Nutrient versus pollination limitation in Platanthera bifolia and Dactylorhiza incarnata (Orchidaceae)

Availability of resources and pollination services have been demonstrated to have impact on reproductive success in some orchid species, but to our knowledge no studies have examined the effects of nutrient application and pollination limitation in the same experiment. In this study, factors limiting reproductive success were studied with two terrestrial orchid species in Central Finland during 1996 and 1997. In a field experiment using a factorial design, plants of nectar-producing Platanthera bifolia and nectarless Dactylorhiza incarnata were treated with nutrient application and hand-pollination. Inflorescence size was considered as an indicator of attractiveness to pollinators as well as of the general condition of the plant. In 1996, fertilizer treatment increased relative capsule production in P. bifolia plants with small inflorescences, indicating poorer store of resources in the underground corm and dependence on nutrient availability during capsule maturation, which was not found in large plants. Hand-pollination and large inflorescence size in P. bifolia did not affect capsule initiation, but increased the proportion of mature capsules. P. bifolia may thus be regarded as both resource- and pollination-limited within a year. Pollination success was observed to be the only factor limiting reproduction of the nectarless D. incarnata within a year, because hand-pollination increased capsule production, and there were no significant effects of fertilizer treatment or inflorescence size in 1996. Capsule production in 1996 did not affect the probability of P. bifolia flowering in the following year, while high capsule production decreased the subsequent probability of flowering in D. incarnata. Species with different pollination strategies differed in the use of resources. The nectar-producing P. bifolia had a lower fruit/flower ratio than the nectarless D. incarnata, also after hand-pollination. [source]


Multiple virulence factors of Cryptococcus neoformans are dependent on VPH1

Acidification of vesicular compartments plays an important role in a number of cellular transport processes, including protein secretion, metal cofactor insertion, glycosylation and pH stability. In the present study, we identify and characterize a component of the vesicular proton pump, Vph1p, to determine its role in the virulence of the AIDS-related fungal pathogen Cryptococcus neoformans. Insertional mutagenesis and plasmid rescue were used to identify the VPH1 gene by screening for mutants defective in laccase activity. Disruption of VPH1 resulted in defects in three virulence factors (capsule production, laccase and urease expression), as well as a growth defect at 37°C, but only a small growth reduction at 30°C. These effects were duplicated by the vacuolar (H+)-ATPase inhibitor bafilomycin A1. Furthermore, the vph1 insertional mutant was also avirulent in a mouse meningo-encephalitis model. Complementation of the insertional mutant with wild-type VPH1 resulted in a recovery of virulence factor expression, normal growth at 37°C and restoration of full virulence. These studies establish the importance of the VPH1 gene and vesicular acidification in the virulence of C. neoformans. [source]

Rosette Recruitment of a Rare Endemic Forb (Gaura neomexicana Subsp. coloradensis) with Canopy Removal of Associated Species

Abstract Gaura neomexicana subsp. coloradensis Munz (Onagraceae), (Colorado butterfly plant), a short-lived perennial forb, became listed as a threatened species by the U.S. Fish and Wildlife Service in October 2000 because of its small global range and population size. This endemic subspecies consists of only 18 extant populations within southeastern Wyoming, southwestern Nebraska, and northeastern Colorado. Wyoming occurrences are on private land with the exception of three occurrences on F. E. Warren Air Force Base, near Cheyenne. Gaura neomexicana subsp. coloradensis may be displaced by many competitors, including Cirsium arvense (L.) Scop. (Canada thistle), an invasive, noxious weed. In June 1998, three G. neomexicana subsp. coloradensis populations were examined for rosette establishment in the presence of and after removal of associated species. The four study treatments removed: (1) Cirsium arvense, (2) grass and forb canopy and associated litter, (3) grass and forb canopy, litter, and C. arvense, and (4) no removal of associated plant species (control). Mature G. neomexicana subsp. coloradensis plants were evaluated for height, number of leaves, and capsule production. The mature plant characteristics monitored in 1998 were not good indicators of subsequent G. neomexicana subsp. coloradensis rosette densities in the following spring. Recruitment was assessed by counting new rosettes in the fall 1999. Although both G. neomexicana subsp. coloradensis and the exotic C. arvense have comparable habitat, removal of C. arvense did not increase G. neomexicana subsp. coloradensis vegetative growth, seed capsule production, or rosette density. Removal of other forbs, grass, and litter, however, increased G. neomexicana subsp. coloradensis rosette density in the following two years, indicating that canopy removal of associated species can enhance rosette establishment of this rare native. The accumulation of dense vegetative cover and litter associated with the absence of herbivory and fire may contribute to the decline of rare species in rich riparian habitats. Return of herbivory and fire in mesic sites to reduce standing biomass accumulations should be considered in restoring recruitment potential to rare monocarpic species. [source]
