a 48-week treatment with pegylated interferon (PEG IFN) ,-2a and ribavirin. A multiplex semi-quantitative reverse-trancriptase polymerase chain reaction (RT-PCR) was used to compare the relative abundance of TLR2,9 transcripts. Results: mRNA levels of TLR2, 3 and 6 were significantly higher in CHC subjects compared with normal controls (n=8). When patients were classified into non-responders (n=8) and sustained virological responders (n=7) according to the virological outcome of the treatment, there was a clear difference in baseline mRNA expression of TLRs and T-helper (Th) 1/2 cytokines. In addition, the mRNA expression of IFN-, and nuclear factor of activated T cells (NFAT), which is exclusively expressed in activated T cells, was inversely correlated with that of TLR4, 6 and 9 in non-responders. Conclusions: TLRs mRNA levels are differentially expressed in baseline PBMC of chronic HCV-infected subjects with or without responsiveness to antiviral therapy. A substantial increase in monomer conversion and molecular weight is observed at room-temperature ionic liquid (RTIL) >60 vol.-%, and the resulting PMMA has a broad multimodal MWD. Renkonen Background:, Previous work in type-I pollen allergies has mainly focused on lymphocytes and immune responses. Here, we begin to analyse with a systems biology view the differences in conjunctival epithelium obtained from healthy and allergic subjects. Methods:, Transcriptomics analysis combined with light and electron microscopic analysis of birch pollen allergen Bet v 1 located within conjunctival epithelial cells and tissues from birch allergic subjects and healthy controls was carried out. Results:, Bet v 1 pollen allergen bound to conjunctival epithelial cells within minutes after the exposure even during the nonsymptomatic winter season only in allergic, but not in healthy individuals. Light- and electron microscopy showed that Bet v 1 was transported through the epithelium within lipid rafts/caveolae and reached mast cells only in allergic patients, but not in healthy individuals. Transcriptomics yielded 22 putative receptors expressed at higher levels in allergic epithelium compared with healthy specimens. A literature search indicated that out of these receptors, eight (i.e. 37%) were associated with lipid rafts/caveolae, which suggested again that Bet v 1 transport is lipid raft/caveola-dependent. Conclusions:, We show a clear difference in the binding and uptake of Bet v 1 allergen by conjunctival epithelial cells in allergic vs healthy subjects and several putative lipid raft/caveolar receptors were identified, which could mediate or be co-transported with this entry. The application of discovery driven methodologies on human conjunctival epithelial cells and tissues can provide new hypotheses worth a further analysis to the molecular mechanisms of a complex multifactorial disease such as type-I birch pollen allergy. Bell Background: There is a need for new clinical models to investigate effectively the development of pain after surgery and the effect, if any, of pre-emptive treatment. Bilateral models are of special interest, since the patient serves as his/her own control. The objective of this preliminary study was to test a clinical model for the study of acute and chronic pain after bilateral reduction mammoplasty. Methods: Eight patients participated in the study where the breasts were randomized to test and control groups. In each patient, one breast was preoperatively infiltrated with lidocaine and adrenaline and the other breast infiltrated with saline and adrenaline. Assessment included visual analogue scale (VAS) pain intensity, thermal thresholds, mapping for punctate hyperalgesia and tactile sensation. Assessments were made preoperatively, postoperatively and at 6 months after surgery. Results: With regard to acute postoperative pain intensity, the model demonstrated a clear difference between lidocaine and placebo treated breasts. There was no difference between lidocaine and placebo treated breasts with regard to chronic pain, but these results are inconclusive due to small number of patients. Conclusion: The model is sensitive and may be useful in studies of mechanisms of development and prevention of chronic pain after surgery. [source] Aberrant expression of pyloric gland-type mucin in mucin-producing bile duct carcinomas: A clear difference between the core peptide and the carbohydrate moietyPATHOLOGY INTERNATIONAL, Issue 8 2005Masamichi Goto The authors have recently defined the clinopathological entity of a mucin-producing bile duct tumor (MPBT), and divided MPBT into two distinct subtypes: ,columnar-type' and ,cuboidal-type' MPBT. Mucin core protein 6 (MUC6), which is present in normal pyloric glands, had higher expression levels in cuboidal-type tumors than in columnar-type tumors. In the pyloric glands, a carbohydrate antigen detected by monoclonal antibody HIK1083 (CA/HIK1083) is also expressed. In order to evaluate the coexpression pattern of MUC6 and CA/HIK1083 in MPBT, expression profiles were evaluated in 38 surgically excised mucin-producing bile duct carcinomas (MPBC; cuboidal-type, n = 15; columnar-type, n = 23), using immunohistochemistry. The staining rate was graded as follows: ,, <5% of neoplastic cells stained; +, 5% to <20%; +,+, 20% to <50%; +,+,+, ,50%. In cuboidal-type MPBC, MUC6 was positive in all cases (+,+,+, 13/15; +,+, 1/15; +, 1/15), whereas CA/HIK1083 was negative in all cases (,, 15/15; P < 0.0001). In columnar-type MPBC, MUC6 was positive in 65% of cases (+,+,+, 6/23; +,+, 8/23; +, 1/23; ,, 8/23), and CA/HIK1083 was positive in 52% (+,+, 3/23; +, 9/23; ,, 11/23; not significant). Our results clearly demonstrate that cuboidal-type MPBC have an aberrant pyloric glandular phenotype, that is, MUC6+/CA/HIK1083,. This unique profile may be related to different outcomes of patients with MPBC. [source] Cyclobilirubin formation by in vitro photoirradiation with neonatal phototherapy lightPEDIATRICS INTERNATIONAL, Issue 3 2001Saneyuki Yasuda Abstract Background: The main mechanism of phototherapy for neonatal hyperbilirubinemia is the production and excretion of (EZ)- and (EE)-cyclobilirubin (4E,15Z- and 4E,15E-cyclobilirubin). Thus, the clinical efficacy of the light source for phototherapy must be evaluated by cyclobilirubin formation from (ZZ)-bilirubin in in vitro photoirradiation. Methods: In the present study, we investigated the in vitro production pattern of bilirubin photoisomers by phototherapy light from the bilirubin,human serum albumin complex. Results: No clear difference was found in the curves relative to (ZZ)-bilirubin and its photoisomers under aerobic and anaerobic conditions. The ratio of (EZ)-cyclobilirubin to (ZZ)-bilirubin increased proportionately to the dose of irradiating light and no photoequilibrium state was observed analogous to that found in configurational photoisomerization. The concentration of (EZ)- and (EE)-cyclobilirubin increased proportionately with the grade of the percentage decrease in A460 nm from 0 to 23%. With a percentage decrease in A460 nm of 23% or more, the cyclobilirubin concentrations reached a steady state. The reason for this appears to be that the concentration of (ZZ)-bilirubin, a substrate for photoisomers, dropped below 1 mg/100 mL. Biliverdin was produced only in trace amounts. However, the absorption at 520,700 nm increased after a percentage decrease in A460 nm of more than 23%. Conclusions: The results of the present study show that little bilirubin photooxidation occurred with in vitro aerobic photoirradiation. Before the concentration of cyclobilirubin reaches a steady state, it is theoretically valid to use the percentage decrease in A460 nm for the evaluation of the clinical efficacy of the light source. A systematic study of the effects of the dipolar interaction on the magnetic configurations is performed in these nanomagnets, and further the coercive field and the remanence as a function of dipolar interaction are analyzed. The results show that the magnetic configuration and thus the magnetization reversal process of nanomagnets are dependent strongly on the strength of dipolar interactions. For the case of small dipolar interaction, the magnetization reversal process is mainly dominated by spin rotation, while the reversal transforms to the vortex nucleation and propagation formations with increasing dipolar interaction. Moreover, if the dipolar interaction is neglected in the calculation of the total energy, no clear difference is found among hysteresis loops of four nanomagnets with same areas, but the inclusion of dipolar interaction can lead to different hysteresis loops for nanomagnets with same areas but different shapes. This indicates that the dipolar interaction is important for accounting for the shape effect of the magnetic properties in nanomagnets. The GC system was analyzed in Curiaú and Pacoval, two remnant Quilombo populations (African-derived populations) from the Brazilian Amazon. There was no significant statistical difference in allelic frequencies between the two populations or between them and three other African-derived Brazilian populations (Mimbó, Sítio Velho, and Gaucinha in Northeastern Brazil). These populations share similarities among themselves and with African populations (high frequencies of GC*1F and lower frequencies of GC*1S), which may reflect the influence of a high level of African contribution to their formation, but there is a clear difference between them and Europeans and South American Indians. It is suggested that the GC system is a useful marker for studying relationships between single populations and major ethnic groups, but does not discriminate between populations which share the same parental stock. The crack propagation along the warp and weft directions, respectively, was considered for the composites cooled at two different rates during laminate molding. The interlaminar fracture toughness was characterized by determining the critical strain energy release rate (GIC) of initiation and propagation measured from the double cantilever beam tests. In the case of a slow cooling rate (1°C/min), most specimens possess pure interlaminar crack propagation and direction-independence characteristics. Nevertheless, the high-cooled (10°C/min) specimens fractured in both directions suffer extensive intraply damage (crack branching, debonding, and bridging of 45°-oriented interfacial yarns) and knit thread breakage, leading to GIC of propagation two times higher than that of the slow-cooled specimens, and the clear difference in the GIC values of initiation between the two directions may be due to the contribution of the knit thread breakage to the fracture energy. We have developed a detergent-based fractionation method which reduces complexity of the protein extracts, is sufficiently fast to allow differential proteomics analysis after treatments of neurons for short time periods, can be applied to small numbers of cells directly in culture plates, and allows differential extraction of proteins in a compartment-specific manner. The sequential use of detergent-containing buffers on neurons in culture plates yields four extracts enriched in cytosolic, membrane-bound or enclosed, nuclear, and cytoskeletal proteins. Fractionation of neurons was validated by comparison of the distribution of known subcellular marker proteins in the four extracts using Western blotting. Comparison of extracts by DIGE showed a clear difference in protein composition demonstrating significant variations with a fold change (FC) of at least 1.20 for 82% of the detected spots. Using proteins identified in these spots that could be assigned a subcellular localization based on descriptions in the Uniprot database, an extraction efficiency of 85% was calculated for cytosolic proteins in extract 1, 90% for membrane-bound and membrane-enclosed proteins in extract 2, 82% for nuclear proteins in extract 3 and 38% for cytoskeletal and RAFT proteins in extract 4. [source] Effects of ruminal dosing of mechanical stimulating brush on digestibility and rumen fermentation status in Holstein steers fed low rice straw at different lengthsANIMAL SCIENCE JOURNAL, Issue 1 2004Ken-ichi HORIGUCHI ABSTRACT The objectives of the present study were to determine the effect of ruminal dosing of mechanical stimulating brush (Rumen faibu; RF) on digestibility and on rumen fermentation status in Holstein steers fed high concentrate and low rice straw. Eight steers (461 kg in average bodyweight) were used. Four steers were orally dosed three RF per head (RF-dosing) and the other four were not dosed (non-RF). All steers were fed enough concentrate and rice straw to gain 1.4 kg/day in bodyweight. Rice straw was cut 2 cm and 30 cm. The organic cell wall content of feeding diets was 17% in dry matter basis. Digestibility and nutritive value were not affected by RF dosing and length of rice straw. Ruminal pH was significantly lower in RF-dosing group than in non-RF group after feeding, and tended to be higher in long rice straw than in short rice straw throughout the day. There were no clear difference on total volatile fatty acid concentration and molar proportion of volatile fatty acid by RF dosing and length of rice straw throughout examined period. These present results suggest that digestibility and rumen fermentation status are not affected by RF dosing when organic cell wall content of feeding diets is approximately 17% in dry matter with rice straw over 2 cm. Smith BSc Huntington's Disease (HD) is an inherited neurological disorder causing movement impairment, personality changes, dementia, and premature death, for which there is currently no effective therapy. The modified tetracycline antibiotic, minocycline, has been reported to ameliorate the disease phenotype in the R6/2 mouse model of HD. Because the tetracyclines have also been reported to inhibit aggregation in other amyloid disorders, we have investigated their ability to inhibit huntingtin aggregation and further explored their efficacy in preclinical mouse trials. We show that tetracyclines are potent inhibitors of huntingtin aggregation in a hippocampal slice culture model of HD at an effective concentration of 30,M. However, despite achieving tissue levels approaching this concentration by oral treatment of R6/2 mice with minocycline, we observed no clear difference in their behavioral abnormalities, or in aggregate load postmortem. In the light of these new data, we would advise that caution be exercised in proceeding into human clinical trials of minocycline. Ginzler Objective To assess the effect of mycophenolate mofetil compared with intravenous pulses of cyclophosphamide on the nonrenal manifestations of lupus nephritis. Methods Patients with active lupus nephritis (renal biopsy class III, IV, or V) were recruited for the study (n = 370) and treated with mycophenolate mofetil (target dosage 3 gm/day) or intravenous cyclophosphamide (0.5,1.0 gm/m2/month), plus tapered prednisone, for 24 weeks. Nonrenal outcomes were determined using measures of whole body disease activity, including the British Isles Lupus Assessment Group (BILAG) disease activity index, the Safety of Estrogens in Lupus Erythematosus: National Assessment (SELENA) version of the Systemic Lupus Erythematosus Disease Activity Index (SLEDAI), and immunologic variables. Results Both treatments were effective on whole body disease activity in the systems examined, as indicated by changes in the classic BILAG index. With either treatment, remission was induced, notably in the mucocutaneous, musculoskeletal, cardiovascular/respiratory, and vasculitis systems, and flares were rare, as measured by the SELENA,SLEDAI. Levels of complement C3, C4, and CH50 and titers of anti,double-stranded DNA antibodies were normalized after treatment with either mycophenolate mofetil or intravenous cyclophosphamide. Conclusion In addition to the efficacy of both treatments on the renal system, this analysis showed that remission could also be induced in other systems. There was no clear difference in efficacy between mycophenolate mofetil and intravenous cyclophosphamide in ameliorating either the renal or nonrenal manifestations. Mycophenolate mofetil is, therefore, a suitable alternative to cyclophosphamide for the treatment of renal and nonrenal disease manifestations in patients with biopsy-proven lupus nephritis. To study cause-effect relationships, olfactory mucosal histopathology, glial fibrillary acidic protein induction and neurobehavioural deficits were re-examined in mice and rats of both sexes given a single intraperitoneal dose of 2,6-diClPh-MeSO2 (16 and 65 mg/kg). There was a clear difference in the character of the olfactory mucosal lesions in the two species. In mice, an extensive metaplasia characterised by severe fibrosis, cartilage and bone formation accompanied with large polyps filling the nasal lumen was confirmed. In rats, a dose-dependent weak metaplasia with patchy loss of olfactory epithelium was observed three weeks after dosing, preferentially at the dorsal meatus, nasal septum, and the tips of the middle ethmoturbinates. Large areas of intact olfactory epithelium remained in all animals, particularly in the low dose rats. In both species, 2,6-diClPh-MeSO2 gave rise to significantly increased motor-activities, impaired performance in the radial arm maze, and glial fibrillary acidic protein-induction. Only rats showed hyperactivity at the low dose. Performance in the Morris water maze was unaffected in rats of both sexes indicating that a general impairment in spatial learning could not be supported. We propose that the observed hyperactivity and radial arm maze acquisition deficits originated from a direct effect of 2,6-diClPh-MeSO2 in the brain rather than being a consequence of the olfactory mucosal lesion. [source] The Significance of Crust Structure and Continental Dynamics Inferred from Receiver Functions in West YunnanACTA GEOLOGICA SINICA (ENGLISH EDITION), Issue 6 2009HE Chuansong Abstract: In our study we collected the teleseismic record of 31 broadband stations and 9 PASSCAL stations in West Yunnan, as well as extracted more than a million receiver functions. Using the waveform model and stacking techniques, we calculated the earth crust thicknesses and Vp/Vs ratios below the stations and obtained 35 valid data points. At the same time, we evenly stacked the receiver functions at the same station and superimposed the two profiles' cross sections of the main tectonic units. The results show a clear difference between the crust thicknesses of different tectonic units. Because of the magma underplatting and delimanition of the lower crust in the role of deep process, the West Yunnan's crust can be divided two kinds,mafic-ultramafic and feldspathic crusts. The research also shows that the mafic-ultramafic crust corresponds to a good background of mineralization. The delamination of the lower crust is one of the leading causes for moderate to strong earthquake prone in central Yunnan. The thinner crust and high velocity ratio as well as the multimodal structure of Ps in the Tengchong volcanic area confirms existence of a deep process of the strong magma underplating. Due to the basic crust structure and nature, it is believed that the Honghe fault is a main suture of the Gondwana and Eurasia continents. Ekholm Abstract Aim:, Fluorescent protein-based indicators have enabled measurement of intracellular signals previously nearly inaccessible for studies. However, indicators showing intracellular translocation upon response suffer from serious limitations, especially the very time-consuming data collection. We therefore set out in this study to evaluate whether fixing and counting cells showing translocation could mend this issue. Methods:, Altogether three different genetically encoded indicators for diacylglycerol and inositol-1,4,5-trisphosphate were transiently expressed in Chinese hamster ovary cells stably expressing human OX1 orexin receptors. Upon stimulation with orexin-A, the cells were fixed with six different protocols. Results:, Different protocols showed clear differences in their ability to preserve the indicator's localization (i.e. translocation after stimulus) and its fluorescence, and the best results for each indicator were obtained with a different protocol. The concentration,response data obtained with cell counting are mostly comparable to the real-time translocation and biochemical data. Conclusion:, The counting method, as used here, works at single time point and looses the single-cell-quantitative aspect. However, it also has some useful properties. First, it easily allows processing of a 100- to 1000-fold higher cell numbers than real-time imaging producing statistically consistent population-quantitative data much faster. Secondly, it does not require expensive real-time imaging equipment. Fluorescence in fixed cells can also be quantitated, though this analysis would be more time-consuming than cell counting. Thirdly, in addition to the quantitative data collection, the method could be applied for identifying responsive cells. This might be very useful in identification of e.g. orexin-responding neurones in a large population of non-responsive cells in primary cultures. [source] |
