Three tetraspanins from Chinese shrimp, Fenneropenaeus chinensis, may play important roles in WSSV infection

Abstract Three members of the tetraspanin/TM4SF superfamily were cloned from Chinese shrimp, Fenneropenaeus chinensis. The deduced amino acid sequences of the three proteins have typical motifs of the tetraspanin/TM4SF superfamily. Phylogenetic analysis of the proteins, together with the known tetraspanins of invertebrates and vertebrates, revealed that they belong to different tetraspanin subfamilies: CD9, CD63 and tetraspanin-3. The three cloned genes of CD9, CD63 and tetraspanin-3 showed apparently different tissue distributions. The CD9 gene (FcCD9) was specifically expressed in the hepatopancreas. While for the CD63 gene (FcCD63), the highest expression was detected in nerves, epidermis and heart, with low expression in haemocytes, ovary, gill, hepatopancreas and stomach and no expression in intestine, muscle and lymphoid organ. Compared with FcCD9 and FcCD63, the tetraspanin-3 gene (FcTetraspanin-3) was more broadly expressed and its highest expression was detected in the intestine. Its expression in nerves was lower than in the intestine, but was higher than in other tissues. Expression in haemocytes, ovary and muscle was much lower than in other tissues. The expression profiles of FcCD9, FcCD63 and FcTetraspanin-3 in different tissues, including haemocytes, lymphoid organ and hepatopancreas, were compared by real-time PCR when shrimp were challenged by live white spot syndrome virus (WSSV) and heat-inactivated WSSV. All three tetraspanins were markedly up-regulated in the live WSSV-challenged shrimp tissues. The data suggested that the three cloned members of TM4SF superfamily in Chinese shrimp may play a key role in the route of WSSV infection. [source]


The effect of vitamin A supplementation in broodstock feed on reproductive performance and larval quality in Penaeus chinensis

Abstract The effect of feeding four semi-purified diets A1, A2, A3, A4, containing different vitamin A acetate levels 0, 20, 40, 60 mg kg,1 diet, respectively, on fecundity, egg hatching rate, larval survival rate and vitamin A content in eggs of Chinese shrimp (Penaeus chinensis) broodstock was compared with a fresh clam diet (control) in a 60-day feeding trial. The broodstock shrimp fed the diet with 60 mg kg,1 vitamin A acetate added exhibited significantly higher fecundity (P < 0.01). Hatching rate was highest with diet A4 (P < 0.05), whereas hatching rates were similar fed diets A1, A2, A3. Increasing levels of vitamin A in broodstock diet resulted in improvement in larval quality. The vitamin A levels in shrimp eggs from broodstock fed with diet A4 were higher compared with those from broodstock fed with diet A1, A2 (P < 0.01). The fecundity and hatching percentages were positively correlated with the vitamin A content in eggs in the present study. The results of this study showed that higher level of vitamin A in broodstock diet may have positive effects on fecundity and larval quality in P. chinensis. [source]


Polymorphic analysis of microsatellite DNA in wild populations of Chinese shrimp (Fenneropenaeus chinensis)

Abstract Primers were designed for eight microsatellite loci from Chinese shrimp Fenneropenaeus chinensis. Microsatellites were used to characterize three wild populations from the China coast of the Yellow and Bohai Seas (HB), and the west coast (KX) and south coast of the Korean Peninsula (KN). A total of sixty-one alleles were obtained, and the average observed heterozygosity ranged from 0.660 to 0.756. Six of the 24 population-locus cases showed a significant departure from the Hardy,Weinberg equilibrium, three of them from population KN, two from KX and one from HB. The Fst values indicated that genetic variation was greater within populations than between populations. Analysis using unweighted pair group method with arithmetic mean showed that the relationship between populations HB and KX was closer than between KN and the other two populations. Polymorphic information contents of the eight microsatellites ranged from 0.598 to 0.918. These results indicated that all eight microsatellite loci would be useful for the analysis of genetic variation in Chinese shrimp (F. chinensis) populations. [source]
