Adams Abstract Aims:, To determine the role of fungal metabolites in the desorption of metals. Methods and Results:, Desorption of Zn from charcoal by three different fungi was compared against metal desorption with reverse osmosis water, a 0·1% Tween 80 solution and a 0·1 mol l,1 CaCl2 solution. All three fungal filtrates desorbed three times more Zn than either 0·1% Tween 80 or 0·1 mol l,1 CaCl2. Metal chelator production in Trichoderma harzianum and Coriolus versicolor was constitutively expressed while chelator production in Trichoderma reesei was induced by Zn. The presence of Zn inhibited the production of metal chelators by C. versicolor. Only C. versicolor was found to produce oxalic acid (a strong metal chelator). All fungi caused a marked decrease in pH, although this was not enough to explain the increased desorption of the metals by the different fungal filtrates. Conclusions:, Metal chelation via organic acids and proteins are the main mechanisms by which the fungal filtrates increase zinc desorption. Significance and Impact of the Study:, The results of this study explain why plants inoculated with T. harzianum T22 take up more metal from soil, than noninoculated plants while metabolites produced by fungi could be used for metal leaching from contaminated soils. [source] Release behavior of freeze-dried alginate beads containing poly(N -isopropylacrylamide) copolymersJOURNAL OF APPLIED POLYMER SCIENCE, Issue 1 2008Jae-Hyung Choi Abstract Beads composed of alginate, poly(N -isopropylacrylamide) (PNIPAM), the copolymers of N -isopropylacrylamide and methacrylic acid (P(NIPAM- co -MAA)), and the copolymers of N -isopropylacrylamide, methacrylic acid, and octadecyl acrylate (P(NIPAM- co -MAA- co -ODA)), were prepared by dropping the polymer solutions into CaCl2 solution. The beads were freeze-dried and the release of blue dextran entrapped in the beads was observed in distilled water with time and pH. The degree of release was in the order of alginate bead < alginate/PNIPAM bead , alginate/P(NIPAM- co -MAA) bead < alginate/P(NIPAM- co -MAA- co -ODA) bead. On the other hand, swelling ratios reached steady state within 20 min, and the values were 200,800 depending on the bead composition. The degree of swelling showed the same order as that of release. Among the beads, only alginate/P(NIPAM- co -MAA- co -ODA) bead exhibited pH-dependent release. At acidic condition, inter- and intraelectrostatic repulsion is weak and P(NIPAM- co -MAA- co -ODA) could readily be assembled into an aggregate due to the prevailing hydrophobic interaction of ODA. J Appl Polym Sci, 2008 [source] The effect of different kinds of electrolyte and non-electrolyte solutions on the survival rate and morphology of zebrafish Danio rerio embryosJOURNAL OF FISH BIOLOGY, Issue 7 2009F. Lahnsteiner The effect of electrolyte and non-electrolyte solutions on the survival and on the morphology of zebrafish Danio rerio embryos was investigated. Embryos in different ontogenetic stages were incubated in electrolyte (NaCl, KCl, MgCl2 and CaCl2) and non-electrolyte solutions [sucrose and polyvinylalcohol (PVA)] of different concentrations for 5 , 15 min. The embryos were hatched to the long-pec stage and the effective concentrations which caused a 50% decrease in embryo development (EC50) were determined. The morphometric changes, which were caused by the test solutions, were measured. Ion channel blockers were used to see if active ion transport played a role for embryo survival. Finally, dechorionated embryos were exposed to the test solutions to get indications about the importance of chorion and perivitelline space. For 12 hours post fertilization (hpf) embryos and a 15 min exposure period, EC50 was highest for MgCl2 (1·60 mol l,1), followed by sucrose (0·73 mol l,1), NaCl (0·49 mol l,1), KCl (0·44 mol l,1), CaCl2 (0·43 mol l,1) and PVA [0·0005 mol l,1 (2·2%)]. EC50 were lower for early embryonic stages than for advanced stages for all solutions with exception of MgCl2 and sucrose. At the EC50, MgCl2 and CaCl2 solutions did not induce morphometric changes. NaCl and sucrose solutions induced reversible morphometric changes, which were compensated within 10 min. Only the EC50 of KCl and PVA solutions induced permanent morphometric changes, which could not be compensated. Incubation of embryos in electrolyte and non-electrolyte solutions together with ouabain (blocker of Na+, K+ ATPase), HgCl3 (dose-dependent inhibition of aquaporine channels), verapamil (inhibition of calcium and magnesium uptake) and amiloride (inhibition of sodium uptake) significantly decreased the per cent of embryos developing to the long-pec stage in comparison to the same solutions without blockers. Ouabain and HgCl3 also induced morphometric changes. For dechorionated embryos the survival rates in water and in the different test solutions were similar to untreated embryos. [source] Chitosan,alginate,CaCl2 system for membrane coat applicationJOURNAL OF PHARMACEUTICAL SCIENCES, Issue 8 2001Lishan Wang Abstract Water-based formulations are preferred for membrane coat application because they do not require the use of noxious solvents. A novel aqueous chitosan,alginate,CaCl2 system was evaluated as a potential formulation to produce water-insoluble membranes of biodegradable polymers. Chitosan,alginate coacervates were prepared by controlled reaction of chitosan (0.25% w/v) and sodium alginate (0.25% w/v) solutions. Coherent membranes were obtained by casting and drying the coacervates suspended in aqueous CaCl2 solutions (0.05,0.07% w/v). Increasing the calcium content did not modify membrane thickness (25,26 ,m), but reduced the water vapor transmission rate from 658 to 566 g/m2/day, and improved the tensile strength of the membranes from 9.33 to 17.13 MPa. Differential scanning calorimetry, Fourier transform infrared spectroscopy, and elemental analyses of the chitosan,alginate coacervates indicated they were stable for up to 4 weeks of storage in distilled water at ambient temperature. Membranes of the stored coacervates required less calcium to attain maximum mechanical strength. They also had higher water vapor transmission rates than corresponding films prepared from fresh coacervates. Manjula Dhevi In this article, nylon 6,6 (NY66) and glass fiber-(30 wt%) reinforced NY66 (GFNY66) specimens were immersed in various aqueous calcium chloride (aq. CaCl2) mixture solutions at different thermal conditions for varying intervals of time, and analyzed using attenuated total reflection-infrared (ATR-IR) spectroscopy, inductively coupled plasma (ICP), energy dispersive X-ray (EDX), gel permeation chromatography (GPC), and mechanical studies. ICP data revealed increasing concentration of absorbed Ca2+ ions with increasing immersion time resulting in disruption of intra- and intermolecular H-bonding as confirmed using ATR-IR results. From EDX data, the ratio of Ca2+ and Cl, ions absorbed by NY66 was calculated and found to follow its stoichiometric equivalence. GPC data exhibited less reduction in Mn and Mw for aq. CaCl2 -treated NY66 specimens suggesting the absence of any significant chemical degradation, but the occurrence of only physical changes involving H-bond breakage and the formation of new CO···Ca2+ dative bond in NY66 matrix. The mechanical properties of GFNY66 samples treated with various types of aq. CaCl2 solutions exhibited pronounced deterioration, possibly due to the interfacial failure between glass fiber and NY66 matrix. The results obtained from this study were quite useful toward understanding the degradation mechanism in NY66 and GFNY66 caused by various aq.
