Highly Enantioselective Biginelli Reaction Promoted by Chiral Bifunctional Primary Amine-Thiourea Catalysts: Asymmetric Synthesis of Dihydropyrimidines

ADVANCED SYNTHESIS & CATALYSIS (PREVIOUSLY: JOURNAL FUER PRAKTISCHE CHEMIE), Issue 18 2009
Yangyun Wang
Abstract The diastereospecific formation of dihydropyrimidines (DHPMs) has been achieved in moderate to high yields with up to 99% ee by a Biginelli reaction. The reaction was performed by using a combined catalyst consisting of a chiral bifunctional primary amine-thiourea 9f and a Brønsted acid with tert -butylammonium trifluoroacetate (t- BuNH2,TFA) as additive in dichloromethane at room temperature. The possible mechanism for the reaction has been proposed to explain the origin of the activation and the asymmetric induction. [source]

Ca(NO3)2·4H2O-catalysed Biginelli Reaction: One-pot Synthesis of 1,2,3,4-Tetrahydropyrimidin-2-ones/pyrimidine-2-thiones under Solvent-free Conditions

CHINESE JOURNAL OF CHEMISTRY, Issue 11 2008
Abdelmadjid DEBACHE
The synthesis of 1,2,3,4-dihydropyrimidinone/thione de- rivatives was achieved in good to excellent yields using calcium(II) nitrate as catalyst to promote the Biginelli three-component conden- sation reaction from a diversity of aromatic aldehydes, , -keto compounds and urea or thiourea. The reaction was carried out under solvent-free conditions. [source]

Biginelli-like three component reaction: Synthesis of some new ethyl 6-ethoxycarbonylmethyl-4-aryl-2-oxo-1,2,3,4-tetrahydropyrimidine-5-carboxylate derivatives

Javad Azizian
In the context of our high-throughput organic synthesis program, we have studied the reactivity of special ,-keto esters toward the Biginelli reaction. We have found that a diethyl-3-oxoglutarate reacts with one molecule of urea and one molecule of aldehyde under solvent-free conditions to give a new family of 3,4-dihydropyrimidin-2(1H)-ones in good yields. A new biginelli reaction procedure using potassium hydrogen sulfate as the promoter for an efficient synthesis of 3,4-dihydropyrimidin-2(1H)-one

JOURNAL OF HETEROCYCLIC CHEMISTRY, Issue 2 2004
Shujiang Tu
Simple and improved conditions have been found to carry out the Biginelli reaction for the synthesis of 3,4-dihydropyrimidin-2(1H)-one derivatives. This synthesis was performed using potassium hydrogen sulfate as the promoter in ethylene glycol solution. Compared with the classical Biginelli reaction conditions, this new method has the advantage of excellent yields (85,95%) and short reaction time (0.5,2 h). [source]

One-pot Synthesis of Bis(dihydropyrimidinone-4-yl)benzene Using Boric Acid as a Catalyst

CHINESE JOURNAL OF CHEMISTRY, Issue 5 2005
Tu Shu-Jiang
Abstract A simple effective synthesis of bis(dihydropyrimidinone-4-yl)benzene derivatives, using boric acid as catalyst, from isophthalic aldehyde or terephthalic aldehyde, 1,3-dicarbonyl compounds and urea or thiourea in glacial acetic acid was described. As the expansion of the classical Biginelli reaction, this method has the advantage of excellent yields 83%,94% and short reaction time 0.5,1.5 h. [source]
